The effect of chronic cold on Ca2+-ATPase (SERCA2) in rat heart

Acute cold exposure is a significant stressor activating heat production by shivering after the prolonged exposure cellular oxidative stress increases. Chronic exposure to cold lasting at least 2 weeks leads to the development of cold acclimatization. The main thermogenic role is taken over by non-shivering thermogenesis taking place in brown adipose tissue, which significantly increases its weight due to cold. Cardiac hypertrophy, hypertension and impaired renal function are frequently observed pathologies of acclimatization at 4-5 řC. Our laboratory recently introduced a model of mild chronic cold acclimatization at 8 řC, during which no damage to the heart or kidneys occurs and has proven cardioprotective effect on reducing infarct size. Hence, the influence of this cold acclimatization model on the other cellular and molecular processes needs to be investigated. The cardioprotective effect of cold acclimatization includes changes in β-AR signaling, activation of anti-apoptotic pathways or augmentation of the antioxidant system. Analysis of PKCα Influence on Cancer Cell Invasion.

7 Abstract Protein kinase C alpha (PKCα) is a serine/threonine protein kinase. PKCα is an important protein regulating cell polarity, protein secretion, apoptosis, cell proliferation and differentiation and tumorogenesis. Previous research has shown a role of PKCα also in a cancer cell migration and cancer cell invasion. The aim of this study was to investigate the role of protein kinase C alpha (PKCα) played in amoeboid mode of cancer cell invasion. We showed that higher expression of PKCα resulted in mesenchymal-amoeboid transition of K2 and MDA mesenchymal cancer cell lines, which was accompanied with decreased cancer cell invasive capability in 3D collage matrix. PKCα overexpression had no effect on the cell morphology of A375m2, however, the results showed a trend in increased invasive potential of A375m2 cells. Conversely, the expression of dominant-negative PKCα resulted in amoeboid-mesenchymal transition of A375m2 cells, and it was associated with decreased invasive potential of K2 and MDA cell lines. Furthermore, a linkage between PKCα and phosphatidylinositol 3-kinase (PI3K) was tested. The theoretical part briefly describes the anatomy and kinesiology of the upper limb with a focus on the hand and wrist, and then presents a summary of the most common fractures in this body part. The last and the longest chapter is about the possibilities of occupational therapy for patients fractures of the distal radius and hand, especially in the period after removal of fixation. Occupational therapy has a very wide application for these patients. Functional disorders of the hand and wrist, which are a frequent complication of fracture treatment, are often closely related to the limitation of self-sufficiency and have a great impact on the patient's quality of life. In the Czech Republic, there is a shortage of outpatient occupational therapists. The involvement of telerehabilitation appears to be a possible solution to the lack of therapists. These methods are already being used with a good effect around the world. The aim of this thesis is to compare the possibilities of telerehabilitation against conventional occupational therapy in patients with ABI. A partial goal is to undertake a SWOT analysis of telerehabilitation and evaluate the communication platform eAmbulance. There are four single case studies. The patients are aged between 27-81 years (two women and two men) with different severity of right-sided hemiparesis. These patients underwent interventions of 60 minutes per week for a period of four weeks. The content of each therapy was different. People with a disorder of these functions tend to have problems, for example with starting an activity and planning individual steps, or are unable to complete the activity correctly and efficiently. Executive functions are localized primarily in the area of the frontal lobes and when they are damaged, an executive deficit is manifested. Clinically, the impairment of executive functions manifests itself in very diverse ways. The applicant for this examination can be the Labor Office, the employer or the client himself. The prevocational assessment examination is attended by an interprofessional rehabilitation team, an integral part of this team are doctors, as well as non-medical professions, including an occupational therapist. The prevocational assessment examination is a very important component of the entire prevocational assessment, as it is the most time-consuming part of this examination.The examination takes place in one of thirteen specialized Prevocational Assessment centers in the Czech Republic. All centers must meet personal, substantive and material standards. Conclusions from individual examinations are presented by experts from the team at a case conference. The secondary goal is to compare the objective evaluation of motor symptoms with the subjective one. The theoretical part summarizes findings of Parkinson's disease.
