Recombinant expression of transcription factor TEAD1.

TEAD1 is one of the members of the TEAD transcription factor family. This transcription factor is very important, for example for development of organs. The structure of the entire TEAD1 transcription factor is not now yet known. Nevertheless, structure of two important domains are known. The TEA binding domain, which is important for the binding of the transcription factor TEAD1 to DNA, and the transactivation domain, to which coactivators bind. TEAD1 binds to DNA and upon coactivator bind it affects the transcription of various genes. Genes, that are affected by the transcription factor TEAD1 includes genes regulating proliferation, differentiation and apoptosis of cells. TEAD1 is also the target of the Hippo signalling pathway, which is active in adulthood and prevents abnormal growth of organs. Important for the activity of transcriptional factor TEAD1 are post-translation modifications, such as palmitoylation and phosphorylation. To discover the entire structure of the transcriptional factor TEAD1 and the way it interacts with DNA, the transcriptional factor TEAD1 was prepared recombinationally by expression in cells of Escherichia coli bacteria. Suitable conditions for production of the transcriptional factor TEAD1 were found and the cleavage of the histidine tag by thrombin was performed....

Sustainable development of tourism in the protected landscape area Iron Mountains

The thesis deals with the issue of sustainable tourism development in protected landscape area of the Iron Mountains. The main objective is to evaluate the current status and potential of the region on issues of sustainable tourism development, sustainable forms of tourism and evaluate conservation in the PLA Iron Mountain. The theoretical part is devoted to sustainable development, sustainable development of tourism and the area of the Iron Mountains. The practical part is to map the nature and landscape protection in the PLA Iron Mountain with regard to small-scale protected areas and analyze friendly forms of tourism. Followed by evaluation of the awareness of sustainable development of the territory by questionnaire from the perspective of local public administration in tourism, residents PLA and business owners. Last theoretical part is the SWOT analysis and recommendations for the future.
Further Education in Social Services

The bachelor's thesis deals with further education in social services. The theoretical part describes social work, its types, forms and it explains the concept of quality in social services in relation to the standards of social services. It also includes a sub-chapter that deals with the financing of social services. The concept of continuing education in social services is mentioned in the theoretical part as well as a professional development and also there is a sub-chapter that explains the accreditation of educational institutions and accreditation of educational programs. The aim of the study was to determine the most commonly used forms of further education applied in social services as well as the most beneficial forms of continuing education for social workers and social service workers and a dependence of funding of various forms of continuing education by providers of social services. The results, obtained from a mixed research for mapping this issue more broadly, are described in the practical part. At first a qualitative research was realized by using semi-structured interview with senior staff in selected social service facilities. In order to process the issue more broadly a quantitative research survey was carried out and was based on email answers from executives of facilities listed in the registry of social service providers from across the country. The results from the mixed research provided not only answers to the research questions but also contributed to meeting the determined targets. The thesis should serve the increase of the awareness in the field of further education of social workers and workers in social services and also could possibly serve as a base for further research.
Level on public knowledge concerning of helth insurence policy

Public health insurance is an important part of the Czech health system. In the Czech Republic the model of national health insurance is administered using system of several insurance companies. The main target of these companies is to collect insurance rate from the payers and to cover health care provided to the insuree. The aim of this work is to map and assess professional and non - professional public knowledge of the system of the public health insurance. Three goals were set within the work. The first one was to find out the level of the professional public knowledge of the public health insurance. The second one was to find out the level of the non - professional public knowledge of the public health insurance. The third one was to compare both groups knowledge of the public health insurance. To realize the above mentioned aims three hypotheses were set. Hypothesis No.1: Most of the professional public is informed about the valid legal arrangement of the public health insurance. Hypothesis No. 2: Most of the non -professional public is informed about the valid legal arrangement of the public health insurance. Hypothesis No. 3: There is no difference between the professional and non - professional public knowledge of the valid legal arrangement of the public health insurance. To prove the set hypotheses the form of quantitative research was used. The technique of questionnaire was used for data collecting. For practical part the identical questionnaire for both professional and non - professional public was used. The research group of the questionnaire investigation was formed by respondents living and working in České Budějovice. The professional public within the research work was formed by people who work or worked in the health or social sphere. The final research group was formed by 200 respondents of professional and 200 respondents of non - professional public. The target of the thesis and partial aims were accomplished. The first two hypotheses were proved. The third one was not proved by the research work. It has been investigated what areas is professional and non - professional public most or less informed in. The results of the research work may be used for publication in the professional journals.
Possible financial resources of nonprofit organisation ESN

This thesis focuses on possible financial resources of the ESN nonprofit organisation. First chapters define the following terms: nonprofit organization and civic society; afterwards the ESN organization on international, national and local levels is introduced. The second part of the thesis offers a review of the possible resources like the EU program Youth in Action, structural funds, grant from the Czech ministry and Sophia Foundation. In the practical part, the research done in the ESN CR network can be found. The thesis also comprises the recommendations for the ESN CR members.
Vývoj veřejných deficitů a státní dluh v ČR

V první kapitole této práce je vymezena teorie veřejných deficitů a státního dluhu, součástí jsou i subkapitoly o vývoji těchto makroekonomických ukazatelů v České republice. Druhá kapitola se zabývá daňovou problematikou, jsou zde charakterizovány nejdůležitější daně vybírané v České republice. Třetí kapitola je věnována státnímu rozpočtu roku 2009, kde je popisován zhoršující se vývoj veřejných financí na základě působení celosvětové ekonomické krize a dále reakce vlády na tuto negativní situaci. Čtvrtá kapitola je zaměřena na státní rozpočet České republiky v roce 2010 a jeho výdajový rámec, tzv. "Janotův balíček". Závěrečná kapitola se zabývá dvěma vládními dokumenty, Konvergenčním programem a "Exit strategy", ve kterých jsou zachyceny návrhy řešení problému zadluženosti českých veřejných financí v letech 2011 a 2012.
