The diploma works demonstrates the thermodynamic calculation and design of the geometry flow profile of the back-pressure steam turbine for specified inlet and outlet conditions of steam. Next target of the work is to design the drawing of the turbine axial cross section. Structuring of this diploma work calculation is following: - regulating stage thermodynamic calculation - turbine stages calculation - equalizing-balancing piston calculation - entire turbine intrinsic efficiency and power rating determination - basic calculation of gearbox parameters and RENK-type gearbox selection for turbine and generator arrangement
Repair of the turbine housing

This diploma thesis deals with the repair of a steam backpressure single-stage turbine. This thesis is assigned to a specific Spilling turbine case. A revisional report of this turbine with repair or exchange suggestions of demaged turbine parts is processed. In addition, a simulation of the contact pressure is carried out on the split plane of the housing. Structural modifications are designed and simulated to increase the parting plane's tightness. Further, the calculation of the tightening torque of the split plane is performed and a control thermodynamic and strength calculation of the labyrinth seals is performed.
The significance of ovarian cancer cell chemosensitivity testing.

Ovarian cancer is a leading cause of death from gynecologic malignancies. Most ovarian carcinomas are diagnosed in an advanced stage. Surgical treatment promotes a high response rate combined with chemotherapy. However, two-thirds of patients experience a relapse, followed by a development of chemoresistance, and, subsequently, death. The biological diversity of individual types of ovarian carcinoma is so varied, that a single treatment regimen can not be considered as a fitting treatment for all patients. Various studies focus on the individualisation of cancer treatment. This study targets the in vitro sensitivity testing in ovarian carcinoma cells on cytostatics using the MTT test. We presume that this method could be utilised in clinical decision-making process. Between the years 2006 and 2010, we have acquired tumor tissue and malignant ascites from a total of 55 patients enrolled in our study and analyzed those samples in vitro for chemosensitivity / chemoresistance using the MTT test. The aim of this research work was to asses chemosensitivity of ovarian carcinoma cells and compare these results with clinical parameters. Carcinoma cells displayed in vitro chemosensitivity to cisplatin in 67% of patients. Tumor cells of 22% patients proved to be sensitive to carboplatin and 16% to paclitaxel....

Diagnosis and therapy of intrauterine pathologies associated with pregnancy.

Introduction: Actually 0.7-1 percent of all deliveries can be followed by secondary uterine bleeding. There is a residual trophoblastic tissue diagnosed in most of these cases and it is often managed by repeat intrauterine interventions. These operations are connected with high risk of formation of intrauterine adhesions and their early diagnosis and management can be important for next fertility. Material and methods: There were generally 188 patients included into the study. All patients underwent ultrasound examination in 6 weeks after delivery and ambulant hysteroscopy after next 2 months without anesthesia. Described intrauterine pathologies (residual tissue and adhesions) were managed in one step. Results: In cases with suspect ultrasound finding, the retained trophoblastic tissue was diagnosed by hysteroscopy in 66 percent vs. in 96 percent with sensitivity 85 percent and specificity 85 percent. Patients with intrauterine adhesions had normal ultrasound finding in 74 percent and it did not recognized patients with severe adhesions in 94 percent. Clinical signs had generally 72 percent of patients with diagnosed left residual tissue. Th1/Th2 cytokine gene polymorphisms in patients with urine fibroid

Background: Uterine fibriod (UF) or leiomyoma is the most frequent benign tumour upon lower genital tract and represents the most frequent indication for hysterectomy. The aetiology remains still unknown. The genetic factors contributing for the development of UF are being intensively investigated. The aim of our study was to look for possible genetic markers which could be used as prognostic tools for evaluation of an increased risk for development of UF. Methods: The study group enrolled 102 patients diagnosed with UF and 145 healthy controls. Ultrasonographic examination of the pelvis was performed and a single blood sample was taken in all women. Histological verification followed the surgery in the patient group. The principal of the cytokine gene polymorphisms detection is based on PCR reaction with sequence-specific primers. Results: A large spectrum of Th1/Th2 cytokine gene polymorphisms in patients with uterine fibroid was compared with control group. The frequencies of the majority of tested cytokine gene SNP in the patient cohort were not statistically different from the cytokine SNP in the control group. Identification of biomarkers involved in the pathophysiology of gestational diabetes mellitus ABSTRACT Gestational diabetes mellitus is a disorder of glucose metabolism that occurs in pregnancy and resolves after delivery. Increasing production of pregnancy-related hormones leads to insulin resistance which is not adequately compensated by increased insulin secretion. Since obesity is an important risk factor for gestational diabetes and is also associated with adipose tissue dysfunction and increased peripheral insulin resistance, the question arises as to what extent is the adipose tissue involved in the development of gestational diabetes. The first part of the thesis focuses on the identification of changes in plasma concentration and mRNA expression of adipokines fetuin A, fetuin B and FGF21. In our study we did not show that the presence of gestational diabetes significantly influenced the plasma concentration of fetuin A, fetuin B and FGF21 during pregnancy. An important finding was that women who had pregnancy complicated with gestational diabetes had a significantly higher concentration of FGF21 several months after delivery in comparison to healthy pregnant women.
