Proposal for Changes of Motivational System in a Selected Company

The bachelor thesis is focused on proposing changes of the concept of the motivation system in selected society. The theoretical part explains some activities related to human resources management as well as concepts related to motivation and stimulation. The analytical part contains a description of the company and analysis of the current state including a questionnaire survey. The last part of the bachelor thesis is focused on suggestions for improving the motivation system.

New amphiphilic ion pairs for the preparation of vesicular systems

The bachelor thesis is focused on the preparation of the new vesicular systems. The aim of this work is to create the most stable system and find out the most optimal working method for its production. The compared parameters were the energy used for dispersion, the molar percentage of cholesterol added, and the type of double-stranded positively charged surfactant. The whole system consisted of negatively charged surfactant sodium dodecyl sulfate and positively charged surfactant septonex. Each system was compared over time to determine stability based on the size of the particle, zeta potential, and turbidity of the solution. By the method of dynamic light scattering, particle size and polydispersity coefficient were measured. On the other hand, the zeta potential was determined by electrophoretic light scattering. The result of the thesis presents the most suitable parameters of ultrasonic dispersion, the molar proportion of cholesterol, and the use of cationic surfactant. Based on these parameters, it is possible to create a physically stable system for a long period of time, thanks to which such a system can be used in the future for other applications.

A Draft of a Corporate Financial Plan

The master's thesis deals about the draft of a corporate financial plan. First part of the thesis focuses on the theoretical basis of financial planning. Second part focuses on introduction of selected company and business analysis at present. After the company's financial plan for the years 2017 - 2020 is proposed in optimistic and pessimistic version. The last part of the thesis is an evaluation of the company's financial plan.

Structural Studies on Extracellular Domains of adhesion G Protein-Coupled Receptor ADGRG4

Bunková komunikácia je schopnosť buniek príjmať, vysielať a spracovávať signály zo svojho okolia. Môžeme teda hovoriť o komunikácii medzi bunkami a orgamizmom. Extracelulárne stimuly spúšťajú signál, ktorý interaguje s receptormi na bunkovom povrchu, čo má za následok aktiváciu intracelulárnych signálnych proteínov. Tieto proteíny sú rozdelené do viacerých rodín podľa ich špecifickej sekvencie, domén ktoré obsahujú, alebo ligandov, s ktorými interagujú. Jednou z týchto rodín je práve receptorová rodina adhezívnych receptorov spojených s G proteínom (aGPCR), ktorá obsahuje 33 členov. Najnovšie štúdie naznačujú, že fungujú z veľkej časti ako mechanoreceptory; avšak, pre niektorých členov boli identifikované aj ligandy pre ich aktiváciu. V porovnaní s inými GPCR sa táto rodina vyznačuje relatívne dlhou extracelulárnou oblasťou (ECR), ktorá obsahuje súbor rozličných štruktúrnych domén a motívov. ADGRG4 (GPR112) predstavuje jeden z najväčších aGPCR s dĺžkou reťazca väčšou ako 3000 aminokyselín (aa). Väčšina jeho reťazca aa sa nachádza v extracelulárnom priestore (viac ako 2500 aa). Je známe, že ADGRG4 je špecificky exprimovaný v enterochromafinných bunkách umiestnených v epiteli ľudského tenkého čreva. Tieto enteroendokrinné bunky sú zodpovedné za produkciu a sekréciu serotonínu a reagujú na chemické a mechanické podnety. K dnešnému dňu však fyziologická funkcia ADGRG4 v týchto bunkách zostáva neznáma. Cieľ tejto práce bol rozdelený na dve časti. V prvej časti bola snaha o poskytnutie štruktúrnych informácií o obrovskej extracelulárnej oblasti ADGRG4. ECR bola rozdelená na viacero častí pre lepšie skúmanie. Následne boli vzorky exprimované v ľudských embryonálnych obličkových bunkách 293 (HEK293S bunky). Výsledné proteínové štruktúry boli purifikované viacerými technikami. Pre zvýšenie možnosti kryštalizácie, bol purifikovaný proteín deglykozylovaný a až potom podrobený na kryštalizačné pokusy. Druhá časť sa zaoberala analýzou NTF-konštruktov, dobre preskúmaných ADGRs. Tieto konštrukty neobsahovali C-terminálny koniec (CTF), iba N-terminálnu časť (NTF). Desať rôznych proteínov bolo úspešne naklonovaných a expresovaných v HEK293T bunkách. Na vyhodnotenie výsledkov oligomerného stavu vzoriek z prvej aj druhej časti boli použité techniky Native PAGE a dynamický rozptyl svetla (DLS). Ako kontrola autoproteolýzy, ktorá je typickým znakom aGPCR, sa využili rozličné techniky Western blotting (WB). Metóda nano-diferenciačná skenovacia fluorescencia (DSF) sa využila na stanovenie pripravených konštruktov a na nájdenie optimálnych podmienok pre roztoky, ktoré sa v práci používali. Výsledky z prvej časti ukazujú za akých podmienok dokážeme nami vytvorený konštruk podrobiť kryštalizácii a následne ďalej pokračovať v jeho výskume. Druhá časť nám ukázala, ktoré nami klonované konštrukty sa dokážu stabilne expresovať v HEK293T bunkách. Na základe získaných parametrov z prvej a druhej časti dokáže výskum v tejto oblasti rozšíriť a do buducnosti získať dôležité informácie ohľadom týchto pozoruhodných receptorov.

Phenomenon of Home

The main theme of this dissertation is the phenomenon of home. The dissertation is intended to be thought-provoking about the issues of home. The aim of this paper is to clarify the phenomenon of home in relevance to present day and to find the answers to home-relevant questions and relating to the housing of man on this Earth. The dissertation itself is arranged into five chapters. The first chapter is dedicated to the ancient Greek roots of the term "home" and briefly describes how the subject is viewed by linguistic scientists and how it is defined by selected social sciences. The second and the third chapters are at the core of this dissertation and describe in detail the phenomenon of home in the conception of two significant phenomenological thinkers, Czech philosopher Jan Patočka and German philosopher Martin Heidegger. The fourth chapter describes the issues of home through a reception of poetry by famous Czech poets. Mečiar's Slovakia: Integration into NATO or the Alliance with Russia?

The primary objective of the Bachelor's thesis: "Mečiar's Slovakia: Integration into NATO or the Alliance with Russia?" is describe Slovak failed integration into North Atlantic Treaty Organization in the context of Slovak-Russian relations. The thesis confronts declared priorities in foreign and security policy with real actions of the government of Prime Minister Vladimír Mečiar. The first part focuses on the Slovak Republic after its formation in 1993, while the Warsaw Pact disintegrated and Slovakia had to seek alternative security. The second part deals with the third Mečiar's government which was critical for the integration failure of Slovakia. The thesis confirmed, that intensive and high-standard Slovak-Russian relations and Vladimír Mečiar had an impact on that.
