Its advantage is that it enables qualitative and quantitative determination of substances even in a complex biological matrix. Biocompatible iron chelators are used in therapy primarily for the treatment of iron overload. However, due to their antioxidant and antiproliferative effects, they could also find application in the treatment of other diseases (Parkinson's and Alzheimer's disease, cancer). The biocompatible iron chelator salicylaldehyde isonicotinoyl hydrazone (SIH) from the group of aroylhadrazones was very promising, with low toxicity and significant antioxidant and cytoprotective effect. However, its use is complicated by the short half-life in plasma. Therefore, chelators were derived from its structure that are expected to have higher stability. The tested substances were:...
Breaking of Rod-shaped Model Material during Compression.

The breakage of a model anisometric dry granular material caused by uniaxial compression was studied. The bed of uniform rod-like pasta particles (8 mm long, aspect ratio 1:8) was compressed (Gamlen Tablet Press) and their size distribution was measured after each run (Dynamic Image Analysing). The compression dynamics was recorded and the effect of several parameters was tested (rate of compression, volume of granular bed, pressure magnitude and mode of application). Besides the experiments, numerical modelling of the compressed breakable material was performed as well, employing the DEM approach (Discrete Element Method). The comparison between the data and the model looks promising.

Pigments are mostly nanometric powder of anisometric particles providing complex rheological behaviour. The interparticle forces of grains (> 100 μm) are based on direct contact, which is mainly influenced by the particle size and shape. On the other hand powder (< 4 μm) and especialy nanometric powder (< 100 nm) is influenced mainly by electrostatic charges and by moisture (respectively by the liquid and solid bridges forming). However the morphology affects the surface area and thus the distribution of surface charge and\nthe moisture content on/in single particle. Last but not least, it is also good to know the history of the powder, i.e. the effect of aging and the memory of the powder on the strength of the material. The nanometric powder TiO2 was chosen as a demonstration material and measuring device was FT4 powder rheometer.

Analytical evaluation of novel drug candidates from the group of iron chelators using HPLC I.
Schöngut, Michal ; Kovaříková, Petra (advisor) ; Kučera, Radim (referee)
Nowadays, high performance liquid chromatography (HPLC) belongs among the most modern and most frequently used analytical separation techniques. Biocompatible iron chelators are mainly used for the treatment of iron overload. Nevertheless, in the light of new information about physiological and patophysiological role of iron in organism, their potential use was widened into other diseases. Iron chelators were proved to have antioxidative, cardioprotective and cytostatic effects. Salicylaldehyde isonicotinoyl hydrazone is a biocompatible iron chelator, which also shows these activities. However, its short half life in plasma makes its use difficult. Nowadays, novel derivates of SIH were prepared to improve its stability in plasma. 2'-hydroxypropiophenone isonicotinoyl hydrazone (HPP-INH) is one of newly prepared SIH derivates. The aim of this study was to develop optimal chromatographic conditions for HPLC analysis of HPP-INH and to develop sample pretreatment method using precipitation for its analysis in plasma. The analysis was carried out on a Merck 250x4 mm column I.D. LiChrospher® 100, RP-18e (5 μm) protected with Purospher® 100, RP-18e (5 μm) guard column. The following mobile phase gradient condition was developed for HPP-INH purity evaluation: phosphate buffer (0,01 mol/l NaH2PO4 · 2 H20 water...
