This theses is focused on the European Union and EU funds. Description of structural policy and its history, with a focus on structural and cohesion funds. Furthermore, attention is focused on the subsidy received from the ROP southeast. In summary is given a new programming framework 2014-2020. The practical part of the thesis was to describe, analyze and evaluate the project co-financed by EU structural funds. The project was implemented within the ROP SE and tackle integrated transport in the region Breclav.

The practical part is given a particular public contract, sources of financing, billing and various ways to pay operating costs.

Manuel Miró Title of the diploma thesis: Automated monitoring of Metridia luciferase released from genetically modified cell line. The diploma thesis reports automatic real-time monitoring of the release of Metridia luciferase from genetically modified cells. A previously optimized method for monitoring the natural release of luciferase from cells was used during the experiment. At the same time, the toxic effect of bile acids (chenodeoxycholic and deoxycholic acid) on changes in cell membrane permeability was tested. Liver epithelial cell line modified with the Metridia luciferase plasmid were used during the experiment. After this genetic modification, the cells were able to synthesize the active Metridia luciferase enzyme. This enzyme generates luminescence upon reaction with substrate coelenterazine. The emitted light was detected in the flow cell of a spectrofluorometer at a wavelength of 485 nm. The measurement was taken in a sequential injection system connected to a 3D printed module containing an insert with the tested cell monolayer. PharmDr. Hana Sklenářová, Ph.D. Title of the diploma thesis: Automated monitoring of sulisobenzone liberation through model membrane This diploma thesis deals with the monitoring of biologically active substance liberation through a model membrane using sequential injection analysis. A UV - VIS spectrophotometer was used for the detection. The tested biologically active substance was sulisobenzone (one of the UV filters used in sunscreens) and the permeation through the polycarbonate membrane was monitored using a Franz cell. At the beginning of the work, a calibration curve was created in two media - water and phosphate buffer of pH 7.4. According to these curves, the concentrations - which were further used in the liberation tests - were selected. The given concentrations of sulisobenzone were prepared in different solvents, namely in water, in 10% ethanol, in 10% propylene glycol and in 5% propylene glycol. The tested solvents were selected based on the assumption of increasing the permeability of the membrane for the liberation of substances with limited solubility in the liberation medium. PharmDr. Ludmila Matysová, Ph.D. Title of diploma thesis: Development and validation of a UHPLC method for determination of meloxicam in a dissolution medium This diploma thesis covers the development and subsequent validation of a UHPLC method for determination of meloxicam in a dissolution medium. The columm LiChrospher®, 5 µm RP-18 100 Å (125x4mm) was used for method development. As mobile phase were used methanol, isopropyl alcohol and phosphate buffer (50 mmol/l) in a 51:9:40 ratio, while the aqueous phase pH was adjusted to value 6.8. Flow rate was set to 1.0 ml/min, column temperature was 30řC and injection volume was set to 20 µl. Analytes were detected on the wavelength of 363 nm. The results of the partial validation showed that it is necessary to further optimise the method in order to provide correct results even in the dissolution medium after dissolution.

Marcela Hollá Title of the diploma thesis: Development of UHPLC-DAD method for separation of phenolic substances in plant material and extraction optimization This diploma thesis deals with development of UHPLC-DAD method for separation of selected phenolic substances (arbutin, chlorogenic, neochlorogenic, cryptochlorogenic, isochlorogenic A and caffeic acid, cynarin, catechin, epicatechin, rutin, hyperoside, hirsutrin, reynoutrin, guaiaverin, quercitrin and quercetin). UHPLC analysis of these phenolic substances took place on a Luna® Omega Polar C18 100 (150 x 2.1 mm; 1.6 μm) column with fully porous particles. As a part of the optimization method, a column with core-shell particles (Kinetex® Polar C18) and a column with a hybrid stationary phase (YMC-Triart C18 ExRS) were also tested. The sample injection volume was 2 μl, the flow rate has been set to 0.3 ml/min and temperature of the column has been set to 30 řC. As a mobile phase ultrapure water acidified with acetic acid to pH 2.8 in combination with acetonitrile has been used.
