Role of PGC-1 coactivators in the regulation of mitochondrial function in adipose tissue

Metabolic pathways in adipose tissue affect the whole-body energy homeostasis. De novo lipogenesis and futile metabolic cycling based on lipolysis and fatty acid re-esterification which is engaged in regulation of fatty acid level in bloodstream are occuring there. These processes are partly regulated by nuclear receptor PPARγ. Mitochondrial biogenesis and oxidative phosphorylation in adipocytes are controlled by interacting of PPARγ with transcriptional coactivators PGC-1α and PGC-1β. The aims of this thesis were to find out whether PGC-1β is connected with regulation of futile cycling and de novo lipogenesis in white adipose tissue and also how specific inactivation of PGC-1β gene in adipose tissue affects phenotype of mice during short-term cold exposure or treatment based on high fat diet enriched by n-3 polyunsaturated fatty acids in combination with mild calorie restriction. The results show that inactivation of PGC-1β probably does not affect futile cycling based on lipolysis and fatty acid re-esterification. In mice with PGC-1β ablation compensation in weight of brown adipose tissue was observed as well as increase in the gene expresion of nuclear receptors PPAR, transcriptional coactivator PGC-1α and UCP1 during cold exposure. Role of mitochonodrial genome in myocardial ischemia-reperfusion injury of spontaneously hypertensive rats (SHR) adapted to hypoxia.

Diplomová práce Abstract - Iveta Brabcová Abstract Ischemia-reperfusion heart injury is one of the most significant diseases affecting mankind and therefore current research pays more attention to its prevention and knowledge of the possible mechanisms which protect the heart. Adaptation to hypoxia has been known for several decades as a cardioprotective intervention but the main issues of protective mechanisms which are induced by the adaptation are still not completely understood. An important role of mitochondria as the main producers of energy and reactive oxygen species which can play a signalizing role in these mechanisms is confirmed in many studies. For this reason a special conplastic strain SHR/OlaIpcv-mtBN/Crl was created. This strain carries the nuclear genome of spontaneously hypertensive rat (SHR) and the mitochondrial genome of normotensive, highly resistant strain Brown Norway (BN). The aim of this study was to compare the expression of selected gene transcripts in the area of energy metabolism, of genes which are related to mitochondrial biogenesis and signaling and antioxidant systems. The mechanism of cell death induction in human malignant melanoma in vitro

Melanoma is a complex malignant disease characterized by increasing incidence and mortality. Melanoma biology is complicated and comprises many aberrant genes and signaling pathways. Treatment of melanoma relies on surgery, the role of radiation therapy and chemotherapy are limited. In advanced disseminating and metastasizing melanoma chemotherapy largely fails and that is why new approaches are being sought including identification of deregulated molecules or processes which could be used as targets. Some of these targets center on frequently aberrant cell death regulating mechanisms. Some traditional cytostatic drugs such as inhibitors of topoisomerases although useful in other types tumors so far proved useless in treatment of melanoma. Nevertheless, expression of topoisomerases in melanoma is often high, thereby enabling at least theoretically their use as targets for chemotherapeutical intervention. Aim of this study was to assess the cytoxicity and proapoptotic effect of campthotecin (topoisomerase I inhibitor), etoposide (topoisomerase IIα inhibitor) and their combination in selected human melanoma cell lines. Green ICT maturity model as a starting point for the implementation of Green ICT practices in small and medium-sized enterprises

The general topic of this thesis is Green ICT and its support. But specifically is this work focused on Green ICT maturity model, as a starting point for the implementation of Green ICT practices in small and medium-sized enterprises in the Czech Republic. The main goal of this thesis is therefore the definition of this maturity model. This definition primarily consists of defining the questions, possible responses and assigning weights for these responses from the perspective of Green ICT maturity. By answering these questions, Green ICT maturity of any small or medium-sized enterprise can be measured. To achieve the main goal, the areas of Green ICT and Green ICT maturity levels for small and medium-sized enterprises were defined. However, for making this maturity model a relevant result of the thesis, testing it in practice was necessary. For this purpose was therefore created an interactive website with self-assessment form, through which companies were able to evaluate their Green ICT. Than the results of this self-assessment, were analyzed. The personal benefit of this thesis is definition of the Green ICT maturity model with a focus on small and medium-sized enterprises, including implementation and verification in practice. This part is considered as practical. For theoretical part I consider defining the areas of Green ICT for small and medium-sized enterprises, which is also seen as personal benefit.

This work should define an information need of a mid-level manager in a small company. An important aspect and the assumption is, that information need relies not only on existing information resources in the company, but also on not yet directly defined topics, such as people management, social behavior, and methods of soft systems methodology of problem solving and team working. So this work isn't only about the information need, but also about topics mentioned above.
