Skalický, Adam ; Skalický, Tomáš (advisor) ; Šimša, Jaromír (referee) ; Klos, Dušan (referee)
Aim of the study: Verification of safety and efficacy of mesenchymal stem cell therapy after extensive liver resection in a pig model. Type of study: Experimental study Name and location of study site: Department of Surgery, Faculty of Medicine, Charles University in Pilsen Set and methodology: A total of 18 experimental animals underwent liver resection in the extent of hemihepatectomy. Mesenchymal stem cells were administered perioperatively in 10 pigs, the remaining 8 pigs were included in the control group. Blood samples were taken at regular intervals for biochemical and immunohistochemical examination. Control sonographic examination was performed at weekly intervals. Experimental animals were sacrificed on the 28th postoperative day and liver tissue samples were subjected to histological examination. Results: There were no statistically significant differences in any of the monitored parameters. Sonographic examinations, however being performed by one experienced radiologist, did not provide valid data from which liver hypertrophy could be assessed. Conclusion: Mesenchymal stem cell therapy appears to be a safe method. However, the results of our study did not show a positive effect of this treatment on liver regeneration after surgical resection. We find sonographic volumetric examination...
Ability of laical public providing first aid in acute condition from point of view of paramedics
SKALICKÝ, Tomáš
This bachelor's thesis examines the issues of providing non-professional first aid in acute condition from paramedics'point of view. The bachelor's thesis is divided into a theoretical part and research part. At first, the theoretical part describes the emergency medical service, its rescue forces and types of ambulance crew in order to explain the connection between the first aid and pre-hospital emergency care. This part analyses the first aid not only from the theoretical and practical point of view, but also from a social psychological one. Then, the theoretical part also deals with anatomy, physiology, pathophysiology and the first aid of six selected acute conditions. The second part is focused on the analysis of data obtained from a research survey. The research survey was conducted by a qualitative survey using as main methods a semi-structured interview and direct observation during university practice. The semi-structured interview includes 33 questions and is answered by eight paramedics that serves in the South Bohemian Region. The aim of this thesis is to find out what the range of capabilities of general public is in providing the non-professional first aid, how effective the general public is in providing the non-professional first aid and whether people are willing to help. The obtained information led to answering all research questions and fulfilling the aim of the bachelor's thesis. The results of the research evaluate non-professional public as willing enough and with sufficient abilities to provide the first aid but on the other hand the effectiveness of performed actions is considerably insufficient. The main contribution of this bachelor's thesis is the rise of awareness about the issues of the first aid provided by laical public, which could contribute to the improvement of the first aid educational method and subsequently to the improvement of providing the non-professional first aid by general public in cases of acute conditions.

Comparison of Methods of Left Appendage Exclusion During Surgical Ablation of Atrial Fibrillation
Kohut, Marián ; Kuntscher, Vilém (advisor) ; Skalický, Tomáš (referee) ; Rohn, Vilém (referee)
Comparison of methods of left atrial appendage exclusion during surgical ablation of atrial fibrilation Abstract Atrial fibrillation is the most common heart arrhytmia affecting a significant part of the population over 60 years of age and it is an eminent source of cardio-embolic events. Leading source of intra-cardial thrombi is the left atrium of the heart, specifically the left atrial appendage. Randomised studies show significant reduction of cerebral ictus by the use of Warfarin in patients with atrial fibrillation, however the use of anticoagulation has its restrictions, side effects and contraindications. There are several surgical techniques of excision or exclusion of the left atrial auricle. This dissertation thesis is focusing on the comparison of the two most common surgical techniques - resection of the left atrial appendage using a stapler and the exclusion of the left auricle using a clip applied from the pericardial side. At the same time, we've set an objective of confirming the electrical insulation of the left atrial appendage as a significant source of fibrillation impulses when using the clip for the left appendage exclusion. In a group of 30 patients with atrial fibrillation and who underwent the surgical atrial fibrillation ablation using a clip or stapler amputation during complex...
Does the assesment of tumor markers in the context of the last negative conclusions still have any impact in ovarian cancer?

Does the assessment of tumor markers in the context of the last negative conclusions still have any impact in ovarian cancer? Svoboda T.: Dept. Of Oncology and Radiotherapy, Univ.Hospital, Plzen, Czech Republic Summary: The incidence of ovarian cancer is very high in Europe and there is still a lot of encertainaity about the best screening method today. The regular use of ultrasound investigation and CA125 testing is not established as a screening, so the greatest number of patients diagnosed with this tumor has a locally advanced disease. Their prognosis is unfavourable even with the treatment by chemotherapy. We describe the role of main factors associated with the primary tumor (staging, grading, histological type) and it's treatment (surgery type and the influence of residual tumor, type and regimen of chemotherapy and inclusion of radiation therapy in the primary treatment) for disease-free and overall survival. We also confirmed that tumor markers (CA125 incl.) used in screening, during treatment measuring response to chemotherapy and for folow-up as an early predictor of disease recurrence have low specificity and sensitivity to be used today. They have role of a prognostic factor - at least some of them - unfortunately with no impact on survival even when palliative chemotherapy can start...
Interactive scheduling and visualisation
Skalický, Tomáš ; Barták, Roman (advisor) ; Surynek, Pavel (referee)
The goal of this thesis was to design and implement a graphical tool for visualization and editing of schedules which would provide a function for automatic repairing of violated constraints in the schedule. The resulting application called a Gantt Viewer is integrated to the FlowOpt project that represents a complex solution for modeling workflows, creation of schedules from them and analysis of these schedules. The application has been developed with the focus on intuitiveness of the user interface and performance during the management of large schedules. It enables the user to visualize extended manufacturing schedules thanks to the cooperation with other modules of the FlowOpt project. Moreover, the Gantt Viewer incorporates a repair tool exploiting a new Repair-DTP algorithm which is both introduced and demonstrated in this work.
Interactive Gantt Charts
Skalický, Tomáš ; Barták, Roman (advisor) ; Vojtáš, Peter (referee)
This work deals with problems of interactive rescheduling in Gantt charts. The goal of the work is to design and implement an algorithm which resolves the inconsistencies in a schedule. First existing approaches are studied. Algorithm for solving our inconsistencies in a schedule is the most important contribution of this work. It is introduced there and there is also it's proof of correctness and finiteness. Demonstration of applicability of the algorithm follows. For this reason, program GanttViewer which is a part of this work is used. Finally, this program is described in detail.
Predictive and prognostic factors of gastric cancer

Predictive and prognostic factors in gastric cancer Šmíd D. Surgical clinic of University Hospital in Pilsen and Faculty of Medicine in Pilsen, Charles University in Prague. Introduction: Gastric cancer is one of malignant diseases which have the worst prognosis. Unfortunately, there are most patients with advanced-stage disease who have to be treated in a palliative way. Patients suffered from the same type of tumor, being at the same stage of disease and treated with the same chemotherapy have various rates of survival, which can be caused by diverse expression of selected genes impacting on the mechanism of cytostatic effects. The determination of these genes or microRNAs which regulate these genes could be used as a predictive factor for prediction of effects of administered chemotherapy. The determination of some microRNAs, or in the combination with suitable plasmatic factors, could be used as a prognostic factor for patients with gastric cancer. It is also possible to use this combination for early diagnosis of cancerogenesis Object: The aim is to verify the possibility to use expression of selected genes and some microRNAs in tumor tissue as a prognostic factor or a predictor for therapeutic effects of chemotherapy in patients with gastric cancer. Methodology: We retrospectively evaluated the group...
