Development of vaccines against porcine circovirus type 2

Porcine circovirus type 2 is a single stranded DNA virus that belongs to the genus Circovirus in the family Circoviridae. This virus is associated with many kinds of diseases in pigs and causes significant economic losses in swine-breeding. In this study, two approaches of vaccination were tested in order to develop an effective vaccine against PCV2. The first approach was to test DNA vaccines. For this purpose, eukaryotic expression plasmids encoding two form of PCV2 Cap protein were constructed. The expression plasmids encoding murine TNF-α and IFN-α1 were also prepared for co-immunization with antigen encoding plasmid to enhance the immune response. The second approach is based on the previous finding that chimeric pentamers of VP1 mouse polyomavirus capsid protein fused with PCV2 can induce protective immunity against PCV2. These chimeric pentamers were further modified by AA substitutions in PCV2 Cap immunodominant epitope in order to enhance protective antibody response directed against PCV2. The chimeric pentamers and DNA vaccines were tested for ability to induce antibody immune response against PCV2 in mice. Silent witnesses - impact of witnessing domestic violence in childhood on future partner relationships

This rigorous work was formed as an expansion of diploma work named Silent witnesses - domestic violence viewed through the eyes of children. The main aim of this work is to highlight the situation of children witnessing violence between their parents and the effects of this negative phenomenon on their development. Within this issue the work is focused on different forms of domestic violence, furthermore it points on the legal questions of this problems and it also outlines the crucial aspects of human development. Devastating impact of witnessing domestic violence is highlighted in the practical part of this work which is focused on group work with children. Particular group sessions and direct quotation of individual children, witnesses of domestic violence, give an evidence of the fact how much the children are endangered by this dismal situation in their family. Negative effects of witnessing domestic violence on human development can be seen in the results of the study which was accomplished among 100 women - victims of domestic violence of which 50% were witnesses of violence between their parents in childhood. Intenzifikace produkce bioplynu

Projekt si klade za cíl dosažení zvýšení produkce bioplynu a lepší využití organických látek ze zpracovávaných substrátů na již existujících i nově připravovaných bioplynových stanicích pomocí intenzifikace procesu biometanizace využitím různých metod předúpravy a kofermentace zpracovávaných materiálů, jakož i optimalizací řízení procesu. Hlavním prostředkem k dosažení cíle je podrobné studium vlastního procesu biometanizace pomocí laboratorních testů a poloprovozních pokusů a jejich podrobným analytickým a mikrobiologickým sledováním. Tyto testy budou prováděny převážně na reálných vzorcích fytomasy v různém vegetačním stadiu. Budou navrženy obecně platné metody intenzifikace biometanizačních procesů vedoucích k prohloubení biologické rozložitelnosti zpracovávaných materiálů a k zvýšení produkce bioplynu v anaerobních reaktorech. Metody budou ověřeny v poloprovozním a provozním měřítku včetně analýzy jednotlivých procesů (produkce fytomasy, sběr, doprava, uskladnění, výroba a energetické využití bioplynu, uskladnění a aplikaci fermentačního zbytku) na bioplynových reaktorech zpracovávajících fytomasu a budou navrženy a ověřeny technologické postupy předúpravy a společné fermentace (kofermentace) fytomasy v anaerobních reaktorech zpracovávajících kejdu hospodářských zvířat s cílem intenzifikace produkce bioplynu a zlepšení energetické bilance procesu.
