Molecular and functional characterization of the death receptor 6
Klíma, Martin ; Anděra, Ladislav (advisor) ; Živný, Jan (referee) ; Kovář, Marek (referee)
Death receptor-6 (DR6/TNFRsf21/CD358) is a receptor from the TNFR superfamily that likely participates in the regulation of proliferation and differentiation of T- and B-lymphocytes and neural cells. The 655-amino acid human DR6 is a type I transmembrane protein containing four cysteine-rich domains in its extracelular part and a death domain followed by the CARD-like region in its cytoplasmic part. Overexpression of DR6 in some cell lines leads to apoptosis, and/or to activation of nuclear factor NF-κB and stress kinases of the JNK family. In the first part of our work we focused on molecular characterization of DR6, including the analysis of its posttranslational modifications. We found that DR6 is an extensively posttranslationally modified protein including S-palmitoylation and both N- and O-glycosylation. Six N-glycosylation and one S-palmitoylation sites were precisely mapped to appropriate asparagines and cysteine respectively. The juxtaposed linker region (between cystein-rich domains and the transmembrane part), which also contains Ser/Thr/Pro-rich region with clustered putative O-glycosylation sites, is required for the plasma membrane localization of DR6. N-glycosylation, but interestingly not S-palmitoylation, may play a role in targeting of DR6 into detergent-resistant...
Effect of iron overload on the induction of apoptosis in mammalian cells
Kabíčková, Tereza ; Balušíková, Kamila (advisor) ; Klíma, Martin (referee)
Iron cations are an important metal ions required to number of essential cell functions. On the other hand, ferrous iron can be very toxic as well. When surplus iron is present in cells, it can catalyze the formation of reactive oxygen species (especially hydroxyl radicals) by Fenton reaction. Iron homeostasis is predominantly regulated by very strict mechanisms on the level of iron uptake into the body. Moreover, iron absorption, transport and storage within the body can be also regulated using complex mechanisms which differ on the level of individual cells and on the level of whole organism. Deregulation of iron homeostasis causing an iron overload and generation of reactive oxygen radicals can evoke serious cell damage leading up to apoptotic cell death. Excess iron storage and subsequent development of oxidative stress can affect lot of different tissues in the body. The organ damages such as fibrosis, cirrhosis, hepatocellular carcinoma, heart failure, loss of β cells and glucose intolerance or diabetes mellitus in patients with iron overload are very often seen. Nevertheless, the apoptosis induced by iron overload has not been well elucidated yet. There are no complex informations about the precise mechanism by which oxidative stress affects different cell types or whether there are other...
Personal communicator for strategic sports
Válik, Martin ; Klíma, Martin (referee) ; Macháň, Ladislav (advisor)
The aim was to design connept of personal communicator useable for strategic sports such as airsoft or painball. Personal communicator provide wireless transmission of position provided by GPS module, eventually other data via RF module. Interactive user interface is solved by TFT touch display. For controlling of modules is used 32-bit ARM based microcontroller. The output is demonstration of functionality of communication and vizualization of distance between two designed devices on TFT display.
Universal timer for sport activity
Janůš, Tomáš ; Klíma, Martin (referee) ; Macháň, Ladislav (advisor)
This bachelor thesis deals with a proposal and implementation of wireless electronic time measurement. In the thesis a proposal and implementation of simple wireless time measurement is described. Its part is also a proposal and creating of the optical barrier. The theoretical part further contains description and usage of various sensors. All sensors are wirelessly interconnected with the main device. The time measurement is being processed in the computer by means of a created programme which enables to control the time measurement and to save measured times as well.
The via interconnection in LTCC
Sedliak, Erik ; Klíma, Martin (referee) ; Kosina, Petr (advisor)

The influence of a pre-print thick layer paste viscosity on forming of layer
Ondráček, Michal ; Švecová, Olga (referee) ; Klíma, Martin (advisor)
This bachelor thesis deals with the theory of thick film technology and applying these layers. The work also includes basics of liquid rheology, methods of measurement viscosity and ways of mixing liquids. The main aim is to determine the influence of viscosity of thick film pastes for printing quality of the test structure using screen printing, in particular its flatness and sharpness. The aim is also measure the dependence of viscosity thick film pastes on mixing time.
The multimedia textbook of microelectronic technologies
Černík, Pavel ; Klíma, Martin (referee) ; Adámek, Martin (advisor)
In this bachelor's thesis, there is summary of technological process used in the manufacture of microelectronic circuits. For easier understanding of the processes, there are appended videos, animations and pictures of these technologies. Textbook is created as web pages, that are dynamicaly generated from data stored in MySQL database according to predefined HTML template. Articles and pictures can be added using simple administrative interface.
