Vascular and osseous cells in polymer structures for tissue engineering

Artificial vascular and bone prostheses are engineered as bioinert, not allowing cell attachment and growth. Our aim was to prepare materials based on natural and synthetic polymers that could modify the surface or create the bulk material of prostheses, and test their bioactivity in vitro. We prepared fibrin assemblies of various thicknesses and evaluated the adhesion, growth and differentiation of endothelial cells (EC) on these layers. We observed increased cell spreading on twodimensional fibrin assemblies and improved cell growth and maturation on thick fibrin gels. Fibrin coated with collagen I, or fibronectin, increased the adhesion area and the proliferation activity of vascular smooth muscle cells (VSMC). Synthetic polymers were based on an inert block copolymer of poly(DL-lactide) and polyethylene oxide (PDLLA-b-PEO) in which 5% or 20% of the PEO chains were grafted with Gly-Arg-Gly-Asp-Ser-Gly oligopeptide, a ligand for cell adhesion receptors. Grafting oligopeptide peptide to the cell non-adhesive copolymer restored adhesion and growth of VSMC, even in a serum-free medium. Synthetic polymers could therefore serve as artificial extracellular matrix analogues for vascular tissue repair and regeneration. Evaluation of efficacy and bio-distribution of AAV5-miHTT in HD minipig brain

Healthy minipigs (n=29) were pre-screened for serum AAV5 neutralizing antibodies (NABs) in serum by uniQure. Conclusions: Intrastriatal/intrathalamic bilateral application of AAV5-CAG-miHtt(3e13/1e13gctotal) or PBS/Sucrose (54μl/216μl) similarly to AA5-CAG-GFP didn´t cause any neurological deficit in transgenic as well as wild type animals. The body weight increased in all animals 84 days after AAV5-CAG-miHtt/PBS-Sucroseintrastriatal/intrathalamic delivery with two exceptions TGanimal L616 (AAV5-CAG-miHtt1e 13g ctotal) and WTanimal L485 (AAV5-CAG-miHtt3e13gc total). The TG boar N176 from group VII (PBS/Sucrose) died during narcosis 56 days after IP/IT application), shortly after CSF and blood collection.The boar breathed heavily and intermittently, then the heart stopped beat and resuscitation failed. We immediately flushed them by 20 L of PBS with heparin and collect all tissues. All animals had normal food consumption. Inflammatory proces in AAV5-CAG-miHtt (1e13 gc total) seems to be similar to PBS/Sucrose injected animals accompanied with no detectable histological pathologies.
Phenotypic Analyses of the HD transgenic Minipig Model

The transgenic model of Huntington’s disease in minipig (TgHD) was created in 2009 and information coding the sequence of N-terminal part of human mutated huntingtin was transferred to subsequent generations from both female and male sides. In each litter, transgenic (TgHD) and wild-type (WT) piglets were born in approximately equal ratio. At present, the Laboratory of Cell Regeneration and Plasticity keeps sets of animals in F2 and F3 generations with identical genetic background and bred in identical conditions of feeding and housing. The present research project was focused on a complex of non-invasive and invasive approaches to WT and TgHD minipigs to achieve the entire phenotypic analysis of HD progression in this large animal model.

This thesis deals with the role of renewable energy sources in the Czech Republic and stresses the importance of various sources of renewable energy and the possibilities for further development, according to the draft of the updated State Energy Policy. The second part of this thesis focuses on the situation of hydro power plants in the Czech Republic with a closer examination of support for this resource. The thesis documents methods and possibilities of operational and investment subsidies, while simultaneously exploring the possibility of using structural funds of European Union in the construction of hydroelectric power plants. The practical part consists of an elaborated business plan for the construction of specific hydroelectric power. The result of this work is an evaluation of the project and the finding that the given investment opportunity is very attractive for the investor. Timeliness of the thesis is mainly in the fact that the work is based on the new situation that has arisen out of the updated legislation on support for renewable energy sources.

The methodology has been developed as a part of solution to program of applied research and experimental development ALFA Technology Agency of the Czech Republic, subprogram Progressive technologies, materials and systems (TA01011466; 80%) and Operational Program Research and Development for Innovation Ministry of Education, Youth and Sports (project ExAM - CZ.1.05./2.1.00/03.0124; 20%).

Quest for protein biomarkers of neural stem cells differentiation

Understanding neurogenesis and neural cell differentiation presents a unique challenge for treatment of nervous system disorders. To gain more insight about molecular mechanism of differentiation of neural cells, we applied different proteomic approaches using classical 2-DE followed by MS and antibody microarrays. Based on 2-DE, profile of constituent proteins of neural stem cells and their differentiated progenies was estabilished at first and then the protein species that are significantly up or down regulated during the differentiation were selected. Differentiation of neural cells was accompanied by changes in the expression of proteins involved in DNA and RNA binding, mRNA processing and transport, stress responses, iron storage and redox regulation. Immunoblot verified changes of hnRNP A1, hnRNP A2/B1, RafB, heme-oxygenasy 2, GRK2 proteins and alphaB-crystallin (S45), CDK1/2 (Y15), PKC mu (S738+S742), proline-rich Akt substrate (T246) phosphorylations during differentiation.
