Prognostic and predictive biomarkers of glial tumors of the central nervous system in the context of personalized medicine

The glial tumors, so called gliomas, represent the largest group of the primary central nervous system malignancies. Gliomas remain generally an incurable disease progressing from the lower grades of malignancy to the more aggressive tumors in the course of time. This finally leads to the rapid patient's clinical deterioration and eventually the death. Recently there has been a significant expansion of knowledge in the neuro-oncology domain regarding the onset and development of neoplastic disease at the genetic as well as epigenetic level. Novel prognostic and predictive molecular genetic biomarkers are emerging that can be used for more precise diagnosis, for more accurate assessment of a patients' prognosis, or for better selection of therapy and prediction of therapeutic response. The fundamental view of the histological- based classification of central nervous system tumors is gradually changing and the molecular biomarkers are incorporating in addition to histopathology to refine the diagnoses of many tumor entities at the moment. The recent findings from molecular genetics of gliomas together with the results from clinical trials incorporating the various biomarkers are discussed in this thesis. Role of blood biomarkers in spontaneous intracerebral hemorrhage Background: The study of blood biomarkers can offer new possibilities in diagnostics, prognostication, determination of etiology, and management of spontaneous intracerebral hemorrhage. The aim of our study was to assess the relationship between a panel of selected blood biomarkers and clinical and radiodiagnostic parameters in patients with spontaneous intracerebral hemorrhage. Primarily, the aim was to find a prognostic biomarker which could help in deciding on the optimal categorization of treatment. Patients and methods: A total of 70 patients were prospectively included in this study. The following blood biomarkers were determined: glial fibrillary acidic protein, S100B protein, matrix metalloproteinase 9, interleukin 6, interleukin 10, 25-hydroxyvitamin D, 1,25- dihydroxyvitamin D, total cholesterol, leukocyte counts, blood glucose and C-reactive protein. These were then correlated with selected clinical and radiodiagnostic parameters. Results: Relative to hematoma volume a statistically significant positive correlation was found for S100B, interleukin 10, interleukin 6 and blood glucose (S100B: ρ= 0,54, p< 0,001, IL-10: ρ= 0,43, p< 0,001, IL-6: ρ= 0,26, p= 0,027, blood glucose: ρ= 0,24, p= 0,045). Expression and meaning. Towards the problem of language in E. Husserl's philosophy 1900-1914

The present thesis deals with Husserl's philosophy of language in the period of 1900 - 1914. It aims to present a complex interpretation of its main features, transformations and inner tensions. We use the method of immanent interpretation, i.e. we do not put Husserl's thought into a broad historical context, but interpret it exclusively against background of his own philosophy as a whole. The text is divided in two main parts. The first one is devoted to the Logical Investigations (1900-01) and presents its basic analysis of the meaningful use of lingual expressions. In particular, we focus on the concept of lingual expression as a meaningmediated intentional relationship to an object and we explain it against the background of Husserl's project of the phenomenological elucidation of fundamentals of an a priori and objective theory of science, namely the pure logic. The second part deals with lectures and research manuscripts dating from 1901 to 1914. First, we explain the important breakthrough of the noematic concept of meaning in reference to the Lectures on the Doctrine of Meaning 1908 and related research manuscripts. Design of early warning system against lightning floods in mountain conditions

The sophisticated local warning system against flash floods in mountain conditions is described. It is designed using the latest knowledge on the hydrodynamic mechanisms of storm runoff. It was found that extreme discharges are caused by these factors and their combinations: (1) saturation of soil cover with water, (2) soil surface hydrophobia, (3) gravity destabilized flow of water in the soil, (4) extreme rainfall, usually after prolonged dry period. Typical combinations causing extreme runoff, are (1) and (3) - the water saturation of soil cover can cause gravity destabilized flow induced by a small rain, (2) and (4) - long-term drought may cause the hydrophobic surface of the soil and simultaneously trigger extreme rain.
