Nanofibrous Membranes as Drug Delivery Systems

Title of Doctoral Thesis: Nanofibrous membranes as drug delivery systems Nowadays, nanofibrous membranes are very quickly developing systems with proportions in nanometre scale. They are intensively studied and used in many technical disciplines, mostly thanks to their unique properties, among them great surface area to volume as a highlight. Their use in the field of biomedicine and pharmacy as drug delivery carriers, wound dressings and tissue engineering scaffolds is currently a topic of great interest. The doctoral thesis deals with newly formulated drug loaded nanofibrous membranes made of several selected polymers with diverse properties produced by electrospinning technology for large-scale production. The purpose was an experimental investigation and demonstration of potential benefits from application of nanofibrous membranes to selected application sites and dosage forms. The prepared membranes were evaluated by scanning electron microscopy and differential scanning calorimetry that proved successful incorporation of the drugs into the structure of the polymeric nanofibers. Human nutrition, Bacteroides fragilis and the immune system

The presence of bacteria in the human body is a crucial factor for immune system development and its appropriate functioning. This bachelor thesis focuses on the beneficial effects of Bacteroides fragilis and explores the possibility of altering its abundance in the colon by dietary intervention. Both Czech and foreign literature was reviewed and the practical part was carried out using a questionnaire survey. The research indicates that it is possible to affect the proportional representation of Bacteroides fragilis in the colon with specific nutrients and dietary habits. Additionally, the theoretical part describes the benefits of this bacterial species on the human immune system. Bacteroides fragilis produces short-chain fatty acids, antimicrobial substances and polysaccharide A, which are the principal components that influence the host. The practical part builds upon the findings of the theoretical part. It investigates the knowledge of a random sample of Czech citizens about the effect of diet on microbiota composition. It also explores people's view on probiotics and their acquaintance with food composition. The conclusion of the thesis is a correlation between the theoretical and practical part. Formulation of Nanoparticle Systems with Antimicrobial Agents

Drug delivery as modern way of therapeutic application is important goal of contemporary pharmaceutical research. The very specific tailor-made drug carriers and special working methods are used. Parametrs which potentially influence nanoparticle size distribution and their zeta potential were studied. Nanoparticles were prepared by emulsion solvent distribution-evaporation method from commercially available, biocompatible and biodegradable polymer PDLLA (poly (DL-lactic acid)) and other polyesters branched on mannitol or pentaerythritol and polyurethanes synthesized at workplace. Polymers were dissolved in different organic solvents (dichlormethane, chloroform or mixture of both in mass ratio 1:1). Poloxamers and polyvinylalcohol with different molecular weight and different degree of hydrolysis, polysorbate 20, and lecithin from soya bean or from eggs as emulsifying agent were used. Nanoemulsion was prepared by homogenizer with stator and rotor. Particle size and zeta potential was measured by Zetasizer. This study demonstrates that monodisperse nanoparticles can be prepared by emulsion solvent distribution-evaporation method in various composition of organic and also aqueous phase.
