Detecting biomarkers of extremophiles in Martian analogues

The main focus of this thesis is on the examination of endolithic microorganisms within gypsum from Sicily. This region is of particular interest because gypsum outcrops are common and accessible there, but the environment is not considered to be extreme in terms of solar radiation or other factors. The research aims to detect endolithic biomarkers and analyse their distribution in their natural environment, to identify endoliths with a focus on phototrophs, and to assign the detected biomarkers to their producers. One of the key tools used in this study was a portable Raman spectrometer with a 532 nm laser, used for the detection of carotenoids under field conditions. Further, the Raman fingerprints of scytonemin and gloeocapsin can be used for the taxonomic identification of some cyanobacteria. With the use of a combination of different excitation wavelengths (445nm, 532 nm, 780 nm), it was possible to detect good quality spectra of various pigments in situ, conditions also in isolated cells and colonies. The results of the molecular analysis also suggest that gypsum habitats in non-extreme regions have a similar microbial composition as those found in extreme regions; i.e., Cyanobacteria being the most abundant group of these microorganisms. Death mediated to pupils through children's literature

This diploma thesis deals with the topic of death and its mediation to pupils of younger school age. The aim of the thesis is to find and introduce appropriate literature for younger school-age pupils and to offer ways of conveying a complicated topic through the literature to younger school-age pupils. The theoretical part of this thesis deals with the definition of death and the concepts related to it. It also defines the concept of the younger school age and describes the emotional development of the child at this age. The thesis also provide space for the definition of literature for the younger school age and for the characteristics of publishing houses concerned with publishing the books with the theme of death for children of younger school age. The theoretical part also introduces, through brief annotations, selected titles of literary works conveying the theme of death. At the end of the theoretical chapter of this thesis, the background of the RWCT program is described, the methods of this program are introduced, and the three-phase E-U-R learning model used in the preparation of the lessons of the practical part of the thesis is specified. Carotenoids of snow algae as biomarkers for exobiology: Raman spectroscopic perspective

The major aim of this thesis is critical evaluation of Raman spectroscopy in the detection of carotenoids of extremophiles, namely snow algae. Extremophilic microorganisms play an important role in exobiology since they set hypothetical boundaries for the presence of life on Earth. Raman spectroscopy will be a part of two mission to Mars. Here, a laboratory Raman microspectrometer was used for the analysis of 11 samples of snow algae from different locations over the years 2002-2017. The acquired spectra were compared with HPLC/UV-VIS analysis. The results showed that the ability of Raman spectroscopy to discriminate between structurally slightly differing carotenoid pigments or several carotenoids in an admixture is limited. Contrary, HPLC/UV-VIS permitted to detect various structurally similar carotenoids (and chlorophylls). However, HPLC/UV-VIS worked with overall pigment extracts during which some structural information can be lost. Raman microspectrometer allowed analysis of cells in different life-cycle stages and thus several various spectra could be studied. Raman microspectrometer was therefore more suitable for carotenoid detection in mixtures of various life-stages than HPLC/UV-VIS. This master's thesis focuses on phenomen of sharing economy in France and its impact on tourism. It analyses the current situation of sharing economy in France and emphasizes the role of France on the market of sharing economy in comparison with other countries, especially with the Czech Republic. The following practical part concentrates on the different knowledge and experience with services in sharing economy between the people living in France and in the Czech Republic. The main aim of this comparison is to emphasize the difference between these countries and determine the situation of sharing economy in France more deeply.

Bachelor thesis is focused on the issue of indemnity, confirmation and debt collection of overdue receivables. In the theoretical part incorporates the topic of the legal framework and defines the economic, accounting and tax impact of overdue receivables on Capital Company. It is focused on the analysis of the selected company receivables and contains appropriate proposals to solve the problem.
Marketing strategy of the hotel Jana a.s.

The goal of this thesis is to create a marketing strategy for the hotel Jana a.s. located in Přerov (cental Moravia). It is divided into theoretical and practical part. The theoretical part introduces the basic marketing terms and the practical part applies them in practise on the example of hotel Jana a.s. Marketing research is included in order to analyze the current situation within internal and external environment of the hotel. The data from the hotel and his annual reports were used together with data from Czech Statistical Office and responsible ministries. Financial situation is analyzed using horizontal and vertical analysis of assets and liabilities plus some selected indicators (ROA, ROE etc.). The strategy is build and marketing mix is designed based on the outcome of all analyses.
Daňová zátěž a její vliv na ekonomiky EU

This diploma thesis deals with tax differences, which occur among states of the European Union. It focuses on the influence of taxes to at-tract foreign direct investment and further impact on the increase of eco-nomic development. Using correlation analysis it quantifies these relationships and proves that generally there are in EU weak links be-tween variables. To sum up, low corporate taxation is a factor which attracts foreign direct investment and foreign direct investment can be under certain circumstances the factor increasing the economic level of the country. Weaker and less obvious results are found for region of Central and East Europe and so called tax havens probably due the po-sition of alternative factors including qualitative characteristics. In addition, these results are discussed in context of recent European effort to coordinate and harmonize corporate taxation.
The Centre JANA Domazlice and it´s client´s ethical motivational aspects of becoming a prostitute

Women, who earn a living as prostitutes, have their own needs, hold certain values and want to fulfil their objectives. The purpose of my work is to make these values and objectives specific and compare the results with fact given in specialized literature. The first part of my work makes basic for the quantitative research. There are chapters about phenomena which can influence the step in prostitution. Secondly, there is a description of motivation phenomenon. Additionally there is a chapter about specific motivation aspects leading to prostitution behaviour. The practical part shows the summary of theoretical data and creating assumptions. All the information are reflected in questions which are involved in these half-structured interviews. All interviews were hold with clients of JANA Centre in the Czech Republic and with clients of Kassandra Centre in Germany. There is the congruence between data gained from my quantitative research and data published in specialized literature. Women working for sex business are not a group which would be easily generalized. Each person experiences various circumstances and all these conditions diversity of their behaviour. Respondents advocate similar values and want to reach suchlike objectives. These is first of all a normal family, a normal job and own children. This term "normal" means for these women a full family and any other job except prostitution.
