Validation of the PCR method for the determination of isotype variants in the ApoE gene

Apolipoprotein E (APOE) was discovered in the early 1970s as a 34 kDa polypeptide of 299 amino acids. It plays an important role in the metabolism of cholesterol and triglycerides. It is an important component of VLDL, HDL and chylomicrons. The APOE gene is located on the long arm of chromosome 19 in region 19q13.32. It consists of three introns and four exons. It occurs in three alleles e2, e3 and e4, which encode three different isoforms. The isoforms differ in two amino acids (arginine and cysteine) at positions 112 and 158. The e3 allele is the most common, the rarer alleles e2 and e4 are associated with various types of diseases, such as cardiovascular disease, Alzheimer's disease or type III hyperlipoproteinaemia. The aim of my bachelor thesis was to write a research on the topic using information obtained from available proffesional publications, to master basic methods of molecular biology (which include DNA isolation, DNA concentration measurement, preparation of PCR reaction and gel electrophoresis) and to perform PCR method validation. The theoretical part contains general information about lipoproteins, associated with them lipoprotein metabolism and apolipoproteins. It also includes knowledge of the APOE gene, its product and the effect of APOE on various diseases. Finally, there is an overview of laboratory methods used for genetic analysis of isotype variants in the APOE gene. The practical part, which I did in the genetic laboratory GENLABS s.r.o in České Budějovice, includes the analysis of the APOE genotype and the validation of the PCR method, which was successful. Therefore, the method can be included in the daily operation of the laboratory.
Changes in selected laboratory parameters in patients with complicated COVID-19

Ilona Fátorová Title of diploma thesis: Changes in selected laboratory parameters in patients with complicated COVID-19 The aim of the diploma thesis is to write a research on the topic using available literary publications. Comparison of changes in selected laboratory markers in COVID-19 disease in hospitalized patients and patients with a mild course of the disease and then statistical evaluation of the obtained data. The theoretical part contains a briefly description of the COVID-19 disease and its originator - SARS-CoV-2 virus. It deals with the description of the disease, transmission, pathogenesis and the course of the disease. Furthermore, risk factors for the serious course of the disease, its complications, clinical and laboratory diagnostics and finally treatment. In the experimental part of the work, a statistical analysis of the measured data is performed. Comparable hematological and biochemical parameters include WBC, lympho #, NLR, Ret-He, ICIS score, sN # (abs), alG # (abs), D-dimers, fibrinogen, CRP, PCT and myoglobin. Regulatory Issues of Organizing Outdoor Activities

Title: Regulatory Issues of Organizing Outdoor Activities Objectives: The objective of this thesis was to analyse legal environment in relation to organizing programs for outdoor sports and outdoor activities, and to analyse terms and conditions for acquiring the necessary qualification. Methods: I performed analysis of the legal environment by applying descriptive analysis, in particular using document analysis as regards their content. I acquired further data based on consultation with experts in the area of individual activities, methodology and legal regulation. I organized and summarized the acquired data. Conclusions: The analysis demonstrated lack of uniformity and visible fragmentation of the regulation, which is missing overall recommendations and rules for outdoor sports and outdoor activities. Part of the rules is also contained in non-binding directives, having no legal force. I also discovered that the system of further education undergoes a transitory period. Implemented changes should lead to better clarity of the legal environment. However, the present development demonstrates that the process will be slow and demanding. Mentoring and child with disability

The subject of this bachelor thesis is the influence of a formal mentoring program on children with disabilities who participate in this mentoring program. The main aim of the thesis is to finding out, how the formal mentoring program affects children with different types of disabilities, namely from the view of parents of the child and of coordinators of formal program of mentoring. The text is divided into two parts, theoretical and practical. The theoretical part of the bachelor thesis deal with concept of mentoring, volunteering and disability. The practical part contains a qualitative research strategy, where the metod of asking questions and the technique of a semi-structured interview was used. The research was conducted with two groups of communication partners, with volunteer coordinators and parents of children with disabilities. The research group consists of 9 comuniaction partners.
