Optimization of the Coil Forming Workplace

This bachelor’s thesis deals with the analysis of the production process and its optimization. Improvement is achieved thanks to a predictive simulation. The theoretical part of the thesis describes the concepts associated with the process management of the company and measurement of work. Furthermore, the theoretical part focuses on the improvement of business processes within the process of digitalization and methodology of simulation projects associated with the creation of a predictive digital twin. The analytical part contains an introduction of the manufacturing company and a global analysis of processes. Then, with the help of several techniques, the production process which will be optimized is analyzed. The design part of the thesis describes the current simulation models and the proposed form of the process. Individual options are evaluated, and the benefits of the proposed solution are stated.
Paint shop Process Improvement through Predictive Simulation

This thesis focuses on the use of simulation to optimise processes in a paint shop. The aim of the thesis is to analyse the existing production processes and suggest improvements using simulation. In the analytical part of the thesis, the processes in the paint shop are described in detail and Petri nets are used to describe the logic of the processes in the paint shop. In the design part of the thesis, the problem and objectives are formulated, and the design of the simulation model is presented. The design describes the elements and methods used in the Plant Simulation software in which the model is created. Furthermore, different scenarios are simulated to optimize the painting process and the deposition areas. The simulation results are compared with the actual state of the paint shop. Finally, the results are discussed and recommendations are proposed to optimize the paint shop processes using simulation. This work presents new approaches to process optimization in industrial manufacturing.
Evolution of the gene regulatory network underlying the formation of the gastrula organizer

During gastrulation, the vertebrate embryo is organized from the clump of cells into a bilaterally symmetric body. This organization process is driven by the gastrula organizer. Its establishment is induced by maternal Wnt/β-catenin signaling and Nodal/Activin signaling localized in the presumptive dorsal region of the embryo. The regulative environment then triggers the expression of the organizer-specific genes which create morphogen gradients in the embryonic body and therefore give each cell positional information. However, the evolution of vertebrate organizer establishment remains vague. Here we aim to test the compatibility of the invertebrate cis-regulatory modules with the vertebrate gene regulatory network (GRN). We introduced fluorescent reporter genes under the control of the invertebrate regulatory sequence of organizer-specific genes into a vertebrate model to observe their behavior in the context of the vertebrate GRN. We found and functionally verified a 500 bp-long amphioxus sequence (an enhancer) that is necessary and sufficient to drive a correct Chordin gene expression in the gastrula organizer in zebrafish. Dlx genes in odontogenesis and craniofacial mophogenesis in bichirs

Bichirs, Polypteriformes, are a basal group of vertebrates with many unique characters as ganoid scales, fleshy pectoral fin, spiraculum or paired lung originated from ventral outpocketing from floor of pharynx. Among the most notable is also the dentition, which contatin teeth among whole area of oropharyngeal cavity. This arrangement is not easily call oral and pharyngeal, what is recently used e.g. for medaka, because it is only one dention with teeth on majority of oral as well as pharyngeal elements. This diploma thesis utilizes expression patterns of Dlx genes to visualize morphogenesis of dentition and pharyngeal arches in the Senegal Bichir (Polypterus senegalus). The very first developing teeth of oral dentition, as well as its further patterning were detected and accurately located using gene expression analyses. It was also discovered an interesting histological context connected with unique mouth opening mechanism during early development of teeth in oral cavity. Gene mutations affecting the development of human testes

Development of mammalian sex organs is remarkable process. Both ovaries and testes rise from the same precursor but then differentiate into morphologically and functionally different structure. The determining factors that decide the fate of undifferentiated structures are not just the genes themselves but also the timing of transcription regulatory genes and specific amount of their products. Development of male and female gonads manages a large set of regulatory genes that interact with each other. Together it forms the gene regulatory network. Crucial role in male sex differentiation plays an SRY gene which regulates the time and quantity of the other factors expression. Even slight errors (mutations) in genes sequences of regulatory genes could change their expression. This leads to disruption of the development of sex organs or even death of the whole organism. Naturally occurring mutations and their phenotype in humans are suitable for studies of gene sex regulatory factors. Thanks to these studies we are able to detect the cause of many birth defects what is a first step toward their eventual treatment.
