Stress resistance of PHA accumulating bacteria against conditions associated with gastrointestinal tract

This bachelor thesis is focused on stress resistance of PHA accumulating bacteria against unfavourable conditions associated with gastrointestinal tract. Two bacterial strains were chosen as model microorganisms for this purpose. Cupriavidus necator H16 producing PHA and its mutant strain Cupriavidus necator PHB-4 without the ability of PHA accumulation. Firstly, three gastrointestinal juices of different concentrations were chosen for the experimental part, namely gastric, bile and pancreatic juices. The stressed bacterial strain was then determined using a spread plate method and a flow cytometry. Other methods used include antimicrobial tests. The agar diffusion method was first tested to determine the size of the inhibition zones. The last method that has been performed is the broth dilution method, which serves to compare the absorbance of pure bacterial suspensions and stressed suspensions with the given juice of a certain concentration. The results show that the production of PHA represents a disadvantage in context of conditions associated with gastrointestinal tract of accumulating bacterial strain.
Biotechnological production of PHA copolymers containing 4-hydroxybutyrate

The proposed diploma thesis aims to study the biotechnological production of poly(3-hydroxybutyrate-co-4-hydroxybutyrate) copolymer. The subject of the experimental part was first to select a suitable bacterial strain from five selected microorganisms with different carbon precursors applied at various concentrations. The five selected microorganisms used in the experimental part include bacterial strains Cupriavidus malaysiensis DSM 19416, DSM 19379, and DSM 25816. Furthermore, the strain Thermomonas hydrothermalis DSM 14834 and Aneurinibacillus thermoaerophilus H1 CCM 8960. The experiment shows that the most suitable candidate for biotechnological production is the bacterial microorganism Cupriavidus malaysiensis DSM 19379. Finally, the biotechnological production of the copolymer was investigated utilizing a batch cultivation technique in a laboratory bioreactor.
Effect of physiotherapy in Puerperium on the Recovery of women after Caesarean Section

1 ABSTRACT The main objective: Caesarean section is currently the most frequently performed obstetric operation and probably the most common abdominal operation at all. The condition after SC is a combination of the condition after pregnancy and the condition after major abdominal surgery. Nevertheless, physiotherapy is not provided as standard in all maternity hospitals in the Czech Republic. Physiotherapy is also not normally indicated after discharge from the hospital. The aim of the present study was to prove that physiotherapy in puerperium influences the recovery of women after SC. Methods: At the beginning, 57 women participated in the study. Data from 45 women (20 women in the treatment group and 25 women in the control group) advanced to further evaluation. During the six weeks postpartum period, the women in TS completed self-therapy consisting of post-operative wound care and rehabilitation exercises. Study participants from KS did not receive a self-therapy plan or other instructions regarding post-operative wound care beyond the information given to them by maternity staff or which they sought out themselves.
