Kučera, Matěj ; Zumr, David (referee) ; Brukner, Bohuslav (advisor)
The subject of the bachelor thesis is to develop a project of a guest house with almost zero energy consumption at the level of documentation for the execution of the construction. The building is situated on the outskirts of Vrbno pod Pradědem. The guesthouse is designed as a brick building with two above-ground floors with partial basement. It has twelve rooms with a total capacity of 24 accommodated persons. Part of the building is a restaurant with its own entrance and facilities. The structural system of the underground floor is combined from monolithic reinforced concrete. The above-ground floors are designed as a longitudinal wall system with load-bearing walls made of aerated concrete blocks. The ceiling of the individual floors is designed as a one-way reinforced concrete ceiling slab. The ceiling of some rooms consists of a visible truss of a sloping pitched roof with metal roofing in a small slope. The corridor necks are topped by a flat extensive vegetation roof. The building has a designed collective garage within the ground plan of the first underground floor, including ski storage and guest house facilities. On the first floor there are 6 residential units, reception, kitchen, restaurant and its facilities. On the second floor we find the remaining 6 residential units.
Fitness center design with minimal environmental impact
Kučera, Matěj ; Kolářová, Eva (referee) ; Kolář, Radim (advisor)
The diploma thesis deals with the design and project documentation of a fitness center. The designed building is situated in the centre of Brno in a busy environment with a direct connection to the city ring road. It is a two-storey non-basement building with a flat green roof. The building is based on a combination of plain concrete strip foundations, reinforced concrete slabs and pads. The load bearing, external and partition walls are designed from the aerated concrete blocks. The outer skin consists of thermal insulation, slats and vertical garden flowerpots. The floor structure is predominantly designed from pre-stressed concrete panels. Facade is ventilated. The building is divided into two floors where each has its own purpose and specification. In the 1st floor there is a gym, room for trainers, dressing rooms and utility room. In upper tract there are exercise rooms and cardio zone. The building is designed as wheelchair accessible. Around the building there is designed a car park for 24 cars with grass paving and a medium-sized trees. The second part of the thesis environmentally deals with the building services. The building will be heated by four heat pumps and will also use energy from photovoltaic panels. Air exchange and cooling will be solved using air conditioning. Third part of the thesis deals with the evaluation by certificate of energy performance of the building comparing the used heating method with the use of fossil fuels. The thesis is mainly elaborated in Autocad, SketchUP, Deksoft, Lumion and Microsoft office software.
Business management of a capital company
Kučera, Matěj ; Tomášek, Petr (advisor) ; Čech, Petr (referee)
1 Abstract Business management of a capital company This thesis deals with the business management of a capital company. This thesis summarizes applicable legislation and conclusions of legal theory. Conclusions of legal theory are calssified in the structure of the thesis and provide complex description of the business management of a capital company. This thesis summarizes court decisions and classifies them in the structure of the thesis. The thesis summarizes and describe important conclusions of legal theory and court decisions, deals with their evolution and identifies and provides interpretation for contradictions. The first chapter deals with theoretical definition of the business management, position of the business management in the structure of a capital company, strategic decisions, common decisions, decisions in relation to a capital company and differentiation of those decisions from the business management. Further the thesis in this chapter deals with definitions of the business management provided by courts and with specific decisions and actions assessed as the business management by court decisions. Of these decisions, most space is devoted to financing of a capital company. The second chapter deals with ability to give instructions to the business management and issues related to who has...
Comparative study of trademark law in legal systems of the Czech Republic and the Peoples Republic of China
Kučera, Matěj ; Růžička, Michal (advisor) ; Pítra, Vladimír (referee)
This paper compares legal systems of trademark law in Czech Republic and Peoples Republic of China with focus on trademark legislation. The goal of this thesis is to provide the reader with a view of Czech and Chinese mentality towards trademarks as it evolved in time. This historical comparison of trademark evolution should mainly serve as a guideline to understanding as to why each legal system chose such legislation even though both are part of the civil law system. Another goal is to present the reader to a complete view of Chinese trademark legislation with focus on those aspects that are the most different for Czech trademark legislation and also to present specifics of Chinese and Czech trademark legislation which would be hard to find in its counterpart. This thesis is divided into three chapters. The first chapter describes the basic concepts that are essential to this thesis, with focus on those, that are significant for the comparison of both legislations. Second chapter provides complete explanation of historical evolution of trademarks in both countries, with focus on historical evolution of trademarks in China. Third and longest chapter deals with the actual comparison of both legislations, with the main focus on Chinese legislation. Each subchapter starts with a description of a...

Technical solutions of payments for wireless network access
Kučera, Matěj ; Kosek, Jiří (advisor) ; Kalina, Tomáš (referee)
Topic of this bachelor thesis is paid WiFi networks and their technical payment solution. The first chapter deals with the various payment methods that can be used for connection to a paid WiFi network. There are described advantages and disadvantages of different payment methods, problems in their implementation. The second chapter describes several ready-made solution that can be purchased as a complex solution for paid WiFi network. There are compared their features, functionality and price. In the third part, which is the practical part, there is described design and implementation of the payment procedure in Hotel WiFi network, operated by the Network Department of the University of Economics, Prague, at all accommodation and recreational facilities of this organization, in order to give the reader a complex demonstration of how the implementation could look like. This part describes the situation before creating the Hotel network, the basic requirements for this network, its design and the final solution from a technical perspective and from a perspective of a paying customer.
Influence of dietary components and redox enzymes on intestinal microbiota proliferation in the tick \kur{Ixodes ricinus}
KUČERA, Matěj
In this work, we have analysed the temporal dynamics of gut-dwelling bacteria and Borrelia in the gut of the deer tick Ixodes ricinus. Using quantitative PCR, we have shown that levels of the tick intestinal microflora are profoundly decreased at later stages of feeding on whole blood but not on serum. Even though we noted that host complement system manages to interfere with Borrelia viability in vitro, we did not see any effect of host complement on Borrelia acquisition in adult ticks in vivo. However, we revealed that host hemoglobin is essential for Borrelia proliferation in the tick gut. All together, these data imply that, during feeding, levels of gut-dwelling bacteria and Borrelia are determined by the host. While hemoglobin seems to be detrimental for gut-dwelling bacteria, Borrelia require it in order to proliferate. During off-host stage, we showed that levels of gut-dwelling microflora are regulated by an intestinal transmembrane enzyme Dual oxidase. In conclusion, we aimed, and mostly succeeded, to perform pilot experiments describing the biology of a complex process of regulating gut microflora in the vector Ixodes ricinus and extend it by its impact on Borrelia acquisition
Molecular characterization of NADPH oxidase in the gut of the tick \kur{Ixodes ricinus}
KUČERA, Matěj
This thesis focuses on characterization of one member of NADPH oxidases - dual oxidase (DUOX) which has been described as the main factor of epithelial immunity in the gut of model organism Drosophila melanogaster. We have identified an orthologous gene coding for DUOX in the tick I. ricinus and described its tissue expression profile. The DUOX is mainly expressed in the gut of unfed ticks and seems to be downregulated upon artificial microbial infection. A fragment of tick DUOX was prepared as recombinant protein and used for preparation of specific antibodies to be used for further characterization of the enzyme. Our main aim is to highlight the importance of tick DUOX producing the reactive oxygen species and their role in the defence against pathogenic organisms within the tick gut.
