Hereby presented diploma paper is concentrating primarily on the Book Of Henryków (BOH), the outcome of Cistercian literature as produced in Silesia at the break of 13th and 14th century. I worked with Roman Grodecki edition, published in 1991. At the beginning I resumed published editions and source related literature. W orks containing some notions on BOH but not dealing with it further were left behind. Here I also made few remarks on topics I did not work with later on, as clashes between Polish, German and Roman/German canon law and the case of the oldest Polish-written sentence. I did not comment largely literature exploited for the purpose of this paper. In first part I reviewed general political and cultural situation in the 12th, 13th and the beginning of 14th century Silesia, and introduced the moment when Grey Monks entered this area. Then I summarized contemporary Cistercian writing, where those produced in communities of Lubi'l_z, Trzebnica, Henryków and Kamienec are the most significant. These were mainly obituaries, annals, chronicles, Bishop' s lists, legends and Boo k Of Henrykow itself. With the help of previous researches and manuscript fascimile, I analyzed handwriting, authors, organization of BOH manuscript and reasons of manuscripťs creation. The Culture Collection of Actinomycetes in České Budějovice (CCACB) was established in 2006 that serves as a depository for cultures of soil actinomycetes. The cultures can be used for research, industrial applications, education and general scientific interest. Cultures are preserved mainly in glycerol or freeze-dried conserves. CCACB will offer strains of actinomycetes, namely streptomycetes (more than 900 cultures at the present moment), in catalogue of cultures www.upb.cas.cz, www.actinomycetes.cz.
The cultivation of bacteria originated in bat guano

The bat guano represents an important source of organic matter in the oligotrophic caves. The high content of chitin from the remains of digested insect in bat diet causes the slow decomposition of the guano. The bat guano deposits can be auccumulated near the bat colonies during hundreds of years. The bat guano represents unique environment with specific conditions and appropriate composition of bacterial community. In this study, the bacterial communities of the guano from Domica and Ardovská Caves (NP Slovak Karst) were investigated using plate method with six different growing media. Total numbers of cultivable aerobic bacteria in guano were determined on the solid complex medium TSBA (Becton & Dickinson]. Total numbers of cultivable anaerobic bacteria were determined on the DifcoTM Anaerobic agar and blood agar (Schaedler agar with 5% sheep blood).
Microarthropods and microbial participation in oak and beech litter decomposition

The aim of our work was to describe the impact of a simulated vegetation zone shift to higher altitudes (i) on the quantity and quality of microarthropods and microbial communities, and (ii) on the decomposition rate of allochthonous leaf litter. In November 2002, 480 litterbags filled with oak or beech leaf litter were placed into spruce and beech forests (950 m a. s. l.) on Kleť Mt. (1083 m; Blanský les Protected Landscape Area, South Bohemia). Three different mesh sizes were used for certain soil biota exclusion: > 2 mm, 0.5 mm and 42 μm. Litterbags were sampled after 2, 4, 6, 12, 24 and 36 months of exposure in the field. Litter pH and moisture were determined in the laboratory immediately after retrieval. Soil microarthropods (Oribatida, Gamasida and Collembola) were extracted using Tullgren funnels. Soil bacteria and micromycetes were isolated using the dilution plate method and specific growth media.
Změny ve společenstvech půdní mikroflóry a mesofauny během rozkladu listového opadu ve dvou vegetačních zónách - litter bag experiment

The hypothesis, whether autochthonous species of soil biota are able to decompose litter from lower vegetation zones in higher zones, was tested by field trial. Litterbags with litter of beech (.i.Fagus sylvatica./i. L.) and/or oak (.i.Quercus robur./i. L.) were exposed in beech and spruce forest on the mountain Kleť (Protected Landscape Area Blanský les). Litterbags layout simulated vegetation zones shift to the higher altitude induced by global warming. There were used litterbags with three different mesh size (42 µm, 0,5 mm, 2 mm) for exclusion of certain parts of edaphon. Several changes of soil bacteria and micromycetes, oribatid mites, springtails and gamasid mites were studied during two years of exposition. There were exposed 480 litterbags at the start of the experiment (November 2002). After 2, 4, 6, 12, 24 months of exposition parts of all litterbags were recovered. Immediately after the sampling litter pH(KCl), moisture and mass loss were measured.
Použití metody Sherlock MIS System pro identifikaci bakteriálních izolátů z lesního opadu – předběžné výsledky

This work has been related to the effect of climatic changes (especially warming) on communities of forest litter bacteria. We simulated this effect with use of litter bags with beech and/or oak litter transported to beech and spruce forest. After 2, 6 and 12 months of exposition we collected and analysed the samples. We measured quantitative and qualitative characteristics of bacteria and micromycetes community structure in all variants of forest/litter combinations (number of CFU, description of bacterial morphotypes, identification of dominant bacterial morphotypes with MIDI Sherlock Microbial Identification System). We found out that there were differencies in diversity in all variants in exposition time. From the first to the third sampling the CFU bacterial number has decreased whereas CFU micromycetes number has increased. Multivariate comparison (PCA based on relative distribution of bacterial morphotypes in samples) separated the oak litter bacterial community in spruce forest soil as the most different variant among the others.

Cílem práce je analyzovat společnost T-Mobile Czech Republic, a. s. v podmínkách současného trhu, zhodnotit dosavadní vývoj podniku, provést srovnání s konkurencí v oboru a v závěru podnik ocenit z pohledu neznámého investora - minoritního akcionáře na základě veřejně dosupných zdrojů k 31. 12. 2005. Pro ocenění jsou zvoleny metody výnosové, protože T-Mobile je dynamicky rostoucí společností, která dlouhodobě vykazuje vysoký zisk. V rámci skupiny výnosových modelů je pro ocenění zvolena metoda EVA a komparace jejího postupu s metodou FCFF. Jako metoda podpůrná je zvolena metoda účetní hodnoty.
