Apoptosis of pancreatic β-cells induced by endoplasmic reticulum stress and its mechanisms in type 2 diabetes

Increasing incidence of type 2 diabetes represents one of the principal threats to human health in the 21st century. Strong evidence indicates that the rise in incidence of type 2 diabetes is correlated with increasing levels of obesity and that important factor playing role in the development of this disease is an elevation in circulating glucose and fatty acids. Chronically increased concentration of these nutrients was shown to induce apoptosis of pancreatic beta-cells that subsequently contributes to diabetes progression. Despite intensive research, molecular mechanisms underlying this beta-cells loss are still unclear. However, there is increasing evidence that one of the key processes involved in glucose and fatty acid-induced beta-cell death is induction of endoplasmic reticulum stress. The aim of this work is to summarize the recent knowledge about induction of apoptosis by endoplasmic reticulum stress in pancreatic beta-cells in relation to type 2 diabetes.
The role of fatty acids in apoptosis induction in pancreatic beta cells

Type 2 diabetes belongs to lifestyle diseases. To its development contributes also the decrease in functional  cell mass caused particularly by long-term action of increased blood levels of fatty acids and glucose. Experiments from recent years demonstrated that apoptosis of pancreatic  cells is induced especially by saturated fatty acids, in contrast to unsaturated fatty acids which do not exert significant cytotoxic effect and they are even able to inhibit the proapoptotic effect of saturated fatty acids. Despite intensive research, the mechanisms of fatty acid-induced apoptosis of pancreatic  cells are not convincingly explained. Understanding of the processes that lead to destruction of functional  cells could enable development of new therapies for type 2 diabetes treatment that would be targeted directly at one of the causes of this disease. The aim of this work is to summarize the recent knowledge about mechanisms by which fatty acids induce and regulate apoptosis of pancreatic  cells.
The role of autophagy in apoptosis induction by fatty acids in pancreatic beta cells.

Type 2 diabetes mellitus represents a metabolic disease reaching epidemic dimensions in the 21st century. Fatty acid-induced apoptosis of pancreatic β-cells significantly contributes to its pathogenesis. Saturated fatty acids (FAs) are strongly cytotoxic for β-cells, whereas unsaturated FAs are well tolerable by β-cells, they are even able to inhibit proapoptotic effects of saturated FAs when co-incubated. According to recent studies, FAs-induced apoptosis in pancreatic β-cells is partly regulated by autophagy, a catabolic process involved in the degradation and recyclation of cell components in lysosomes. The aim of this diploma thesis was to contribute to the clarification of the role of autophagy in FAs-induced apoptosis regulation. We induced apoptosis in human pancreatic β- cell line NES2Y by 1 mM stearic acid (SA) and inhibited it with 0.2 mM oleic acid (OA) co- incubated with SA. We revealed, that the saturated SA used in apoptosis-inducing concentration simultaneously inhibits the autophagic flux in pancreatic NES2Y cell line. When SA is co- incubated with unsaturated OA in concentration sufficient for inhibition of proapoptotic effect of SA, OA is also able to inhibit the block of autophagy induced by the effect of SA. The association of type 2 diabetes with obesity is apparent and dysfunction and apoptosis of pancreatic β-cells caused by elevated levels of fatty acids in circulation are considered as an important factor contributing to the development of this disease. However, molecular mechanisms that underlie these detrimental effects of fatty acids are only partially understood. The aim of this research project was to contribute to elucidation of mechanisms by which saturated and unsaturated fatty acids regulate viability and apoptosis induction in human pancreatic β-cells in vitro. Employing human pancreatic β-cell line NES2Y, we showed that increased levels of relevant dietary saturated fatty acids (palmitic and stearic acid) induce apoptosis of pancreatic β-cells, in contrast to relevant dietary unsaturated fatty acids (e.g. palmitoleic and oleic acid). We found that stearic acid-induced apoptosis is accompanied by significant activation of caspase-2, -6, -7, -8 and -9, but not by significant activation of caspase-3. Analysis of circulating markers in patients with solid tumors

Circulating cell-free DNA (cfDNA) and its tumour-derived circulating tumour DNA (ctDNA) fraction are considered an innovative prognostic and predictive biomarker in oncological diagnostics. Many studies have demonstrated higher levels of cfDNA concentration and integrity, as an indicator of the amount of ctDNA in cfDNA, in body fluids from patients with cancer diseases in comparison with healthy individuals, which suggest its potential as an effective biomarker for monitoring of the tumour dynamics. This study focused on optimisation and validation of measurement methods later used for analysis of cfDNA concentration and integrity in blood samples from patients with four different solid cancers. Two different commercial isolation kits have been tested in plasma and serum samples. Quantitative real-time polymerase reaction (qPCR) and PicoGreen dsDNA assay were optimized to effectively quantify low concentrations of cfDNA, subsequently compared to each other and to droplet digital PCR assay tested on selected samples. The concentration and integrity of cfDNA from plasma samples of breast, ovarian, colorectal and pancreatic cancer patients were evaluated. Expression and regulation of the ABC transporters in tumour cells

Estrogen signalling pathway plays crucial role in carcinogenesis of breast cancer. Estrogen receptor (ER) is a prototypical hormone receptor that upon binding its ligand, estradiol, translocates into the nucleus and turns on target genes related to cellular proliferation and survival. Although estrogen signalling physiologically supports normal breast tissue development, deregulations of this pathway contribute to development of breast tumours that are estrogen receptor dependent. One of the main obstacles in breast cancer treatment is acquired resistance to common anticancer drugs also known as multidrug resistance (MDR). The switch between chemotherapy responsive to chemotherapy resistant cell phenotype is usually accompanied by increased expression of ABC transporters, special membrane proteins responsible for export of various kinds of commonly used anticancer drugs from the intracellular to extracellular space and is also linked to the existence of cancer stem cells (CSCs). Impact of weight loss in obese subjects on the sensitivity of adipose tissue cells in relation to stress of endoplasmatic reticulum.

Adipocytokines released by the adipose tissue play an important role in the regulation of immune and inflammatory responses. In obesity their production is dysregulated, which is one of the major factors contributing to the onset of a chronic low-grade systemic inflammation representing a risk factor for the progression of other diseases, such as atherosclerosis or type-2 diabetes. The main goal of this thesis was to analyze the secretion of selected adipocytokines (adiponectin, IL6 and MCP1) by in-vitro differentiated adipocytes, isolated from the adipose tissue prior to and after a dietary intervention, and this under basal conditions and during stimulated lipolysis. In case of adiponectin, the secretion of its isoforms was analyzed too. The concentration of adiponectin, IL6 and MCP1 was determined by the ELISA method, the Western Blot method was used to determine the distribution of the adiponectin isoforms. The thesis also concentrates on the gene expression of ATF3, ATF4 and HSPA5, factors engaged in the ER stress in the course of the differentiation of adipocytes.
