Nováková, Anežka ; Skálová, Lenka (advisor) ; Karlíčková, Jana (referee)
Charles University Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Anežka Nováková Supervisor: prof. RNDr. Lenka Skálová, Ph.D. Consultant: Ing. Lenka Langhansová, Ph.D. Title of diploma thesis: The effects and metabolism of anthelmintics in fodder plants Drugs are potentially dangerous contaminants of the environment. The entry of anthelmintics into the environment is mainly via excrement of treated animals. Anthelmintics can negatively affect the ecosystem by acting on non-target organisms such as plants and soil microorganisms. The accumulation of medicines in plants is also at risk. The consumption of these plants may promote the development of resistance in helminths. This thesis looked at the study of the influence and biotransformation of frequently used anthelmintics of albendazole (ABZ), ivermectin (IVM) and monepantel (MOP) in the forage plants of alfalfa seté (Medicago sativa) and the meadow clover (Trifolium pratense). Plants cultivated in in vitro conditions and plants grown in a greenhouse were used for evaluation. Phytotoxicity was monitored on seeds and whole plants by measuring the length of the roots, by determining the content of proline, light-collecting pigments and the total protein content. Mild phytotoxicity was observed after ivm...

Charles University Faculty of Pharmacy in Hradec Králové Department of Pharmaceutical Botany Candidate: Radka Rzepecká Supervisor: Ing. Kateřina Macáková, Ph.D. Title of Thesis: Interactions of alkaloids with transition metals I. Copper is one of the essential trace elements that are necessary for the proper functioning of the organism. Copper is a significant component of many enzymes that affect various metabolic processes in the human body. It is important that the level of copper in the body is regulated because its deficit or excess lead to a variety of pathological conditions. Alkaloids are secondary plant metabolites that are distinguished by numerous biological activities. In this thesis the copper-chelating and copper-reducing activity of eleven isoquinoline alkaloids was measured: boldine, isocorydine, (+)-bulbocapnine, (+)-corydine, glaucine, (-)-sinoacutine, (-)-californidine, (-)-escholtzine, platycerine, (-)-fumaricine, and (+)- parfumine. Alkaloid activity was measured at four (patho) physiological pH values by a verified spectrophotometric method using two indicators: bathocuproinedisulfonic acid disodium salt and hematoxylin. Based on the results, structure-effect relationships were derived. The results show that none of the tested substances was able to chelate copper ions. In...

Hradecká, Michaela ; Karlíčková, Jana (advisor) ; Kašparová, Marie (referee)
Hradecká M.: Natural compounds applicable for the treatment of metabolic syndrome, Diploma thesis 2018/2019, Charles University, Faculty of Pharmacy in Hradec Králové, Department of Pharmaceutical Botany, pp. 60. Occurence of metabolic syndrome is increasing worldwide in children and adults. The use of natural compounds is one of possibilities in the prevention and treatment of metabolic syndrome. These compounds may act complexly or affect individual risk factors associated with metabolic syndrome. Selected plants with these effects are incorporated into my diploma thesis (for example Allium sativum, Crataegus laevigata, Hibiscus sabdariffa, Persea americana, Rosmarinus officinalis, Silybum marianum and Vaccinium myrtillus). The alga Undaria pinnatifida and fungus Pleurotus sajor-caju are also mentioned. It is necessary to carry out other clinical studies, where the positive effect of mentioned constituents will be confirmed, which could be added to the list of phytopharmaceuticals used to prevent, slow down or treat the metabolic syndrome in the future. Keywords: natural compounds, metabolic syndrome

Charles University Faculty of Pharmacy in Hradec Králové Department of Pharmaceutical Botany Candidate: Zuzana Lomozová Supervisor: PharmDr. Jana Karlíčková, Ph.D. Title of Thesis: Interactions of tamarixetin and isorhamnetin with copper Keywords: tamarixetin, isorhamnetin, chelation, reduction, copper, bathocuproine, hematoxylin Copper is a biogenic trace element important for proper function of human organism. It is an essential part of several enzymes and is involved in metabolic processes in the body. Excess or lack of serum copper can lead to pathological conditions. Copper chelating agents are used to treat copper toxic effects. Flavonoids are polyphenolic substances belonging to secondary metabolites of various plants. They are part of the human diet and have a positive impact on our health. They exhibit antioxidant and anti-inflammatory effects and are able to chelate transient metals, especially iron and copper. Chelation therapy is currently used in Wilson's disease in which copper is overloaded. In the future, the chelating effects of flavonoids could be used to treat neurodegenerative diseases, cardiovascular diseases or cancer. In this diploma thesis, interactions of two flavonoids (namely tamarixetin and isorhamnetin) with copper ions in different buffers were tested....
Environmental risks of anthelmintics

Charles University Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Nikoleta Dojčanská Supervisor: prof. Ing. Barbora Szotáková, Ph.D Title of diploma thesis: Environmental risks of anthelmintics Anthelmintics are drugs used in human and veterinary medicine for the treatment and prophylaxis of infectious diseases caused by parasites- helminths. They are admittedly beneficial to the treated organisms. However, their overall effect on the ecosystem has not yet been sufficiently explored. Anthelmintics, together with their metabolites, enter the environment in multiple ways. One possibility is directly through urine and excrements of treated livestock or indirectly by leakage of contaminated manure into surface and groundwater. Such pollution has an effect on various animal organisms and plants that have the ability to uptake xenobiotics and remove them through metabolic pathways. The aim of this work was to investigate the effect of antihelmintics ivermectin and fenbendazole on soybean plants (Glycine max). We investigated the concentration of total phenolic compounds spectrophotometrically using the Folin-Ciocalteu method. Using the colorimetric method of aluminum chloride, we found the concentration of flavonoids in soybean plants. The concentration of isoflavons-...

Comparison of interactions of equol and desmethylangolensin with iron and copper
Němcová, Hana ; Karlíčková, Jana (advisor) ; Matysová, Ludmila (referee)
Charles University Faculty of Pharmacy in Hradec Králové Department of Pharmaceutical Botany Candidate: Hana Němcová Supervisor: PharmDr. Jana Karlíčková, Ph.D. Title of Thesis: Comparison of interactions of equol and desmethylangolensin with iron and copper Iron and copper are essential trace elements, which are important for our body. Both elements have a significant effect on the correct function of organs and make part of many enzymes. They are able to accept or donate electrons - conversion between oxidized (Fe3+, Cu2+) and reduced (Fe2+, Cu+) forms. If these metals are excess in organism, they are accumulated in the cells and mediate the creation of free radicals, that destroy cell structures. This deficiency is treated with chelators, which facilitate the excretion of metals from the body. Isoflavonoids are polyfenolic substances, which can have antioxidant effects and they are involved in the scavening of free radicals. Isoflavonoids can have also a pro-oxidative effect, because they are able to reduce metal ions. In this study were tested interactions (chelation and reduction) between the metabolites of isoflavonoids (equol and desmethylangolensin) and ions of iron and copper. Both metabolites only weakly chelate metal ions, but they significantly reduce cupric ions. KEYWORDS: Iron,...
