Methodology of road traffic accident investigation Abstract This thesis, entitled "Methodology of Road Traffic Accident Investigation", responds to the long- term high number of road traffic accidents that result in many deaths and serious injuries. The purpose of this thesis is to provide readers with up-to-date and comprehensive information on the issues and specifics of road traffic accident investigation. The thesis also presents a number of planned innovations and proposals that could significantly reduce the occurrence of road traffic accidents and thus increase safety on Czech roads. The thesis is designed chronologically, with the introductory chapter outlining the theoretical basis and nature of road traffic accidents. In the subsequent chapters, attention is then given to typical clues, investigative situations, peculiarities of the subject matter and stimuli for investigation. The sixth chapter then deals with initial investigative tasks, in which special attention is given to the issue of detecting alcohol in road users. Methodology of Investigation of Murders Abstract This thesis examined methodology of investigation of murders. As this is a very broad topic, the focus was on selected aspects of the investigation methodology. The introduction of the thesis was devoted to murder from the perspective of law and its comparison with the crime of killing and murder of a newborn child by the mother. The nuances that differentiate them are discussed there. The second chapter focuses on the criminological characteristics of murder. It provides the reader with an insight into the characteristics of the perpetrators of murder and who the victims most often are. The following chapters deal with the individual components of the methodology itself. They describe typical clues as well as investigative situations that criminologists most often encounter in practice. Attention is also paid to the methods of crime scene examination, examination of a corpse and forensic examination. The eighth chapter discusses the planning and organisation of the investigation and the way in which criminal investigators construct versions based on the evidence and clues found, which are then tested. Interrogation as evidence in criminal proceedings

This thesis deals with interrogation as the most commonly and widely used means of evidence. The thesis focuses on the so-called questionable methods of verification of testimony, which is a controversial issue on which there are different opinions. The aim of this thesis is to describe and analyse selected methods which can lead to the verification of testimony, but the opinion on their use is still not clear. The thesis focuses on the issue of the use of these methods and the evaluation of the possibility of their use in criminal proceedings. The thesis is divided into two main parts. The first is the theoretical part, which is further divided into two large chapters. The first chapter is devoted to interrogation in general, defines this term and describes its preparation, course and documentation. It also outlines the different types of interviews, including the specific requirements for interviewing children. Finally, this chapter also covers the credibility of testimony. Cytokine networks and their impact on the immune profile of acute myeloid leukemia (AML) patients

Acute myeloid leukemia (AML) is a malignant hematological disorder characterized by aberrant expansion of blasts in the bone marrow and peripheral blood. The immune system protects the body from leukemia by eliminating transformed cells. However, in AML, the abilities of immune cells are affected both by direct contact between leukemic cells and effector cells, as well as by cytokines, metabolites and other soluble proteins that, together with the cells, form the specific AML microenvironment. The effects of cytokines and other soluble molecules in the AML microenvironment are not sufficiently described yet. This thesis aimed to optimize and implement a multiparametric flow cytometry panel for the measurement of cell populations and to implement multiparametric assays for the analysis of cytokine levels, chemokines and other soluble proteins in plasma. The following goal was to use these methods to characterize the frequency and functional phenotype of cell populations and the levels of the soluble proteins and to describe their influence on disease severity and overall survival of the patients. Investigative hypothesis Abstract This thesis deals with the topic of investigative hypothesis as a part of criminalistic science and a method of criminalistic practice. The aim of the thesis is to provide a thorough definition and essence of investigative hypothesis, to describe their types, creation, process of verification and proving of versions and last but not least to show their practical significance and use on real criminal cases. The thesis uses the methods of descriptive analysis, comparison and interpretation. In the first chapter of the thesis, the concept of criminalistic versions itself is examined in detail, first through its systematic classification, then through an analysis of its definition put forward by some of the most important figures in criminalistics. Furthermore, the first chapter is devoted to a brief analysis of the concept of the term hypothesis and its relevance to investigative hypothesis. The thesis then goes on to explore the concept further by comparing the method of creating investigative hypothesis with behavioural analysis (criminal profiling) as a method of constructing a profile of an offender. Optimizations and applications of non-linear spectral unmixing in flow cytometry

Recent advances in flow cytometry techniques enable high-throughput single-cell experiments with extensive marker sets. In order to leverage this technology the measured signal must be unmixed to recover interpretable re- sults. Current approaches to unmixing typically leverage linear deconvolution algorithms such as fitting by ordinary least squares method, that tend have issues dealing with various noise sources inherit to the data collection pro- cess. This thesis evaluates the performance of a novel non-linear approach of unmixing called nougad. For the evaluation, we have generated realistic arti- ficial data with known ground truth for testing, implemented multi-threaded version of nougad and tuned its hyperparameters using Bayesian optimiza- tion, and collected several performance metrics of nougad and the other algorithms on the testing datasets. The results show that nougad is able to outperform the tested linear algorithms making this non-linear method more suitable for practical applications and a good candidate for further refinement and optimization efforts. 1

RESUMÉ Homicide investigation methodology is certain set of instruction or aid for guiding investigators progress, but it does not mean that if the methodology is followed, there is a sure result in capture of perpetrator. It depends on every specific act performed by the investigator. Correct way of search of crime scene/corps, interrogation, expertise, apartment inspections, recognition, reconstruction and others are already dependent on human activities and their quality depends on those activities. Certain rules or limits provides law that determine all the formalities which must be completed for potential enforcement in court against offender. The first important task is to search the crime scene and the corpse, which must be thorough, because it is important to secure all traces that may be on the crime scene. It is decisive to properly ensure traces to prevent deterioration or destruction of found material and especially biological traces, where a DNA sample or other important fibers of significant probative value are found. Interrogation is an act where it is best to see that methodology is only a guide, but how good it will be depending on investigator (his preparedness and experiences) who performs it and also on the willingness of an interviewee to cooperate. Public procurement-related crimes (in Czech republic) (summary of the dissertation) Vladislav Hřebíček In his paper, the author focuses on the area of public procurement-related crimes in the Czech Republic. As there has been no comprehensive coverage on the subject so far, the paper's primary ambition is to sum up the current state of knowledge while critically considering particular resources (mainly legislation and practice of courts), overlapping significantly into the area of daily practice of detecting such crimes and gathering evidence. In the first chapter, the author provides definitions for key terms and examines the term procurement from the perspectives of its legal definition as well as its interpretation based on court practice. Furthermore, the chapter specifies what is to be understood as procurement-related crime, placing this type of crime within the broader context of economic crime. At the same time, it offers answers to the related criminological questions and presents statistical data provided by the Police of the Czech Republic and the Czech Ministry of Interior. The second chapter gives the reader a historical overview of this area of law mainly from the perspective of criminal law.
