Criminal proceedings against legal entities
Novotná, Eva ; Jelínek, Jiří (advisor) ; Vanduchová, Marie (referee)
This diploma thesis discusses the criminal proceedings against legal persons. The main idea of this diploma thesis is to describe single juridical institute of criminal procedure which is specific for proceedings against legal persons. For better understanding is necessary to find and understate all the sources of law. The primary authorities which will be used in the criminal proceedings are above all Act of Criminal Procedure against Legal Persons, Rules of Criminal Procedure and also Constitution of the Czech Republic and Charter of Fundamental Rights and Freedoms. It is also important to looking the sources at international level. Fundamental source is European Convention on Human Rights. The thesis deals with institutes of criminal law specific for legal entities, like imputability, a person committing a crime or enumeration of criminal acts for which can be affected legal entities. The main chapter contains analysis of each section from special part of Act of Criminal Procedure against Legal Persons. Subchapters discuss some of elemental principles in criminal proceedings as local jurisdiction in criminal proceedings and principle of joint trial of a criminal case, then provisional and protective measures, procedure for the cancellation, termination and conversion of the legal entity during...
New Technologies in Education from the Phenomenological Point of View
Novotná, Eva ; Hogenová, Anna (advisor) ; Dvořáková, Michaela (referee)
This diploma thesis describes connection between education and technology from the phenomenological point of view. The hypothesis of this diploma thesis is the idea that new technologies influence changes in perception of the real world. There are students who use new technologies - the aim of the thesis is to evaluate the influence of technologies in educational process and changes in their perception of the world. The thesis describes the approach to education (Jan Patočka), new technologies in education (Branislav Frk, Marc Prensky), phenomenology and technologies (Maurice Merleau-Ponty, Paul Virilio) from the point of view of chosen authors. The thesis shows how perception of young generation (that Prensky called digital natives) is changed compared to the older one called digital immigrants and what consequences can this change cause. The thesis describes threats arising from the use of technologies and how to precede. KEYWORDS phenomenology, upbringing, education, technologies, media, perception
Investigation of proteolytic enzymes expression in different tissues at the transgenic animal model of Huntington disease by means of biochemical and immunohistochemical methods
Kocurová, Gabriela ; Dršata, Jaroslav (advisor) ; Novotná, Eva (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Bc. Gabriela Kocurová Supervisor: Prof. MUDr. Jaroslav Dršata, CSc. Title of diploma thesis: Investigation of proteolytic enzymes expression in different tissues at the transgenic animal model of Huntington's disease by means of biochemical and immunohistochemical methods Background: Huntington's disease (HD) is a neurodegenerative disorder that is caused by an expansion of a polyglutamine (polyQ) domain in the huntingtin (Htt) protein. Because it is known that mutant Htt and especially its small proteolytic fragments are toxic to neurons (particularly those in the striatum and cortex), it has been suggested that proteolysis of mutant huntingtin (mHtt) might play an important role in HD pathogenesis. Therefore, the aim of the present study was to examine the expression of endogenous and mtHtt and possible participation of the proteolytic enzymes from the group of caspases, matrix metalloproteinases (MMPs), kallikreins (KLKs) and calpains in HD pathology of brain tissue. Methods: In this study we used WT and TgHD minipigs for N-terminal part of the human mtHtt (548aaHTT-145Q, both F2 generation, age 36 months; F3 generation, age 48 months in additional experiment), R6/2 mice were used as...
Biochemical diagnostics of mitochondrial disorders in muscle tissue
Bradová, Radka ; Dršata, Jaroslav (advisor) ; Novotná, Eva (referee)
Mitochondrial diseases are serious hereditary metabolic disorders caused by insufficient energy function of mitochondrion. The cause of the disorders is biogenesis and structure of oxidative-phosphorylation system (OXPHOS) fault. Mitochondrial diseases have extensive clinical implications with a very poor prognosis. Incidence of these diseases is approximately 1: 5000 and thus they ranks among one of the most common congenital metabolic disorders. Given it that treatment is still unavailable, the only way to help the affected families is properly and early find out the true cause of the mitochondrial diseases to ensure the possibility of genetic counseling in affected families. In recent years, although novel genes that are responsible for the MO are discovered, the majority of the genes causing the disease still remains unknown. The only possibility is the analysis of mitochondrial functions at the biochemical level. This thesis focuses on the analysis of biochemical parameters of mitochondrial enzymes in skeletal muscle sample. The thesis describes the biochemical diagnosis process involving the collection of tissue, isolation of mitochondria and analyses for enzymatic activity of OXPHOS complexes. Mitochondrial fraction of native muscle was isolated by the differential centrifugation. The...
Laboratory investigation of thyreoidea in pregnancy
Stehnová, Jana ; Dršata, Jaroslav (advisor) ; Novotná, Eva (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Bc. Jana Stehnová Supervisor: Prof. MUDr. Jaroslav Dršata, CSc. Consultant: MUDr. Mgr. Sylvie Špitálníková, Ph.D. Title of diploma thesis: Laboratory investigation of thyreoidea in pregnancy Thyroid diseases are very common in the Czech Republic. It is reported that up to 46% of the population suffer with thyroid diseases. Women are afflicted with the desease 4 to 8 times more often than men, and particularly in pregnancy, hormones of the thyroid gland play an important role in developing, maintaining and progress of pregnancy. In this work, we focused on creating the reference intervals for TSH, fT4 and antiTPO in the first trimester of pregnancy, which are recommended as the main markers of thyroid assessment of thyroid function during pregnancy. Reference intervals created by us were compared with the recommended reference intervals of the supplied Roche diagnostics. Our reference file was compiled from a group of 81 pregnant women and divided into individual weeks of the first trimester (weeks 11, 12 and 13). Reference intervals were calculated according to the recommendation, such as 2.5th and 97.5th percentile of our reference group for each week and then the resulting average...
Optimization of human carbonyl reductase overexpression in Escherichia coli
Ježková, Veronika ; Novotná, Eva (advisor) ; Zemanová, Lucie (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Bc. Veronika Ježková Supervisor: RNDr. Eva Novotná, PhD. Title: The optimization of expression of the selected human carbonyl reductases in Escherichia coli The present thesis called The optimization of expression of the selected human carbonyl reductases in Escherichia coli is divided into two parts. The first one is theoretical and the second one is experimental. The theoretical part presents a brief information summary about selected enzymes: carbonyl reductases 1 (CBR1), quinone oxidoreductase 1 (NQO1) and DHRS2 (Hep27). It also includes a theoretical summary of laboratory methods which were used in the experimental part. The experimental part aims to optimize the express conditions of the selected enzymes in the bacterial expression system Lemo21(DE3). The plasmid pET-28b(+), which encodes the selected enzymes, is transformed by a thermal shock into competent bacterial cells Lemo21(DE3). In these cells a protein expression was carried out at different concentrations of L-rhamnose. We also purified the expressed recombinant protein NQO1 by the affinity chromatography method. The success rate of protein expression was examined by SDS-PAGE or Western Blot. Based on the obtained results...
Management and description of geographic data in the field of environment of the Czech Republic
Zrzavecká, Lada ; Souček, Martin (advisor) ; Novotná, Eva (referee)
This thesis describes and analyzes the current state of environmental management of geographic data within geoinformation infrastructure of the Czech Republic in response to European legislation. The work is divided into four main parts. The first part deals with geoinformation infrastructure of the Czech Republic in its basic components due to the efficiency of the management of geographic data. Another part deals with the general characteristics of geospatial data underlying the description methods, organization and retrieval in the next section. The final part of its qualitative research describes in detail the management of geospatial data in the environmental sector, which is affected by the phenomenon of the implementation of INSPIRE and draws conclusions and recommendations. The work also supports the analysis of the availability of data sources for the implementation of the INSPIRE directive, the questionnaire used as a guide for research purposes, to illustrate the services Geoportal three pictures.
