The molecular and biochemical basis of primary coenzyme Q10 deficiency

This diploma thesis focus on disorders of coenzyme Q10, which lead to decrease of its concentration in certain patient`s tissues (CoQ10 deficiency). This thesis introduces two molecular-genetics methods for CoQ10 deficiency analysis. The first one is a screening method - High Resolution Melting Analysis, which is a useful tool for screening of a large group of patiens with possible mutations in PDSS1, PDSS2, COQ2, COQ9, CABC1 and APTX genes, which encode some enzymes of CoQ10 biosynthetic pathway. The second one is direct sequencing of coding sequences of these genes including intron sequences neighbouring exons. This work also describes a model of CoQ10 deficiency applied in vitro using cultured human skin fibroblasts and HEK293 cells. In this case CoQ10 deficiency is evoked by CoQ10 biosynthesis inhibitor - 4 aminobenzoic acid (concentration 1 mmol.dm-3) . 4 days treatment by 4 aminobenzoic acid caused decrease in CoQ10 level in fibroblasts and HEK293 cells (60 - 70 % of control, resp. 41 % of control in case of HEK293 cells), although cell viability and morfology of mitochondria remained unchanged. Our results declare that there was slightly increased reactive oxygen species concentration after treatment, especially the amount of superoxide radicals; nevertheless the ultrastructure of mitochondria stayed...

The old people opinions of past in today context

This diploma work enters theoretically and practically on old people opinions of their life. The introductory chapter describes the life cycle, spiritual ageing and individual value system. It is also concerned with the psychotherapeutical method, created by V. E. Frankl, which is called logotherapy and with chances of social - spiritual help in difficult situations. The second chapter is focused on ageing and the old age - on changes, which this process fetch along and on the ways of living in the old age - in the concrete on the old people's life at home or in the institucional care. The closing chapter contains the investigation, which aims to know, how old people perceive their own life in today context - how much they are satisfied and frustrated, how succesful they were with achieving the purpouse which they had or still have got.

Energy balance and its influential factors

The theoretical part of the thesis deals with the issue of energy balance. Principles of its regulation and factors that affect it through energy intake or energy output are described in the thesis. It also describes methods of measuring and calculating energy output for proper adjustment of energy needs and achieving energy balance. In addition, the thesis presents energy needs in in specific life stages and pathological conditions that may occur during prolonged disruption of the energy balance. Both physiological and pharmacological factors which may impact energy metabolism are mentioned. The main objective of the practical part was to analyze freely available articles on the Internet providing different ways of weight reduction. The articles were subjected to a critical assessment from the point of view of a nutritional therapist and compared with expert studies. The secondary objective was to evaluate the questionnaire which also dealt with weight reduction. The individual responses were focused on the respondents' personal experience with weight loss. The results showed that up to 90 % of respondents attempted to reduce weight and that freely available articles, which do not always provide relevant information, were often used for this purpose. Uncontested proceedings in cases involving minors The aim of this thesis is to provide a comprehensive overview of non-contentious proceedings in cases involving minors with a defined focus on the adoption proceedings and court custody of minors. The purpose of my thesis is to analyse the area of international child abduction and the proceedings of that. Chapter One describes the contentious and non- contentious proceedings and distinction between that with an overview of non-contentious proceedings in cases involving minors and their distribution. For the purpose of this text, non- contentious proceedings in cases involving minors are divided into two subdivisions, namely the adoption proceedings and court custody of minors. Chapter Two concentrates on court custody of minors. Chapter Three focuses on the international child abduction, which is selected from court custody of minors. New substantive law was the cause of creation of the new system of procedural law, which shows a certain degree of shortcomings regarding the procedure for the adoption of an full age. Wenceslao Wodiczka and his Works for the Flute

The Bachelor thesis collects the biographical data of Wenceslao Wodiczka, which can be found within the Czech Republic, and builds on the existing research on his place of birth. It also deals with the compositions for flute as comprehensive information about these pieces has not been published in the Czech literature yet. The work includes modern edition, formal-harmonic plan and the interpretation analysis of "Concerto à 5 stromti : Flauto Traversiere, Violino Primo, Violino Secundo, Alto Viola et Basso", and the incipits of sonatas collections - "Huit sonates pour le violon et la basse dont il y en a quatre pour la flute traversiere...oeuvre second a Sei sonate a flauto solo e basso e violoncello...opera terza".

SPE of Organic Sulphates on Ionexchangers

Diploma work deal with possibility of synthesis and determination 6- sulphatoxymelatonin on weak ion exchanger. First part is focused on synthesis, isolation and determination of 6-sulphatoxymelatonin from obtained publications, description SPE techniques and reports brief summary extraction of organic sulphates using SPE from literature. Second part of work presents results of synthesis 6-hydroxymelatonin and 6-sulphatoxymelatonin based on performed experiment.
