Dientamoeba fragilis is a cosmopolitan intestinal protist colonizing the human gut with varying prevalence depending on the cohort studied and the diagnostic methods used. Its role in human health remains unclear mainly due to the very sporadic number of cross-sectional studies in gut-healthy populations. The main objective of this study was to expand knowledge of the epidemiology of D. fragilis in gut-healthy humans and their animals in the Czech republic.

The main aim of this study was to summarize available information about the gut protist Dientamoeba fragilis with special focus on the influence of selected factors such as age, gender, travelling or lifestyle to its colonization. It was first described in 1918 by Jepps and Dobell. First, D. fragilis was classified as an amoeba but later, based on molecular studies, was revealed that it is closely related to the trichomonads. Dientamoeba fragilis has three life forms - cyst, precyst and trophozoite. Until recently, only trophozoite of D. fragilis stage was known and the way of transmission between individuals was not clear. Thus, it was hypothetised that D. fragilis trophozoites are transmitted with eggs of some worms colonizing gastrointestinal tract. Recently, the cyst and precyst stages were discovered. In addition to humans, the presence of D. fragilis has been observed in primates, pigs and some pets. Mice seem to be successful experimental models. In terms of age factor, D. fragilis is most common in young children. There have not been revealed any correlations between occurrence of D. fragilis and gender. Pathogenicity of D. fragilis is another controversial fact about this organism. In past, it was considered a pathogen causing gastrointestinal disorders, mainly in children. However, a few recent epidemiological studies focused on healthy human population highlighted that D. fragilis appears to be more common in people without no gastrointestinal problems. Despite of this, there is still a small number of epidemiological studies on people with no intestinal problems, thus, it is difficult to compare prevalence of D. fragilis between healthy populations and patients suffering from intestinal diseases. Occurrence of D. fragilis is more prevalent in developed countries in contrast to Western populations. RT-PCR was evaluated as the most optimal method of detection of this organism from stool samples. The role of this intestinal protist in health and disease remains the subject of scientific discussions. But according to some new studies this organism tends to commensal character in the human body. At the end of this theoretical work is a brief summary of the current experimental work, which will be presented in detail in the master thesis.
Assessment of motor skills in children with osteogenesis imperfecta

Diplomová práce Hodnocení motorických dovedností dětí s OI Abstract The thesis deals with the assessment of motor skills and quality of life in children with osteogenesis imperfecta. The theoretical part summarizes aetiopathogenesis, clinical features, kinesiological abnormalities and the treatment strategies. A particular part describes therapeutic interventions during the various stages of childhood with an alignment to multidisciplinary cooperation. Furthermore, tests of motor skills and quality of life are presented. The practical part assesses the applicability of the BOT-2 for the evaluation of motor skills and PedsQL for the evaluation of the quality of life in children with OI. Moreover, the BOT-2 standardized test score evaluates and compares motor skills and quality of life in children with osteogenesis imperfecta to their peers. Contemporaneously, it investigates the effect of motor skills on the quality of life in children with OI. Based on the results the thesis confirms the hypothesis - worsened both gross motor skills and fine motor skills and the dependency of quality of life on the level of motor skills. For my thesis I chose the topic - Business plan. The introductory part of this work was devoted to defining the terms of business in the Czech Republic and the possibility of establishing a business. Next, I looked at creating a theoretical basis for optimal processing of a business plan and principles that the project must meet. Application of the work was focused on the establishment of a specific business plan - A comprehensive wedding services. The business plan was created based on the principles defined in the theoretical part.

Tato práce je věnována managementu kvality a jeho přístupům, které vedou k dosahování vysoké úrovně jakosti výrobků a služeb. V teoretické části byly vymezeny hlavní přístupy systému managementu kvality, které lze v podniku zavést. Jednalo se o přístupy správné výrobní a hygienické praxe, přístup podle normy ISO 9001, celosvětově uznávaný TQM a jeho model Excelence EFQM.Praktická část je věnována principům správné výrobní praxe a problémům, které byly spojeny s jejím dodržováním s ohledem na čistou finální produkci.
