Kristýna Krasulová Progression in the diagnostics of prostate disorders Bachelor thesis Charles University in Prague, Faculty of Pharmacy in Hradec Králové Healthcare bioanalytics - Medical Laboratory Technician Background: The Bachelor thesis deals with the diagnostics of prostate disorders. It is focused on immunohistochemistry of prostatic biopsy especially on the proof of the antidote against high molecular weight keratins (HMW CK). It was been discovering if certain biochemical values of the prostatic specific antigen comport with the Gleason Score, which is determined according to the result of the immunohistochemistry. Methods: The values of PSA were measured in Oblastní nemocnice Náchod a.s. thanks to their gadget Immulite 2000. The analyzer works on the principle of sequential enzymatic chemiluminiscence immunoassay reaction in the solid phase. Preparates for immunohistochemistry were signed by antidotes against HMW CK. The mouse monoclonal antibody anti-high molecular weight cytokeratin (klon 34βE12) was used. The cell nucleuses were stained by haematoxylin Gill III. The Imaging Software NIS-ELEMENTS AR 3.20.00, camera ProgRes CT 3 (JENOPTIK) by NIKON was used to document the preparates. Side effects of antiepileptic drugs (questionnaire study).

1 ABSTRACT Charles University in Prague Fakulty of Pharmacy in Hradec Králové Department of Biological and Medical Sciences Candidate: Markéta Janů Supervisor:Doc.MUDr.Josef Herink, DrSc Title of bachellor's thesis : Side effects of antiepileptic drugs ( questionare study ) The aim of bachellor's thesis was the comparison of occurence of side effects of anticonvulsant drugs by means of retrospective questionnare. A total number of 12 patients with partial or generalized seizures was involved in this study.The newly applied therapy or non-responsiveness to previous treatment for the latest 12 months were incoming kriteria.The evalution or questionnaires revealed that the most frequent side effects were increased iritability, anxiety and headaches. The resultts obtained were compared with publisher data concerning particular antiepileptic drugs.

The introduction of immunohistochemistry in laboratory practice for prostate disease Němečková Lenka ABSTRACT Increased incidence of benign prostatic hyperplasia (BPH) has been associated with age of men. The quality of lives gets worse in men with BPH. Higher numbers of reported malignant tumours of prostate with age of men, too. Adenocarcinoma of prostate (CaP) was the most frequently diagnosed oncological disease in males after other malignant neoplasms of skin. Physical examination and laboratory methods, especially the measurement of prostatic specific antigen (PSA) in serum, are applied in the diagnosis of CaP. The histological examination of tissue sample obtained by needle biopsy of the prostate confirms the diagnosis of CaP. The immunohistochemical examination of histological sections helps to differentiate malignant and benign prostatic glands in tissue of prostate usually using antibody anti - high molecular weight cytokeratins (HMW CK) highlighting basal cell of benign prostatic glands. Basal cell is missing in malignant glands. The Chronic Wound Therapy

The chronic wound is a secondarily healing wound, which didn't healed during 6 - 9 weeks despite adequate healing. Chronic wounds grow up from the acute wound crossing into chronicity as a result of an illness or an infection. The aim is to heal not only the defect but the illness as a complex. The suitable media of wound is necessary for re-issue of healing process. The balance and distribudion of MMPs and TIMPs is breaked in benefit of MMPs in non-healing wounds. In chronic wound liquids there are increased levels of MMP-2 and MMP-9. More increased proteases MMP-1, MMP-2, MMP-8, MMP-9, MMP-13, neutrophil elastese, uPA can be found directly in the chronic wound. That leads into substrates degradation which are necessary for healing (colagen, gelatin et al.). There was found out, that the decreasing of pH media leads to the decreasing of proteasis activity. The optimal pH of the media is 4 - 7. The epidermal growing factor is significantly more degraded and that leads into inhibition of proliferation of epidermal cells The experimental part target for healing by "wet" therapy. By all twelve pacients there were focused cleaning of wound, and regulation of a wound internal balance. The wound reparation was started. Allergy is regarded as one of the major epidemics of 20 century and even nowadays it still does not lose its power. The number of patients who suffer from various types of allergy is still rising, despite the intensive effort of clinical immunology and allergology experts to reduce it. The rising number of patients suffering from allergies is evident in the area of food industry as well. For one thing, we try to solve all the problems that regularly appear. However, we are not able to avoid the spread of reasons leading to allergic illnesses. We usually know such reasons. Unfortunately, we are very inconsequent to banish them fully. This state is caused by modern life style and the life priorities of individuals and society. This thesis aims to introduce the area of uneligible reactions to consumed food- stuffs. I managed to state the basic difference between the authentic food-stuffs allergy and the food-stuffs intollerance. I also aimed to describe the food-stuffs allergens that apper the most often. Such allergens cause wide range of uneligible reactions and they are followed by serious health problems, especially by sensitive individuals. Effects of cholesterol on the dynamic behavior of muskarinic acetylcholine receptor in the cell plasma membran.

- EN The muscarinic acetylcholine M2 receptor that was originally identified as the predominant muscarinic receptor subtype in the heart is also widely distributed in the central nervous system. Its signal transduction is effected by both the βγ dimer of heterotrimeric G-protein that activates potassium or inhibits calcium conductance and the αi subunit that preferentially inhibits cAMP synthesis. However, M2 muscarinic receptors expressed in CHO cells (CHO-M2) directly activate signalling pathways of all three major subclasses of G-proteins, i.e. preferred Gi/o subclass and at concentrations higher than needed for standard inhibition of forskolin-stimulated cAMP synthesis also Gs and Gq/11 subclasses to cause stimulation of cAMP synthesis and accumulation of inositolphosphates (IP), respectively . In the present experiments we investigated influence of membrane cholesterol content on activation of signalling pathways of these three G-protein subclasses in CHO-M2 cells by carbachol, a non-hydrolysable acetylcholine analogue. Treatment of cells with methyl-β-cyclodextrin decreased cell and membrane cholesterol content by 74% and 39%, respectively, and incubation in the presence of cholesterol-saturated methyl-β- cyclodextrin increased cholesterol content by 169% and 137%, respectively . 5 Abstract of diploma thesis Author: Roman Karaščák Title: Dysmetabolic mechanism in the definition of metabolic syndrome Diploma thesis Charles University in Prague, Faculty of Pharmacy in Hradec Kralove Field of Study: Pharmacy The aim of this thesis is to assess the significance of insulin resistance in the pathophysiology of metabolic syndrome by measuring levels of the body fat burning during fasting and during the oral glucose tolerance test (OGTT). Next, we tried to establish a typology of different insulin groups. Under this classification, we expect identification of stage of pathology of the metabolism by insulin response to glucose load and the consequent effect on tissue metabolism. The experiment included 273 subjects who underwent OGTT and evaluation of the current fat burning device DeltatracTM . The withdrawn blood samples were determined in order to provide levels of insulin and c-peptide by methods imunoradiometric determination/analysis (IRMA). The determined values were statistically processed in order to assess particular relationships. Results section describes the obtained relations. Explains the concepts associated with it. Devote with the most frequent disease in renal carbohydrate metabolism and point to the complications of it is the emerging. The experimental section examines glycemic responses within a heterogeneous group of subjects studied at medical centre of Poliklinika in Hradec Králové using a device for continuous blood glucose monitoring. The purpose of this study was to observe the glycemic response to recommended meals and to determine the glycemic variance of different individuals subjects. Results were individual recommendations based on monitoring the patients during this investigation.
