Pentoses as energy sources of Mastigamoeba balamuthi
Novotná, Monika ; Verner, Zdeněk (advisor) ; Čepička, Ivan (referee)
Mastigamoeba balamuthi is a free-living amoeba, a sister group to one of the most widespread parasitic agent Entamoeba histolytica. During its lifetime, M. balamuthi can utilize at least four different energy sources to produce ATP. In addition to glucose, a standard cultivation energy source, the amoeba can utilize pentoses xylose and arabinose, as well as an amino acid L-arginine. In this work, energy source preference was assessed. Morphological and physiological characteristics including cell growth and size, motility, number of nuclei and metabolic end products were evaluated. It was concluded that glucose appears to be a less preferred energy source than pentose upon evaluating statistically significant differences. Therefore, it was proposed to replace the commonly used hexose with the preferred xylose in the culture medium of M. balamuthi.
Censor and writer Jan Nepomuk Václav Zimmermann (1788-1836)
Novotná, Monika ; Marek, Jindřich (advisor) ; Šípek, Richard (referee)
This work exposes, evaluates and summarises academic and professional career of Jan Nepomuk Vaclav Zimmermann (1788-1836), who worked as a scriptor and a censor. It reviews his censoring and book-examining skills across all fields of book culture. It investigates and evaluates whether contemporary critical testimonies depict Zimmermann's rather negative character rightfully and accurately or not. Zimmermann devoted his time to examining and censoring Czech books and also Hebrew books. As a scriptor at University library, he was responsible for collecting, archiving and organizing manuscripts into catalogues. These catalogues have been filed in National Library for many years. Palaeographical analysis of the catalogues will bring in new facts concerning Zimmermann's autorship. This thesis also presents a list of Zimmermann's work with its historical critical evaluation and compares it to the recent research. Keywords: Jan Zimmermann, Manuscript Catalogues, University Library, Censorship, Censoring Hebrew Literature, Censoring Czech literature, of the Cross with the Red Star, Reformation of Catholic Church under the reign of Joseph II
The determination of fatty acid levels in the tissues of healthy, suddenly deceased persons and polymorbid patients
Novotná, Monika ; Kuchařová, Monika (advisor) ; Bárta, Pavel (referee)
Charles University Faculty of Pharmacy in Hradec Králové Department of Biophysics and Physical Chemistry Candidate: Monika Novotná Supervisor of Diploma Thesis: Mgr. Monika Kuchařová, Ph.D. Title of Diploma Thesis: The determination of fatty acid levels in the tissues of healthy, suddenly deceased persons and polymorbid patients The aim of the thesis was to determine the levels of 14 fatty acids in tissues of suddenly deceased, otherwise healthy individuals and in the group of polymorbid, chronic patients. It was a comparison of fatty acid levels in seven tissues of the human body: subendocardial left ventricular tissue, liver parenchyma tissue, kidney cortex, adrenal tissue, skeletal muscle, abdominal subcutaneous adipose tissue, and brain tissue. Each group included 10 deceased patients. The theoretical part incudes fatty acids as the main component of lipids. It deals mainly with the group of polyunsaturated fatty acids and their relation to pathologies in the human body. Gas chromatography, thanks to which we analyzed the fatty acid spectrum, is also described. The experimental part consists of the basic characteristics of the research groups, the description of the workflow and the results. Statistically processed data are divided into categories by individual fatty acids and other monitored...
Virulence factors of Entamoeba histolytica
Novotná, Monika ; Tachezy, Jan (advisor) ; Tůmová, Pavla (referee)
The parasitic protist Entamoeba histolytica causes intestinal disease called amoebiasis (amoebic dysentery), which is one of the most significant diseases worldwide, mainly in developing countries. The goal of this bachelor thesis is to summarize current knowledge about virulence factors of E. histolytica. It is primarly focused on adhesive lectin Gal/GalNAc, cysteine proteases, phagocytosis of apoptotic cells, amoebapore forming pores in the membranes of the target cells and trogocytosis. Keywords: virulence factors, Entamoeba histolytica, parasite, protist, amoebiasis, lectin Gal/GalNAc, cysteine proteinase, phagocytosis, trogocytosis, amoebapore

Graphical sign in the context of school and town
Novotná, Monika ; Kafková, Helena (advisor) ; Raudenský, Martin (referee)
NOVOTNÁ, Monika. Graphic sign in context of school and town. Prague, 2014.Thesis. Charles University in Prague, Faculty of Education, Department of Fine Arts. Supervisor Mgr. Helena Kafková, 90 pages. In the theoretical part I focus on a sign, the graphical sing. I sort out its forms. I present the sing as a nonverbal system of communication and a way of labelling objects. I search for the connections and contradictions of the existence of legal [traffic signs, orientation signs, signboards] and illegal graphic signs [graffiti and street art] in the public space. In the didactic part I suggest and realize the art projects where some technics of graffiti and street art are used. The artistic themes: SING - LETTER - PICTOGRAM - PUBLIC SPACE are processed at my art lessons at both primary and secondary schools. We discuss the meaning of these words in the public space. The part of the thesis is a research study among the pupils of a primary and secondary school which tries to point at either different or the same art perceptions by the pupils and their mental constructions of these. There I try to connect the street art and graffiti with the topics of private property, vandalism and personal attitudes versus the state and society. I offer to the children the question of personal freedom of an...
