Electrical stability of the heart during hypothermia-induced potassium plasmatic level changes and after modulation of the autonomic nervous system by renal denervation.
Kudlička, Jaroslav ; Kittnar, Otomar (advisor) ; Nováková, Marie (referee) ; Danzig, Vilém (referee)
Malignant ventricular arrhythmias are a common cause of sudden cardiac death. Moderate therapeutic hypothermia (MTH) is routinely used in post-resuscitation care for anticipated neuroprotective effects. However, the safety of MTH in terms of the electrical stability of the heart has not been satisfactorily proved yet. Also, the increased sympathetic tone in patients with heart failure contributes to a higher incidence of malignant ventricular arrhythmias. The aim of this work was to verify the safety of MTH as regards the inducibility of ventricular fibrillation (VF) in the pig biomodel, especially in relation to spontaneous changes in the kalemia and QT interval. Furthermore, we assumed that renal denervation (RDN) could reduce the inducibility of VF. In the first part of the thesis, the extracorporeal cooling was introduced in fully anesthetized swine (n = 6) to provide MTH. Inducibility of VF was studied by programmed ventricular stimulation (8 basic stimuli with up to 4 extrastimuli) three times in each biomodel under the following conditions: during normothermia (NT), after reaching the core temperature 32 řC (HT) and after another 60 minutes of stable hypothermia (HT60). VF inducibility, effective ventricular refractory period (ERP), QTc interval, and potassium plasma level were measured. In...

Nováková, Marie ; Němec, Zbyněk (advisor) ; Hájková, Vanda (referee)
This diploma thesis deals with respite service and issues with providing respite services to parents of children and adults with autism and mental retardation. The opening chapters focus on autistic spectrum disorder in combination with mental retardation and potential challenging behaviour. This work describes therapeutic process for persons with autism and mental retardation and it also describes the issues with providing social service to these persons. This thesis focuses mainly on respite services. The empirical part describes the availability of respite services for families with autistic person and mental retardation in the Czech Republic.
Children's groups in counseling in the context of the active cooperation with parents
Nováková, Marie ; Pavlas Martanová, Veronika (advisor) ; Heider, David (referee)
This diploma thesis discusses the children's groups in counselling in the context of active parenting. It maps experiences of parents in groups from the point of view of mothers and therapists. The theoretical part focuses on the definition of the group psychotherapy, the specifics of child psychotherapy and the support groups. Furthermore, it focuses on the experiences of a parent of a problem child and its active role in the process of a child psychotherapy. It also considers the work of a therapist. The qualitative research is presented in the empirical part. Semi-structured interviews with 5 mothers and 4 therapists were conducted. These interviews were analysed based on the method of creating thematic categories. The evaluation of the data is also inspired by the method of grounded theory. The results show that the views of parents and therapists are the same in most areas. Both groups perceive the importance of a parent participating in the group. The group acts therapeutically for both the child and the parents. This affects the process of the whole therapy. KEYWORD: child psychotherapy, group psychotherapy, support groups, parent of a problem child, cooperation with parent
Influence of cervical spinal cord lesion on cardiovascular fitness
Machač, Stanislav ; Kolář, Pavel (advisor) ; Vilikus, Zdeněk (referee) ; Nováková, Marie (referee)
Traumatic damage to the cervical spinal cord has a dramatic impact on determinants affecting cardiovascular fitness. In tetraplegia, in addition to diminished skeletal muscle function, impaired sympathetic cardiovascular innervation together with a dysfunction of catecholamine release is typically present. Individuals with tetraplegia are at a high risk of long-term inactivity, which increasingly contributes to the development of common civilization diseases. The aim of the current dissertation project is to investigate the cardiovascular fitness in males with cervical spinal cord injury (n=20) as compared to able- bodied males (n=27). Both groups completed exercise testing on an arm-crank ergometer. Arterial blood pressure response to exercise, peak oxygen consumption and peak heart rate were the primary outcome variables. In the first part of the study, a graded maximal arm- crank ergometer test to the subjective exhaustion was performed for the inter-group peak exercise comparison. The peak power dramatically differs between the groups and could be an important confounding factor limiting the evaluation of a physiologic response related to the equivalent physical loading. Therefore, in the second part of the study, individuals of the control group completed a second graded arm-crank ergometer...
Expert teachers perspective on bullying and disturbed relationships in the classroom
Nováková, Marie ; Mudrák, Jiří (advisor) ; Horková, Lenka (referee)
This thesis focuses on the view of experienced teachers on bullying and disturbed relationships in the classroom. The theoretical part describes the characteristics of bullying and its typical characters. It defines the main protagonists of bullying. It also describes the procedures for dealing with bullying, investigation, intervention and prevention. The theoretical part also describes the role of the teacher, the definition of his profession and experiences. The empirical part presents a subjective view of experienced teachers on bullying and disturbed relationships in the classroom. It focuses on their personal experience with the phenomenon. It maps the tools and strategies that experienced teachers use to dealing with bullying and its prevention.
Regulace receptorů spřažených s G proteiny Studie muskarinových a β-adrenergních receptorů u M2KO myší
Beneš, Jan ; Mysliveček, Jaromír (advisor) ; Kuncová, Jitka (referee) ; Nováková, Marie (referee)
(in English): The aim of the work was to perform in-depth analysis of M2KO mice both at baseline and upon a challenge with a cold stress and to explore the role of opposing receptors (i.e. adrenoceptors) in adaptation to lacking M2-receptors in the heart. We have performed receptor binding studies, study of receptor gene expression, echocardiography, telemetric monitoring of heart rate, body temperature and activity, heart rate variability and biorhythm analysis, analysis of heart rate response to the application of drugs (carbachol, atropine, isoprenaline, propranolol), assessment of adenylyl cyclase and NO synthase activity, measurement of catecholamine blood concentration and gene expression of catecholamine-synthesizing enzymes. We have found that the disruption of M2-receptor gene caused a compensatory decrease of cardiostimulatory β1-adrenoceptors and β2-adrenoceptors with corresponding down-regulation of the gene expression, M3-receptors were down-regulated as well. Missing M2-receptors were functionally replaced by the main cardioinhibitory β3-adrenoceptors that were up-regulated, not by cardioinhibitory M4-receptors. β3-adrenoceptors were found to signal through adenylyl cyclase instead of NO synthase. All these changes were found in the left ventricle only, so heterologous regulation is...
Introduction of microbiological diagnostic agents of invasive infections - Bartonella henselae and Bartonella quintana
Nováková, Marie ; Melter, Oto (advisor) ; Chrenková, Vanda (referee)
Bartonellae are agents of infections with varied clinical symptomalogy, including serious, such as endocarditis. Direct detection of Bartonella so far in the Czech Republic and therefore does not need to address this issue. Proper diagnosis of these infections is a prerequisite for targeted antibiotic therapy for patients. The genus Bartonella today are more than 20 kinds of bacteria. They are small Gram-negative rod-shaped bacteria that cause the new emerging infections such as cat scratch disease (CSD), showing the typical regional lymphadenitis, as well as bacillary angiomatosis (BA), whose typical symptoms are skin and subcutaneous nodules (nodules). They also cause a disease called trench fever or South American bartonelosis. In the experimental part of the work was performed genotypic and phenotypic methods for the diagnosis of these bacteria, specifically on two types of Bartonella, Bartonella henselae and Bartonella quintana . The results show that by suitable methods to diagnose Bartonella are amplification genus- specific genes ribC and ftsZ, further embodiment of a method pulsed electrophoresis (PFGE) as typing method and the determination of susceptibility to antibiotics, the most appropriate testing showed E-test. This work was carried out optimization of genotypic and phenotypic...
