3 Summary Title: Study of new prognostic markers in patients with chronic lymphocytic leukemia Chronic lymphocytic leukemia (CLL) is the most common leukemic disorder of adults in Western hemisphere. It is characterized by clonal proliferation and accumulation of morphologically mature lymphocytes in bone marrow, peripheral blood and lymphatic tissues. Clinical course of CLL is extremely heterogeneous with some patients living for decades without need of therapy while others succumbing to the disease within several years. Thus, there has been great interest in identifying prognostic markers that could be used to distinguish patients with an aggressive form of CLL, because they might benefit from early intervention. The process of angiogenesis has been shown to be crucial for growth and metastasizing ability of solid tumors. Angiogenesis in "liquid" tumors was supposed to be less important, nevertheless numerous recent studies have shown enhanced angiogenesis in many hematological malignancies including CLL. Regulation of liver regeneration as assessed by means of molecular genetics Liver regeneration is a noteworthy phenomenon of a vigorous tissue recovery following a wide variety of damaging events. Regenerative ability of the liver is unique among the mammalian visceral organs and plays its role in pathophysiology of heterogeneous hepatic diseases. The recovery process occurs in three phases of which the termination remains the least elucidated. Even the questions of regeneration termination timing and of completeness of organ build-up are still controversial despite decades of studies. The aim of the present thesis was to analyse gene expression data pertinent to late phase of liver regeneration.We evaluated the course of spontaneous recovery in rats subjected to 2/3 partial hepatectomy. Because of lack of generally agreed parameter of hepatic regeneration completeness we proposed a proprietary indicator denoted as %LReg, based on organ and whole body weight assessments performed throughout the study. Title: Defensive combination dealing with ball screening on an attacking ball-handler in a training process of the Czech Female Basketball League teams. Results: The main goal of the thesis is to find out how important is the defensive combination which deals with screening on an attacking ball-handler by her teammate who is off-ball and how much time is spent dealing with this combination according to the coaches of the teams of the Czech Female Basketball League. Methods: The data for the thesis were collected using the unstandardized questionnaire. The methods of evaluation and analysis were applied to examine the results of each question in the questionnaire. Results: I found out that the majority of the Czech Female Basketball League teams spend more time practicing the defensive variation of screening that the offensive one. I also found out that the Czech Female Basketball League teams practice all the possible variations of the defensive combination dealing with ball screening on an attacking ball-handler. Molecular monitoring of residual disease in chronic lymphocytic leukemia

The detection of residual tumor cells in patients with chronic lymphocytic leukemia (CLL) was the goal of this study. Since there is no CLL-specific molecular marker, minimal residual disease detection (MRD) is not simple. The outcome of 29 patients treated with fludarabine, cyclophosphamide and rituximab (FCR) was monitored using PCR with "consensus" primers and comparative PCR. 16 patients subsequently underwent autologous stem cell transplantation (ASCT) which did not significantly prolong the length of molecular remission reached by these patients in comparison to patients treated with FCR only. However, it seems that the length of molecular remission of transplanted patients was not affected by eventual contamination of progenitor cells with leukemic cells while it was affected by the presence of detectable CLL cells in their bone marrow at the time of ASCT. Since "consensus" PCR is not a method sufficiently sensitive for MRD detection in CLL the usefulness of detection of DNA methylation was explored for this purpose. Main issue of this thesis deals with economic information systems used for accounting. It is aimed to analyse current economic information system used in AIRCONDITION.CZ, s.r.o. There was used a survey combined with interviews to analyse strengths, weaknesses and demands of accounting information system. There are suggested optimal solutions for current situation based on this analysis.

Master‘s Thesis deals with the problem of entreprise and it‘s success rate. The aim of accent is analysis of current business in joinery Antonin Hála – HAANT. To provide the analysis are used analysis of internal and occupational environment of the company but also the important financial analysis. All of them are provided for the purpose to identify the gaps and the possibilities of extension of current enterprise. Based on acquired results of analysis is proposed the extension of entrepreneurial activities.
The effect of tick´s serpin IRS-2 on dendritic cells activated by TLR4 ligand

IRS-2 is the inhibitor of serine proteases from the Ixodes ricinus tick. My task in this thesis was to find out the effect of the IRS-2 on dendritic cells activated by TLR4 ligand or by Borrelia afzelii. This effect was studied on several levels. I focused on the cytokine production, the expression of costimulatory molecules and cell signaling pathways. The results show that the IRS-2 may inhibit the expression of costimulatory molecules CD-80 a CD-86 on the cell surface, but this finding needs to be confirmed again. The production of cytokines was not affected by the IRS-2. The effect of the IRS-2 on the activity of p38, Erk1/2 nor NF-?B in LPS stimulated cells vas not observed. The fosforylation of STAT 3 in cells activated by the B. afzelii was lowered by the IRS-2.
