Analysis of functional interaction between PKN3 kinase and CARMIL1 protein

Cancer cell motility and cytoskeletal rearrangements are crucial for metastasis formation. These complex changes involve multiple cellular processes affected by many different proteins. One such protein is the Ser/Thr kinase PKN3. This kinase has been shown to be essential for metastasis formation in some aggressive types of breast and prostate cancer. Interestingly, the PKN3 kinase is not only important in malignant cancers but also in normal tissues. In endothelial cells, the PKN3 kinase can alter their adhesion, or in osteoclasts it helps to promote bone resorption. The effects of the PKN3 kinase on cancer malignancy and cell motility are well documented, but the mechanism behind these effects is still unclear. Therefore, our laboratory seeks to identify novel substrates and interaction partners of the PKN3 kinase. This work focuses on a novel potential substrate of the PKN3 kinase, CARMIL1. This protein is involved in actin cytoskeleton rearrangements by regulating actin polymerisation and thus cell motility. Here we provide evidence that the PKN3 kinase interacts with CARMIL1. Practical part includes: Detection of premature ventricular complexes and detection of bundle branch blocks using statistical analysis of QRS complex and their classification with K-nearest neighbors
Interpreting the learning process of an atrial fibrillation classifier

In the theoretical part of the bachelor thesis the problems of atrial fibrillation (AF) detection and principles of convolutional neural networks (CNN) are discussed. Next, two classifiers were created in the practical part. The first was designed to classify sinus rhythm, atrial fibrillation and other pathologies, while the second further distinguished the category "atrial fibrillation" according to whether it was present in the whole recording or only in a part of it. The resulting accuracies are 82.12 \% and 85.14 \% for the first and second classifiers, respectively.

This bachelor’s thesis examines ECG classification using convolutional neural networks. Two models were created -the first one for classification of sinus rythm, atrial fibrillation and other pathologies and the second one for classification of sinus rythm, atrial fibrillation in the whole record, atrial fibrillation in part of the record and other pathologies. Both neural networks were implemented in Python programming language.
Detection of atrial fibrillation using intracardiac ECG

Atrial fibrillation is one of the most common heart rhythm disorders. It is caused by irreg- ular activation of the atria, which causes uncoordinated atrial contractions with a very high frequency, and therefore its early diagnosis is very important. The aim of this bachelor thesis is the automatic detection of atrial fibrillation in intracardiac recordings. The automatic detection allows finding the sections with atrial fibrillation and thus medi- ate a faster diagnosis. Testing of the developed algorithm was carried out on the available database of paediatric patients who underwent electrophysiological examination at the Children’s Hospital of the University Hospital Brno in the Czech Republic.
Advanced sleep scoring

This diploma thesis focuses on classification of sleep stages using a smart watch. Two signals were used – heart rate and acceleration. A model called TinySleepNet composed of convolutional neural network and LSTM was chosen for this task. The model was first trained for the classification of five sleep stages using only heart rate, achieving F1 score of 49%. Acceleration was converted into an SVM vector, on which the second model was trained. Due to the lack of information in the SVM vector, the model was trained only for binary classification of wake/sleep, achieving F1 score of 62.3%. Both SVM and heart rate were combined in the last model. The classification of heart rate and SVM vector into five sleep stages achieved F1 score of 51%. The calculated parameters evaluating sleep quality were then compared with data evaluated by a sleep expert.

This bachelor thesis consists of two parts - theoretical and practical. First chapter of the theoretical part deals with the issue of renaissance woman in society. It shows a woman in role as a virgin, wife and widow; it describes education that every woman had to go through; and it looks at noble woman in point of view of aristocracy. Second chapter of the theoretical part is dedicated to a specific noblewoman Polyxena of Lobkowicz, maden name Polyxena of Pernstein and describes her lifeway. Last chapter of the teoretical part deals with the problematics of perception of visual impairment and focuses on the methodology of compensatory aids, especially haptic models for visually impaired ones. Practical part of this bachelor thesis has the goal to transform the Polyxena's paintings into the haptic model in the form of relief which is primarily intended for the blind and partially sighted.

As the construction of villa is especial oportunity how to create own housing so it is important to carefuly weight all building steps.
