THE ANALYSIS OF MARKETING STRATEGY OF A HOTEL
KUČERA, Lukáš
The thesis is related to the sector of services in the hotel industry. One of partial aims of this pthesis was a thorough analysis of the historical advancement till the present days. The marketing and management methods of SWOT analysis, STEP analysis and a questionnaire have been used to elaborate the work. In addition, various marketing strategies which could be put to use for the hotel in question have been evaluated. However, the main target of the work was a draft of such a marketing strategy that will appear the most advanced in the future.
Accurate pedometers function using the autocalibration
Kučera, Lukáš ; Slabý, Kryštof (advisor) ; Procházka, Michal (referee)
v angličtině Aim: The aim of this study is to explore the possibilities of accurate pedometer functions through a partial removal of individual measurement errors for a particular pair of pedometer- proband, using the auto-calibration of walking for the previously known distance. Methods: They will be used pedometers of mechanical type YAMAX SW-200 and fully electronic type OMRON HJ-720. Thirty adults from 18 to 69 years old (11 men, 19 women) took part in this study. In the pilot phase will be suggesting of a sufficiently long sections of known length (100 m, 300 m a 1000 m) and auto-calibration on a flat track. Own attempt will consist in landscaping measurement. Resulsts: From everage lengths and from the number of steps, which we measured, is possible to determine distance, which probands covered. The biggest aproach was to real distance 1000 m in case we used everage length of step from distance 300 m. In this case undervaluation of the distance reached in Omron 7 % and with Yamax just 10 %. Howewer the distance as well for Omron was overvalued in 27 % probands and for Yamax it was in 20 % probands. Conclusion: The best possibility seems to be average length of step, which is couted from distance for 100 m. Although there is undervaluation more frequent, overvalution is minimal. It will be...
Preparation of affinity surfaces for mass spectrometry
Kalaninová, Zuzana ; Pompach, Petr (advisor) ; Kučera, Lukáš (referee)
One of the most potent toxic substances of biological origin known are botulinum toxins - nerve toxins produced by strains of Clostridium bacteria. They affect neuro-muscular connections where they cleave proteins of the SNARE complex, block neurotransmitter release, and thus stop signal transmission. This leads to flaccid paralysis, the main symptom of a disease called botulism. Due to their extremely low lethal dose and high toxicity, BoNTs take a place on the list of agents of bioterrorism. Despite numerous methods available to this date, the mouse bioassay remains the gold standard for the detection of BoNTs. Regardless of the ethical aspects of these experiments, the time they require is also inconvenient. In this work, a unique combination of protein surfaces and detection of active BoNTs through their endopeptidase activity and mass spectrometry, EndoPep-MS, is introduced. Functionalized surfaces were prepared by ambient ion soft landing - a technique utilizing a beam of hyperthermal ions (energy range 1-100 eV) as a tool for controlled modification. Two approaches to the detection of botulinum toxin A1 were applied. One of them, based on avidin- biotin interaction, used a neutravidin-modified surface for enhancement of biotinylated peptides (substrate peptide for BoNT/A and peptide products...
Design of Supplier-Customer Relationships in Terms of FinancialTies
Kučera, Lukáš ; Sedláčková, Magda Piknová (referee) ; Jurová, Marie (advisor)
The bachelor’s thesis deals with an evaluation of the situation of individual entities from a selected company on the basis of information and documentation provided by the company. The thesis consists of theoretical part and practical part, of which theoretical part consists of individual definitions which are related to the thesis, it deals in more detail with claims which then helps realization of the analysis in the practical part. The practical part focuses on the analysis of supplier-customer relationships within individual entities of the selected company. The invoices issued in 2020 within the entities provided by the company were subjected to the analysis. The results of the analysis were then evaluated, commented and on the basis of the results are written recommendations for improvements and innovations in the company.
Residue analysis from archaeological ceramics using specific antibodies and mass spectrometry
Pavelka, Jaroslav ; Kučera, Lukáš (konzultant)
Important information preserved in the form of biological markers - so-called organic residues containing food residues - can be preserved on ceramics under special conditions. By analyzing them, it is possible to determine the type of food being prepared or consumed in the past. For this reason, a laboratory procedure based on the ELISA method (Enzyme-Linked Immuno Sorbent Assay) was developed, primarily aimed at determining the presence of denatured proteins from medieval ceramics, which uses available and certified kits for immunological analysis. The approved methodology is intended for scientific experts from the ranks of archaeologists, bioarchaeologists and immunologists working with archaeological material.

Technology of a selected part of the vacuum pump
Lišková, Tereza ; Kučera, Lukáš (referee) ; Chladil, Josef (advisor)
The content of study is related to reducing the production times of milling of aluminum component of vacuum pump. The primary aim of the thesis is to experimentally verify the proposed cutting conditions when milling the rotor blades and then evaluate them according to predetermined parameters. The output is reducing machining time with respect to cost, tools and machine.
Evaluation of dynamic postural stability of soccer players
Kučera, Lukáš ; Vomáčková, Helena (advisor) ; Malá, Jitka (referee)
Title: Evaluation of dynamic postural stability in football players Objectives: The main objective of this diploma thesis is to investigate the difference of dynamic postural stability between football players and the common population and measure all the data by computerized dynamic posturography SMART EquiTest System from company of Neurocom. Methods: This is a quantitative cross-sectional study. The research is divided in two groups, an experimental group involving 25 football players aged between 20 and 30 years without injuries, an active career over 5 years and a control group involving 25 participants, who are recreational athletes aged between 18 and 31 years from the Faculty of Physical Education and Sport, Charles University. Measurements of dynamic postural stability were performed on Neurocom SMART EquiTest in the Laboratory of applied kinesiology of Charles University Faculty of Physical Education and Sport. Everyone was tested just one time for all seven test batteries - Sensory Organization Test, Motor Control Test, Rhythmic Weight Shift, Unilateral Stance, Weight Bearing Squat, Adaptation Test and Limits of Stability. One testing session took about 45-60 minutes. The measured data was subsequently processed by Neurocom Balance Manager Software. For the analyses of the data there...
