Diagnostic and prognostic impact of miR-122 and miR-142 in colorectal cancer

Colorectal carcinoma is one of the most frequent cancers and it is also one of the most common causes of cancer related death. It is often diagnosed at a late stage which is associated with a worse prognosis. Therefore, there is an effort to identify new biomarkers that allow earlier diagnosis. These biomarkers could predict prognosis of disease and patient's response to treatment. Short noncoding RNAs - microRNA - appears to be promising candidates, these molecules are involved in many cellular processes through RNA interference. The aim of this thesis is to summarize knowledge about application of miR-122 and miR-142 as diagnostic and prognostic biomarkers for colorectal cancer. Possible approaches to the treatment of cystic fibrosis

Cystic fibrosis is a genetic disease caused by a mutation in the CFTR gene. This leads to an absence or a malfunction of CFTR chloride channel, which is also a crutial regulator of other ion channels. This thesis was aimed at gene therapy of cystic fibrosis using CFTR- mRNA gene transfer. To determine the expression of CFTR protein, methods of indirect immunofluorescence and Western blot immunodetection were utilized. Also relative gene expression levels of CFTR gene were assessed by quantitative PCR. Experiments were carried out on a healthy lung epithelial cell line (NuLi-1), a lung epithelial cell line with F508del mutation (CuFi-1) and an embryonic kidney epithelial cell line (HEK293S). CFTR protein was visualized by previously mentioned methods using six primary antiobodies (432, 450, 570, 596, 769, CF3). Primary antibodies 570 a CF3 were found as optimal for the detection of CFTR protein by the method of indirect immunofluorescence, whereas for the detection of this protein by the method of Western blot only the CF3 antibody was suitable. It was also determined that CFTR gene is expressed in overall small levels. Its relative gene expression in the CuFi-1 cell line was approximately six times higher than in the NuLi-1 cell line. Field determination of unconfined compressive strength of rocks according to ISO 14689-1

Abstrakt/Summary The aim of the following bachelor thesis is to determine the strength under the uniaxial compression, according to the norm ČSN EN ISO 14689-1. This norm as replaced the norm ČSN 73 1001. Several changes in the rock classification take place in the chapter entitled The Field Determination of Strength Under Uniaxial Compression. Moreover, my own research also address the issue of rock identification and classification. The comparison of the own measurements with the data of laboratory reported in the literature results take place thorough the thesis.
Secondary compression of Brno Tegel

Observation and measurement of long-term volumetric deformation of clay are important in tackling different geotechnical and geological issues. The area of interest is the northern part of Vienna Basin, where Miocene clays sedimented. It is locally known as Brno Tegel. The Thesis deals with the secondary compression of clays, which can be defined by the Cα/Cc concept. The one-dimensional creep can be determined using the numerical values of the coefficient of secondary compression Cα and subsequently the thickness of overburden of sediments can be estimated. The two different theories, known as Hypothesis A and B are also discussed, which have been used to interpret the effect of creep during the primary consolidation phase. The laboratory measurements of long-term oedometric tests are presented for two undisturbed samples and one reconstituted sample of Brno clay. Construction damages in the undermined Kladno-Švermov district

The thesis is dealing with construction damages in the undermined Kladno-Švermov district. This area is situated in the Kladno Coal district, which is a part of the Kladno- Rakovník Basin. Kladno-Švermov is one of the places which have been affected by mining activity for more than last 150 years. Firstly the historical documentation, structure development and engineering geology investigations were reviewed. Secondly the area was investigated and the catalogue of 39 documentation points was made. The aim was to register visible construction damages on buildings, roads, or bricked fences, which could be caused by effects of undermining. The consequences of undermining in the Kladno-Švermov district are still apparent and the impacts of the hard coal mining are not finished. The main impact in the area is represented by subsidence basin. It has affected all the area and the damages are mainly caused by unequal descent. They are visible on some buildings, roads, or fences. Attachment number 3 is represented by commented catalogue of the main damages in the area and map of the undermined district with documentation points is enclosed as an attachment number 4.
Engineering-geological assessment of the dam construction works in the area of Giant and Jizera Mountains in the first quarter of the 20th century

The thesis is mainly about Intze's Dam contructions in 1st qaurter of the 20th. Century in the area of Giant and Jizera Mountains. In the thesis are briefly characterized nature conditions. Than is Ing. Otto Intze dr. h.c. introduced and his technical pripncples, defined for the dam construction foundations are described. Main part of this project is about realization of the reservoirs in the area of interest and 1 example of foreign dam collapse. History, contruction, technical parametres and engineering geological aspects of individual dams are described in detail. In the conclusion is an evaluation of the quality and the current state of the cuntructions.
Power supply for acquisition and communication subsystem of a current transformer

This bachelor thesis is focused on designing a power supply for measuring part and communication part of a device, which is supposed to analyze current waveforms on high voltage lines. The power supply consists of a current transformer, control electronics, and accumulation elements. The thesis analyzes the current waveforms on high voltage lines to get an overview of input data so that the right current transformer can be chosen. A working principle of these transformers is described, as well as the options for accumulation elements. Power supply requirements for the power supply are determined as well. The next part of the thesis summarises this theoretical knowledge and applies it to the schematic design. This is followed by a description of the schematic with an included description of function blocks. Additional adjustments that would improve the power supply's properties might be mentioned. This will be followed by designing a PCB layout. The final part includes the manufacturing process and determination of parameters like power, efficiency, etc. In the manufacturing process, the PCB will be printed, soldered, and evaluated on an electrical test. The electrical test inspects the functionality of the designed power supply by simulating real-life scenarios of high voltage lines. At last, an impact of the power supply on the measuring system will be analyzed.

Nowadays there is only paliative treatment available, which can be further subdivided into two groups: acetylcholinesterase inhibitors (AChEIs) and N-methyl-D-aspartate (NMDA) receptor antagonist. Donepezil, rivastigmine and galantamine represents AChEIs currently available for AD treatment. Tacrine is the first AChEIs to be approved for AD treatment, however, it was withdrawn from the market due to its side effects, especially due to its hepatotoxicity. 7-Methoxytacrine (7-MEOTA) is less toxic tacrine derivative preserving pharmacological profile of tacrine. This diploma thesis describes synthesis of multifunctional 7-MEOTA- amantadine derivatives as potential drugs to confront AD. Using Ellmanʼs method, we have established their efficacy to inhibit acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) in vitro. The objective oft his paper is to explore the issue of the opposition to the U.S. Constitution during the period of its ratification. The Constitution was produced by the Philadelphia Constitutional Convention of 1787 and altered the existing constitutional framework of the Articles of Confederation in many significant ways. Virtually all aspects of advocacy of the new Constitution have been thoroughly discussed in American historiography for obvious reasons: the Constitution of the United States continues to be the supreme law of the land and represents the oldest written federal constitution still in use. Pathophysiology of colorectal cancer. Colorectal cancer screening effect and the role of microRNA in pathophysiology of colorectal cancer.

Colorectal cancer is a serious malignant disease with an incidence of over 1.8 million new cases per year worldwide. There are about 8 000 patients diagnosed with CRC in the Czech Republic each year, and about half of them present with an advanced disease. Screening program identifies patients in the early stages of CRC resulting in overall better prognosis and survival. There is also a lack of biomarkers of early CRC detection and of response to treatment. The first aim of our project was to conduct a national multicentre prospective observational study to evaluate the impact of CRC screening within the framework of a Czech population screening programme. Between March 2013 and September 2015, a total of 265 patients were enrolled in 12 centres across the Czech Republic. Patients were divided into screening and control groups and compared for pathology status and clinical characteristics. Screening was defined as a primary screening colonoscopy or a colonoscopy after a positive FOBT in an average-risk population. The distribution of CRC stages was significantly favourable in the screening group compared with the control group (stages 0, I and II, 63% versus 43.3%; p <0.001).
