Effect of vitamin D on ABCB1 and CYP3A4 expression in human precision-cut intestinal slices
Mazurová, Tereza ; Vokřál, Ivan (advisor) ; Matoušková, Petra (referee)
Charles University Faculty of Pharmacy in Hradec Králové Department of Pharmacology and Toxicology Student: Tereza Mazurová Supervisor: PharmDr. Ivan Vokřál, Ph.D. Title of diploma thesis: Effect of vitamin D on the expression of ABCB1 and CYP3A4 in human intestinal slices When drugs are administered orally, their absorption is significantly affected by the intestinal barrier. This barrier expresses a variety of efflux and uptake transporters, as well as first and second-phase biotransformation enzymes. The most important efflux transporter in the intestinal barrier is P-glycoprotein, which has a broad substrate specificity. Among the first-phase biotransformation enzymes, cytochrome P450 3A4 is the most important. Their function is to protect the human body from the toxic effects of xenobiotics, including drugs. Many clinically important drugs act as substrates, inhibitors or inducers in relation to these proteins, which may result in an increased risk of drug interactions. A plethora of dietary supplements or medicines containing vitamin D can be found in the pharmacy. It is used for the proper development of bones and teeth, for the proper function of the immune system or for the treatment of osteomalacia, rickets or osteoporosis. Limited information is available on its effects on P-gp and...
The optimization of fenbendazole extraction from environmental samples
Brodská, Denisa ; Matoušková, Petra (advisor) ; Raisová Stuchlíková, Lucie (referee)
Charles University Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Denisa Brodská Supervisor: doc. Ing. Petra Matoušková, Ph.D. Consultant: Mgr. Martina Navrátilová Title of bachelor thesis: The optimization of fenbendazole extraction from environmental samples Anthelmintics are drugs used for the treatment and prophylaxis of infectious diseases caused by parasitic worms. In the Czech Republic, they are mainly used in veterinary medicine. During the biotransformation of the active substance, metabolites are formed, which are excreted together with the parent substance in faeces and urine. Fenbendazole and other similar substances can be absorbed into the soil in this way and subsequently act on non-target organisms in the environment. Fenbendazole is widely used in livestock farming, and since animal excrements are often further used for fertilization, the risk of the content of medicinal substances should be strictly monitored. For this purpose, it was necessary to develop and optimize the extraction method and also to carry out the corresponding validation of the analytical method, which will subsequently be applicable to real samples in the environment. The assumption of absorption of fenbendazole and its metabolic products, fenbendazole sulfoxide and...

Bezdíčková, Kristýna ; Matoušková, Petra (advisor) ; Nguyen, Thuy Linh (referee)
Charles University Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Bc. Kristýna Bezdíčková Supervisor: doc. Ing. Petra Matoušková, Ph.D. Title of diploma thesis: Implementation of the diagnosis of gastrointestinal nematodes using RT-PCR Gastrointestinal nematodes can cause various diseases in farm animals. Individuals are often infected with several types of parasites at once. Since the symptoms tend to be similar, the disease is called parasitic gastroenteritis. Not only do nematodes threaten the animal health, but they can also have a major impact on the economy of the entire farm. Diagnosis is not always easy. In addition to morphological and coprological methods, biochemical or molecular analysis can be used, which tends to be more sensitive and accurate. We tried to diagnose gastrointestinal nematodes using the molecular RT-PCR method. In the thesis we focused on 6 species of nematodes (Haemonchus contortus, Teladorsagia circumcinta, Trichostrongylus spp., Nematodirus battus, Chabertia ovina and Ashworthius sidemi). The material used was feaces samples from sheep from which we isolated gDNA. The control material was gDNA isolated from H. contortus eggs and gDNA obtained from whole parasite bodies. RT-PCR was tested using the fluorescent dye SYBR Green I and...
New fungal fucosidases and their synthetic potential
Glozlová, Michaela ; Beránek, Martin (advisor) ; Matoušková, Petra (referee)
Charles University Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Bc. Michaela Glozlová Supervisor: prof. PharmDr. Martin Beránek, Ph.D. Consultants: doc. RNDr. Pavla Bojarová, Ph.D., Mgr. Pavlína Nekvasilová Title of diploma thesis: New fungal fucosidases and their synthetic potential The literary research focuses on α-L-fucosidases, their occurrence, functions, and use in the synthesis of fucosylated compounds, which were also a part of the research, particularly their presence in the human body and involvement in pathological conditions. Furthermore, it explores the oligosaccharides found in human milk, their impact on infant health, and their synthesis, which can be facilitated through the use of α-L-fucosidases. The experimental section involves the analysis of a library of filamentous fungi with the aim of identifying a representative strain with α-L-fucosidase activity towards pNP-α-L-fucoside. Recombinant α-L-fucosidases were selected based on searches in genetic databases and prepared. The feasibility of their production in Pichia pastoris and Escherichia coli was tested, and the produced enzymes were purified by ion exchange chromatography, characterized and their regioselectivity and synthetic potential for the synthesis of fucosylated...
The efflux transport activity of P-glycoprotein - 9.2 fromHaemonchus contortus
Kohoutová, Kateřina ; Matoušková, Petra (advisor) ; Boušová, Iva (referee)
Charles University Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Kateřina Kohoutová Supervisor: doc. Ing. Petra Matoušková, Ph.D. Title of diploma thesis: The efflux transport activity of P-glycoprotein-9.2 from Haemonchus contortus Haemonchosis is a globally widespread disease affecting small ruminants. The causative agent of the disease is the gastrointestinal nematode Haemonchus contortus, which feeds of the blood of its host. The treatment of haemonchosis with anthelmintics is complicated by the worldwide spread of resistance. One possible cause of the development of anthelmintic resistance is a decrease in anthelmintic concentration in the worm due to increased expression of P-glycoproteins from ABC transporter superfamily. The study of the transport activity of these transporters could help to understand the mechanism of resistance development and make the treatment of haemonchosis more effective. The aim of this thesis was to characterize the transport activity of P-glycoprotein-9.2 and its interaction with a selected inhibitor using a heterologous expression system. The adherent cell line HEK293 was transfected with a plasmid carrying the Pgp-9.2 gene of the Haemonchus contortus. Successful expression was verified by dot blot. Unfortunately,...

Training of Employees in the Banking Sector
MATOUŠKOVÁ, Petra
The bachelor thesis deals with education in the banking sector of the Czech Republic. It defines the indi-vidual concepts related to education, methods of education and also includes the characte-ristics of the banking sector of the Czech Republic. In the theoretical part, corporate education, its individual phases and their sub-parts are detailed. This part of the thesis also deals with individual methods of education, which are then described in more detail, including their main advantages and disadvantages. The second half of the bachelor thesis is the research part. This was conducted during 2023 and the results were then compared with the CVTS questionnaire survey. This survey is carried out by Eurostat with a regular five-year periodicity in the countries of the European Union, which includes the Czech Republic. It has been participating in this survey since 2000. The most up-to-date data are from 2015, as the Covid-19 pandemic is set to hit the world in 2020. Inspired by the CVTS survey, a questionnaire of its own was created, looking at the methods currently used in training, the forms of training or what further courses the employees themselves would like to take and where they feel there is room for improvement. The questionnaire also included questions on the fi-nancing of training, which is a very important item in banks' budgets and an investment in their future. Finally, further training progress was recommended based on the needs of banking sector employees and a future outlook was given in which the whole current training system could be rapidly transformed by aug-mented or virtual reality.

"Being close to another person, being with them, that's the most important thing - in love, in life and in dying." Being close to another person is a basic and easily attainable need for a healthy person. Spending time with either our family relatives or friends is actually quite simple for the majority of us. Foremostly, we just put our work aside and sit on a bus, train, or plane and after a certain period of time we can be together with our closest loved ones. However, for the families with disabled children it is not that easy. If the family finds itself in a difficult life situation, often the simple “being around” might be challenging. For this particular reason, my thesis proposal aims to create a safe place for all of those who could possibly end up in a such situation – children hospital where a person can be available for a help any time needed.

The main aim of the bachelor thesis is to find a use for the Piarist monastery in Lipník nad Bečvou, which is currently abandoned and does not fulfill any function. Based on the analysis, we decided to design a building for the Faculty of Pharmacy that would combine space for education and science. At the same time, we try to preserve the original character of the building, while all incorporated elements should underline its historical beauty and functional potential. We open up the original space with several cramped rooms and we design laboratories, classrooms, auditoriums or garden greenhouses and fields in the exterior, which are used for educational and research activities of the faculty. Besides that, it includes spaces for ordinary people of Lipník nad Bečvou, such as a teahouse, library, pharmacy or exhibition spaces so they have the opportunity to use the local services in a pleasant environment and learn information about the field of pharmacy or the cultivation of medicinal plants. The arrival of young, educated people to the city is also a significant benefit of the monastery's reconstruction. Two open atriums that are surrounded by the building are parts of the monastery. Thanks to them, a lot of daylight enters the building. They connect the outdoor space in front of the monastery with the garden at the back. The new proposed entrance is therefore made up of glass so you can see the atrium and the garden when you enter the building. The important point in the central part of the monastery is a large, open and airy space with a staircase that connects the various parts of the building. It provides a meeting place for students, teachers and other members of the university who can spend time together, exchange knowledge, and learn.

The bachelor thesis deals with the reconstruction of the building of the Piarist monastery in Lipník nad Bečvou. The building has been unused for more than 20 years and it is difficult to find a suitable use for it, although it has great potential due to its size and the corresponding extensive garden. The proposal is aimed at revitalizing the building by inserting a multifunctional hall, opening the monastery garden to the public and other interventions. In addition to restoring the monastery, the goal was to create a space for people that would support and enrich their cultural and social life.
