Evaluation of basic characteristics of thermal water with separation methods

The presence of inorganic ions is one of the criteria for thermal water assessing. The most important anions that affect the water quality and allow their use for therapeutic purposes, include sulfates, chlorides, nitrates and bicarbonates. Ions (as sodium, potassium, calcium and magnesium) are the significant cations. The ion chromatography is used for separation of these substances and also some methods based on electrophoretic migration of ions in an electric field. The electromigration method of isotachophoresis was chosen for determination of selected ions in water samples collected from thermal boreholes in Pasohlávky.
Determination of musk compounds in biotic matrices

This diploma thesis deals with actual issues of the contamination in the environment with synthetic fragrances. Musk compounds are infiltrated to many environmental components (especially an aquatic ecosystem) because of their biological persistence and the ability of accumulation. This diploma thesis is focused on the selection and the optimization of method for the determination of musk compounds in real biotic matrices (fish). The isolation of analytes was realized by PSE method and the purification of extract was realized by the method of the adsorption column chromatography. Identification and quantification of individual musk compounds was carried out by GC-MS. In conclusion were results and the contamination of fish from the river Svratka disscissed and evaluated. These fish were caught at the inflow and at the outflow of the wastewater treatment plant.
Caffeine degradation by high frequency discharge in solutions

The bachelor thesis deals with the degradation of caffeine in solutions by a high frequency discharge generated directly in the solution. The influence of experimental conditions such as the initial pH and conductivity of the solution, caffeine concentration or electrode geometry was studied. The prepared caffeine solutions had a concentration of 10–100 ppm and their initial pH was adjusted with electrolytes (NaCl, Na3PO4, Na2HPO4 and NaH2PO4). All experiments were performed in a batch plasma reactor consisting of a 400 ml beaker and two electrodes. UV-VIS spectrophotometry was used to determine the amount of degraded caffeine. It was found that the experimental conditions (electrical conductivity, power, type of electrode used, electrolyte used) have an effect on the degradation of caffeine. The most suitable electrolyte appears to be NaCl and the initial specific electrical conductivity set in the range of 400–500 S·cm–1.
Modern vegetable oils useful in food and cosmetics

The aim of this Bachelor’s thesis was to make a literary research of currently most used vegetable oils in food technology, cosmetics and pharmaceuticals focusing on health benefits, possibilities of use. Two selected modern vegetable oils were specified that are currently a subject of research in respektive fields. The selected modern oils were carrot seed oil and raspberry seed oil. A literary recherche was drawn up dealing with these oils from the perspective of their composition, charakteristics, properties and use in food technology, cosmetics and pharmaceuticals. In the experimental part of the thesis the fatty characteristics of carrot seed oil and raspberry seed oil were determined: acid value, saponification value, ester number, iodine value, peroxide value. Also fatty acids present in the oils were identified and quantified using gas chromatography with flame ionization detector. Fatty acids were analysed as their methylesters. Derivatization was was performer by alkalic esterification using methanolic solution of potassium hydroxide. In conclusion, selected oils were compared in terms of fatty characteristics and fatty acid profiles. Fatty acid composition was also compared to fatty acid composition found in literature sources.
The Application of Separation Techniques Based on Gas and Liquid Chromatography with Different Types of Detectors for the Determination of Biologically Active Compounds and Selected Xenobiotics

This work deals with the using and application of separation techniques for analysis of polymers degradation and polycyclic aromatic hydrocarbons. Thereby this work is separated to two special parts. In the first part, the degradation properties of synthetic biopolymers based on lactic acid, gylcolic acid and poly(ethyleneglycol) PLGA-PEG-PLGA and ITA-PLGA-PEG-PLGA-ITA (modified by itaconic acid) were studied. These copolymers (firstly their thermosensitive hydrogels) should be used for therapy of fractures in orthopedy (as adhesives). Therefore, the sol-gel and gel-sol phase diagrams were determinated for selected samples of copolymers. The samples forming gel at 37 C was used for other study. Polymer samples were depredated in phosphate buffer at 37°C. The degradation process of physical hydrogels was described by the decrease of molecular weight and the increase of concentration lactic acid and glycolic acid in phosphate buffer. The obtained results confirmed that the degradation of polymer modified by itaconic acid is faster process than no modified polymer and polymers with lower ratio PLGA/PEG degrade also faster than lower ration PLGA/PEG. The influence of pH it was also tested. The rate of degradation of polymers was follow pH 4,0
Quantification of selected pollutants in waste waters

The focus of this thesis is to detect the basic indicators of waste waters released from waste watter treatment plants. The following parameters were discovered: biochemical oxygen demand, chemical oxygen demand, dissolved solids, inorganic nitrogen, total nitrogen, total phosphorus, pH these result led to a conclusion whether the specimens meet the legislative requirements for releasing the treated waste waters into the surface waters.
Determination of organic tin compounds in the environment

Organotin compounds are one of the most produced and most used organometallic compounds. Some of these substances are endocrine disruptors, persistent organic polutants and their high toxic effects are observed. That’s why their presence in the environment caused by human activity could endanger many organisms. The aim of this thesis is summarize their properties and their occurrence in the environment. Then the quick, easy and relatively cheap method for determination of trialkyltin compounds in heavily poluted aquatic sediments using capillary zone electrophoresis is developed.
Determination of organic substances in tar formed after biomass combustion and gasification

Biomass pyrolysis and gasification techniques count among basic technological procedures for its use as a source of energy. As a side-effect, production of tar can be considered. Tar is a complex mixture of various organic compounds and affects negatively both the environment and the facilities where biomass is processed. Within the scope of this master thesis the analysis of tar samples from different materials was performed. As an appropriate analytical method the gas chromatography combined with flame ionization detection (GC-FID) and time-of-flight mass spectrometry (GC-TOF-MS) was chosen. The concentrations of volatile organic compounds known as BTEX, polycyclic aromatic hydrocarbons (PAH) and phenolic compounds were assessed.
Determination of antibiotic residues in soil

This master‘s thesis is focused on the optimization of sample preparation and analytical method for the determination of fluoroquinolone. The theoretical part describes the principles of analysis in soil samples, environmental aspects of antibiotic resistance, characteristics of UPLC-MS / MS methods, description of quinolone. The experimental part deals with the identification and determination of fluoroquinolone substance (ciprofloxacin, enrofloxacin) in soil in which manure from poultry litter was incorporated using liquid chromatography with mass spectrometry (MS / MS, triple quadrupole). Internal standard and standard addition methods were used to quantify ciprofloxacin and enrofloxacin. The amount of CIP in the samples ranged from 0.2030 to 12.7088 µg·g-1 and for ENR from 0.2537 to 10.0224 µg·g-1.
Pesticide glyphosate and possibilities of its determination

This bechelor thesis deals with determination of glyphosate employing capillary isotachophoresis technique. The literature review is devoted to pesticide, their distribution and use, properties and methods for the glyphosate determination, capillary zone isotachophoresis and its use. Experimental part focuses on the application of capillary isotachophoresis for determination of glyphosate, for determination of glyphosate in commercial preparations and testing of glyphosate sorption in soil.
