Hif1a role in cardiovascular function and heart disease

Hypoxia inducible factor 1 alpha (HIF1A) is a transcriptional factor, which plays a central role in the maintenance of homeostasis under hypoxic conditions. It regulates a wide variety of genes encoding proteins that influence metabolism, extracellular matrix composition, oxidoreductase activity or angiogenesis in response to reduced oxygen levels. When HIF1A protein function is impaired, the organism is unable respond appropriately to hypoxia. Altered HIF1A regulation can result in severe tissue damage and eventually lead to death. The heart, as an organ with a huge oxygen consumption, is susceptible to various pathologies caused by hypoxic stress. The role of HIF1A in the heart is rather ambiguous and remains to be elucidated. It plays a role in cardioprotective mechanisms but also promotes the development of inflammation and apoptosis. This thesis aims to clarify the role of HIF1A in maintaining physiological functions of the heart during adaptation to hypoxic conditions using a mouse model with heterozygous Hif1a+/- deletion. The Private and the Public in the Work of Libuše Jarcovjáková

The bachelor thesis is dedicated to the interpretation of the work of the Czech female photographer Libuše Jarcovjáková and aims to emphasize its importance and uniqueness for Czech and world photography. The theoretical part introduces the author, her cultural background, consisting of important personalities of the Czechoslovak art scene, and her still- evolving work and projects. It offers an insight into the world of photography through the search for similarities and differences with the work of Nan Goldin, with whom she is often compared in the Czech and foreign context. It focuses mainly on the work from the 1970s and 1980s, presented in the author's book Černé roky (The Black Years), its setting in the contemporary context, and the interpretation of selected photographs in terms of form and content. The interpretation focuses mainly on the search for the boundaries of the private and public, the issue of censorship and self-censorship, and the projection of these influences into the author's photographs. The didactic part of the thesis is a pedagogical project combining the knowledge and experience from the theoretical and practical parts of the work. Transcriptional regulation in the development of the cardiac sympathetic system

To improve modern therapeutic and diagnostic methods, it is crucial to understand the development of the cardiac sympathetic system and to identify the genes involved in its regulation. Neural crest cells give rise to the sympathetic precursors that migrate towards the dorsal aorta. This migration is regulated by the NRP1/SEMA3A and neuregulin/ERBB signaling. The differentiation towards the sympathetic phenotype is regulated by transcriptional factor networks, including ASCL1, PHOX2A/B, GATA3, HAND2, HIF1A and ISL1. Next, neurons migrate to the final paravertebral position, which is regulated by the BDNF/TRKB signaling. The final step in the development of cardiac sympathetic neurons is the axon growth and guidance towards the heart. This is regulated by the NGF/TRKA and NRP1/SEMA3A signaling. This thesis aims to map current knowledge of different regulation pathways involved in the cardiac sympathetic development (especially in the mouse model) with emphasis on transcriptional factors. This type of information should help us better understand the pathophysiology of some cardiovascular diseases associated with the dysfunctional sympathetic system, such as arrhythmias, congestive heart failure or myocardial infarction, which remain to be main causes of death worldwide.
Possibilities of a nature reserve V Lisovech in the development of tourist trade in SO ORP Jihlava and Pelhřimov.

This bachelor thesis deals with the analysis and evaluation of meaning of the nature reserve in Lisova. It is also focused on the possibility of utilization of a V Lisovech nature reserve in the development of tourist trade in SO ORP Jihlava and Pelhřimov. The main aim of this thesis is to develop drafts and materials which should help to the development and propagation of the tourist trade in the observed region. Results gained from the propagational materials are made into logo, web application and information booklet. The processing of results, information booklet and logo was created thanks to the programs as Adobe InDesign CS6, ArcGIS ArcMap and Zoner Photo studio 17. On the other hand the web application was made due to the ability to link maps with GoogleMaps. An essential part of this thesis is also my own maps, photographs and charts.

The thesis describes and explains the issues relating to non-profit organizations, in particular governmental organizations. The theoretical part enlarged on ways to finance non-profit organizations, and customer relationship management. Pursuant to available information are compiled an internal and external analysis, an extensive marketing mix analysis and describes the results of marketing research and proposed recommendations to increase competitive.
The formation of biogenic amines in flesh of selected fish species

The thesis deals with the use and effectiveness of some less common methods of conservation of fish meat. The formation of biogenic amines in meat is observed in connection with the non-traditional preservative methods. Amines can serve as indicators of protein degradation. The quality of fish was considered in connection with the increasing content of selected biogenic amines (putrescine, cadaverine, spermidine, spermine, 2-fenylathylamine, histamine, tyramine and tryptamine). Ultra performance liquid chromatography (UPLC) was used as the method for determination of biogenic amines. Amines were derivatized by dansylchloride before their UPLC separation. The fish flesh was vacuum-packed. Samples were stored for several weeks in a thermostat at the selected storage temperature after the application of selected preservative technique. Beta-irradiation and high hydrostatic pressure were used for the preservation of fish flesh. Control samples were not exposed to the â-irradiation and high pressure. The organoleptic properties were studied for all samples (smell/odor, insight and shape). Beta-irradiation was applied to fish meat of common carp (Cyprinus carpio) and rainbow trout (Oncorhynchus mykiss). Both these species of freshwater fish are economically significant. Carp and trout are the species being mostly consumed in the Czech Republic. Fish meat is considered to be provided the flesh is fresh. A testing series of samples was created with common carp to determine the appropriate dose of â-irradiation. The maximum permissible dose of irradiation for fish meat is 3 kGy. Fish samples were exposed this dose in the first experiment. The dose of irradiation was reduced in following experiments based on the experience from the initial experiment. The doses of 0.25, 0.5, 0.75, 1.0 and 2.0 kGy were applied to rainbow trout. The value of 0.75 kGy of â-irradiation or higher (1.0, 2.0 and 3.0 kGy) prolonged the shelf life of fish meat, which was stored for three months (98 days). Prolonging of the shelf life of fish meat to approximately 98 days at 3.5 °C is redundant from technical point of view. For that reason lower doses 0.25, 0.5 and 0.75 kGy were tested in more detail in the repeated experiment with carp meat. Lower doses of â-irradiation are considered to be more acceptable and-at the same time-sufficiently effective for delaying the beginning of degradation processes. 6 High hydrostatic pressure was applied to meat of common carp (Cyprinus carpio), rainbow trout (Oncorhynchus mykiss) and pike (Exos lucius). Pike is another very popular kind of freshwater fish. Pike flesh is very tasty, but in spite of this, pike is not so much popular among consumers compared to carp and trout. The cause is its high price. Samples of pike were stored at standard temperature 3.5 °C and also at higher temperature 12 °C (unlike experiments with â-irradiation). Lower temperature of storage (3.5 °C) followed the conditions of storing of fish meat in industrial refrigeration facilities and households. The high pressure might not be sufficient for preservation at higher storage temperatures. This assumption was based on available information. Samples were treated by high pressure and stored at both 3.5 °C and 12 °C to verify this assumption. Higher temperature simulated either failure of refrigeration equipment or unsuitable store temperature of meat. In all species selected freshwater fish two levels of high pressure were applied ? 300 and 500 MPa. Both levels had significantly reduced the formation of biogenic amines, especially in samples stored at 3.5 °C. At this temperature, the effect of 300 and 500 MPa delayed start of degradation processes in fish meat by 3?4 weeks. At 12 °C and 500 MPa, high pressure extended the sustainability of meat by no more than one week. 500 MPa is effective treatment at the lower temperature of 3.5 °C. High pressure is not reliable preservative techniques at higher temperature.
Modernity in Controversy. Zikmund Winters´ work in the Journal Zvon (1900-1912)

This piece of work will be talking about famous works (such as Peklo, Mistr Kam- panus) and margine contributions of Zikmund Winter. These contributions were published in 1900 ? 1912 in magazine called Zvon. These texts will be considered especially from the poit of view of the contributors of magazine Zvon and Moderní revue. These contributors had many polemics between each other. Moderní revue was the centre of decadent and symbolist orientation. My aim is to describe the literaly method of Zikmund Winter that can be considered sometimes as opposed, sometimes as identical with programme of Česká moderna. The solution will be not only the interpretation of chosen works but also the analysis of comments, editorial notes and reviews concerned these works.
