Identifikace genů vázaných na pohlavní chromozomy u Silene latifolia a Silene dioica

Silene latifolia and Silene dioica are closely related dioecious plant species where sex is determined by the presence of sex chromosomes, X and Y. The sex chromosomes of S. latifolia and S. dioica are younger (10-20 million years) than in mammals (about 200 million years). These two species are thus important models for the study of early stages of sex chromosomes evolution. In this work a group of genes linked to sex chromosomes (Sl3, Sl7, DD44) was analyzed. These genes were de-novo isolated from S. dioica by screening of the BAC library. The expression of these genes in different tissues was compared using Real-Time PCR and HRM in S. latifolia and S. dioica. Using the low-copy FISH (fluorescence in situ hybridization) method, a physical map for individual genes was created. This map is the most detailed physical map created for closely related Silene species.

The master thesis titled "Self-immolation of Jan Palach - from reality, newspaper/TV news to film cover and film reviews of Burning Bush" concerns of film cover of real events in three parts drama called Burning Bush, which is the main topic of the movie. The thesis aims to ascertain to what extent it is the movie with a documentary elements, where authors would have to represent only real events and to what extent it is artistic movie where authors have more space in creativity and could change reality to impress spectators with theatricality and emocionality, with the risk they will change the meaning of real events, therefore they would manipulate with the spectator. The thesis also essays the media content before the 1989 and after the 1989, while it aims to find out not only what is the difference in the newspaper and TV news, but also what difference is in the quantity of the content regarding to Jan Palach in the times of totality and democracy.
The Impact of scientific knowledge on Aesthetic Appreciation of Nature

The bachelor thesis is focusing on the question of the impact of the scientific knowledge on aesthetic appreciation of nature. The question is if scientific knowledge does in fact have any impact on the aesthetic appreciation of nature, and if so, how does it influence our aesthetic appreciation. The thesis is solving the question with the aid of selected authors, particularly Ronald Hepburn, Allen Carlson, Nick Zangwill and Cheryl Foster. The initial item of the thesis is to show the cleft between two attitudes, which are cognitive and formal attitudes. The thesis demonstrates how these two attitudes tackle the question of the impact of scientific knowledge on aesthetic appreciation of nature and in the final chapter it offers a new attitude, which interconnects two previous attitudes. Detekce mikrobiální kontaminace molekulárně-biologickými metodami v kořeninové paprice

Dryed pepper is one of the most used spice in many parts of the world. The presence of undesirable pathogens can be a serious problem for consumers. Especially contamination by microscopic fungi, some of which produce mycotoxins representatives, poses a great threat to humans. Today's conventional method for determining contamination by microorganisms are time consuming and some less common types of pathogens can be difficult to determine. In the context of this thesis were applied molecular biology methods for detection of contamination in samples of pepper. There were selected ITS regions for sequencing, which show high variability between species and organisms are an appropriate tool for the taxonomy. The sequence analysis of the ITS regions were identified by sequence high similarity with the ITS regions of many microscopic fungi especially representatives of the class of Ascomycotina and several yeast species. The work was a qualitative rather than a quantitative detection, which does not exclude the wholesomeness of the samples, which is proven by health protocols.
