Effect of chromatin on the repair of double-strand DNA breaks after cleavage by CRISPR/Cas and other programmable nucleases in plants

Plants are highly resistant to ionizing radiation, also thanks to a high-quality repair system for repairing double-stranded breaks. Double-strand breaks in plants are repaired by four repair pathways. Most often, double-strand breaks are repaired by non-homologous end joining (NHEJ), which joins the broken ends without further processing. More accurate but slower and more complex is repair through homologous recombination (HR), which repairs the break using a homologous sequence. HR repair takes place preferentially in a region with active transcription and during the S and G2 phases of the cell cycle. Alternatively, repair further proceeds through single-strand annealing (SSA) or Theta mediated end joining (TMEJ). Both pathways are based on short homology between the overlapping ends of the double-strand break. An often neglected part of repairs is the overcoming of repressive chromatin, which protects the genome from DNA damage and prevents access of nucleases but also acts as a barrier for repair proteins. This bachelor's thesis deals with the issue of employment relations in terms of optimizing wage costs. The main goal is to present propositions in order to optimize the wage and personnel costs of the selected employer. Based on the knowledge gained in the theoretical and analytical part, propositions are then submitted to solve these problems.

The dissertation solves the thickness of built-up areas. It is possible to thicken by housebuilding not only a town but also a village or countryside. This has become the starting point for me. I have chosen for my work the municipality Skrbeň and its cadastral territory. It is situated in the north of Olomouc. It is unique not only because of its location in the fertile Haná but also for its preserved agricultural farmyard which has been creating the centre of the village since the 16th century. The whole agricultural premises was completed in the 1960s by the second part of the cooperative in the north of Skrbeň. The large agricultural complex does not fulfill its function nowadays similarly as many other villages. The aim of my work is to study what has happened with agriculture in our country and why it is no longer able to work effectively. According to the research I want to propose a new recovery system of agricultural countryside and a new agricultural typologie that could bring back life into the countryside and many villages.
Garden city

Housing cannot be simplified to its sole function of living. It must be extended to the relationship to its place and environment. What is the relationship of a person (inhabitant) to the space of his living? How to live? How to live differently? What is housing today and what can be predicted of its development? How can we, the architects, seek a solution to this seemingly banal housing problem in the current context? In the new territory of the South Center of Brno, in connection with the new railway station, large areas have been freed up, which make it possible to organize the urban structure of the new stop. The garden city is one of its designed parts. The new housing environment is offered in the expansion of the city center in the scale of the city district. Living in a garden city is not only a link to natural elements but also an opportunity to restore direct relations to the place of work and to the place of living and other forms of urban living. The work is based on an urban study prepared by the association Ivan Koleček - Pavel Jura architects Lausanne - Brno.
Theories of Czech Syntax in Textbooks for Czech Courses in the Second and Third Grades

Ve své práci jsem se zabývala pozorováním a porovnáváním zpracování učebnic českého jazyka určené druhým a třetím ročníkům ZŠ. Zaměřila jsem se na učivo o skladbě v souvislosti s obsahem osnov uvedených ročníků ZŠ. Zajímaly mě otázky, jak je učivo skladby v jednotlivých učebnicích pojato a následovně seřazeno. Také jsem sledovala, zda se v učebnicích látka o syntaxi vyskytuje kompletně a uceleně bez absence některých částí, které jsou součástí požadavků osnov ZŠ. Nedílnou součástí mého pozorování bylo také sledování estetické úpravy učebnic, která má neodmyslitelný vliv na kvalitu učebnice v oblasti přehlednosti a vlastně její celkovou funkčnost. Dále jsem se zaměřila na způsoby a metody seznamování s učivem a procvičování získaných informací o syntaxi prostřednictvím uvedených cvičení. Následovně jsem mé pozorování ucelila metodickými poznámkami k uvedeným cvičením, popřípadě jsem se snažila cvičení obohatit vlastními metodickými návrhy a zpracováním. Na úplný závěr jsem přiložila několik jazykových her, které mohou sloužit ke zpestření vyučovacích jednotek. Powered by TCPDF (www.tcpdf.org)
"The importance of screening examination methods in prenatal diagnosis from a midwife{\crq}s perspective"

The subject of this bachelor thesis is to create a well-arranged and comprehensible review of the current status and possibilities of the prenatal diagnosis, as it nowadays represents an integral part of almost all pregnancies and many pregnant women decide whether to undergo a prenatal diagnosis and which method of the prenatal diagnosis should be employed in order to achieve a maximum guarantee that they will not give birth to a child with a defect. This thesis has a total of six aims. To find out if the midwives are informed about possibilities of screening examinations in the pregnancy, whether they proceed in compliance with the recommendations of the Society for Gynaecology and Obstetrics during these examinations, whether the pregnant women according to the midwives understand the meaning of the screening examinations, whether the pregnant women according to the midwives are interested in performance of screening examinations in pregnancy and whether the pregnant women are interested in optional, above standard examinations that are not fully covered from the health insurance. In order to ascertain these aims, a total of six hypotheses were postulated on the grounds of which an anonymous questionnaire was created that was focused on evaluation of the theoretical knowledge of the midwives about possibilities of screening examinations in pregnancy and on evaluations of pregnant women{\crq}s interest in these examinations based on the opinions of the midwives. The questionnaire contained a total of 26 questions. The cohort of respondents consisted of the midwives working with pregnant women in 79 selected gynaecological-obstetric outpatient departments in Prague where the research was carried out. A total of 146 questionnaires were distributed, the rate of return of which was 66%. The interpretation of the results is based on the total of 94 respondents. The first hypothesis was based on a presumption that the midwives are informed about possibilities of the screening examinations in pregnancy. This presumption was confirmed as the opinions of the respondents were in 66% in accordance with this hypothesis. In the hypothesis 2, there was a presumption that in the field of the screening examinations, the midwives do not proceed according to the recommendations of the Society for Gynaecology and Obstetrics. This hypothesis was not confirmed as nearly all midwives claimed to proceed in compliance with these recommendations. In the third hypothesis, there was a presumption that according to the midwives, the pregnant women do not know the meaning of the screening examinations in pregnancy. This hypothesis was not confirmed as the average of 86% midwives gave a positive answer at this point. The fourth hypothesis was based on a presumption that midwives positively evaluate the importance of the screening examinations in pregnancy. This hypothesis was confirmed as the average of 92% of responders evaluated the importance of these examinations positively. In the fifth hypothesis, there was a presumption that the pregnant women are according to the midwives interested in performance of screening examination in pregnancy. This hypothesis was confirmed as the average of 95% of respondents gave opinions that were in accordance with this hypothesis. The last hypothesis was based on a presumption that the pregnant women are not interested in the optional, above standard examinations that are not fully covered from the health insurance. This hypothesis was not confirmed as the average of 51% of respondents reported the pregnant women to be interested in these examinations. This thesis and the results of the research part should help the midwives and other medical staff to increase the awareness on screening examination methods of the prenatal diagnosis.
Publi attitudes to NATO

In January survey CVVM SOÚ AV ČR investigated public attitude to membership of the Czech Republic in NATO and to some aspects concerning this membership. The question about satisfaction with membership in NATO was positively answered by more than half of respondents (55, 3%), on contrary 29, 2 % of addressed citizens shared opposite opinion . In comparison to year 2002 the same question was positively answered by 61 % and negatively by 23 % of respondents.More than 15 % of people answered “I do not know“, which is almost the same number as in year 2002.

December CVVM survey was also focused on relationship of Czech citizens to other nations and nationalities. They had to choose from 24 nationalities and evaluate their attitude to these nationalities by marks (1-7) according to their attitude to them.
