Proposal for Improving the Information System of the Temporary Help Agency

This thesis aims to propose an improvement to the information system of JOBINN&HOSTESSINN s.r.o. Agency employment has some specificities that the information system must account for. The first part of this thesis describes JOBINN&HOSTESSINN s.r.o., compares it to the competition, and analyzes the current information system. The second part proposes improvements to remove a fundamental problem and recalculates the return on investment.
Objectified Valuation of the Business Entity

The aim of this work is to determine the value of the selected business entity. Valuation will be served to the company's management for internal purposes and will be conducted to 12/31 2014. The thesis will be divided into three parts. The first part illustrates the theoretical background needed when examining the issue, primarily methodological approaches of valuation. Second, the analytical part, applies the information processed in the previous section directly to the selected company. Based on the obtained information it will be possible to valuate the company.
High-dimensional single cell cytometry approach for immune system analysis

Technological advancement allowed for the advent of single-cell technologies capable of measuring a large number of cellular features simultaneously. These technologies have been subsequently used to shed light on the heterogeneity of cellular systems previously considered homogeneous, identifying the exclusive features of individual cells within cellular niches. Today, single-cell technologies represent an essential tool for studying the underlying immunological mechanisms correlating with disease. In this context, cytometry is one of the diverse high-throughput methods capable of examining more than 50 features per cell. However, utilising cytometry at its full potential requires the development of optimized assays. Additionally, the resulting high-dimensional data represent a challenge for existing computational techniques. This thesis attempts to address these challenges. The first part of the thesis is focused on developing a non-linear embedding algorithm for rapid analysis of cytometry datasets called EmbedSOM. The comparison of EmbedSOM with other state-of-the-art algorithms suggested the superiority of EmbedSOM with faster runtime. Detection and clonogenic assay of cancer stem-like cells using flow cytometry

The Diploma Thesis deals with an implementation of the new method for an assessment of a cloning efficiency of the cancer stem cells separated by a high speed cell sorter. The cell-sowing on the microtitration plates was performed by the flow cytometry method in a combination with the high speed cell sorter. In the first part of the Diploma Thesis the new method was introduced and tested on the selected cell lines. The obtained results were compared with the results of the limiting dilution assay within four cell lines. As for the second part of my Diploma Thesis, the method was practically applied to analysis of the cloning capacity of two subpopulations of cE2 cells based on the expressions of characteristic markers of stem and cancer stem cells - CD44 and CD 133. Based on the findings, the new method can be introduced as an approved proceeding for the cloning capacity assessment of cancer stem cells in other workplaces that possess analogical device equipment.

The introductory part of this work is dedicated to the conversion of fuel energy into mechanical energy by internal combustion engines. Furthermore, an analysis of technical solutions to reduce fuel consumption. The paper addressed the issue of fuel consumption during operation of the combustion engine and its optimization. It is studied and assessed the impact of systems operating on the principle of magnetic resonance in the fuel in internal combustion engine operation.
