Study of the DNA recombination mechanisms in plants

DNA double-strand break (DSB) is a dangerous type of DNA damage, but it also serves in controlled increase of genetic variability. The two major DSB repair pathways are homologous recombination (HR) using homologous sequences and non- homologous end joining (C-NHEJ). Two model plants Arabidopsis thaliana (Arabidopsis) and the moss Physcomitrella patens (Physcomitrella) differ in DSB repair strategies. Arabidopsis uses C-NHEJ, however Physcomitrella prefers HR. These plant models are compared on the basis of measurement of DSB and single strand breaks (SSB) repair by comet assay. The half-life of the first rapid phase of the DSB repair is about 5 minutes in both plant species. Although the C-NHEJ is considered as the main DSB repair pathway in Arabidopsis, rapid repair is independent of AtLIG4 and AtKu80, suggesting the existence of the effective backup non-homologous repair pathways (A-NHEJ). In Physcomitrella, the rapid DBS repair dominates in mitotically active cells and is also independent of PpLIG4. Conversely, PpLIG4 is surprisingly involved in the repair of the DNA alkylation damage. Cloud computing and its application in practice

Goal of this work is to define the concept of cloud computing. Then I analyzed the market for cloud computing, the use in business for small and medium enterprise. The work deals with the analysis of the Czech market, SaaS, IaaS and for the world market we provide an overview of catalogs, which provides cloud computing providers. After analyzing the markets deal with a case study to compare the solution of the classical way and the use of technology solutions for cloud computing. The definition of cloud computing, I used the definition of many foreign authors, and the definition of NIST. When analyzing the market, I searched online applications and SaaS applications and evaluate whether the SaaS model, or ASP. In the case study I conducted an analysis of TCO costs for ICT / IT solutions in the classical pathway and the use of cloud computing. When we came out in the analysis as a better solution using cloud computing. As the main contribution of the paper I define a case study that compares the address ICT / IT in a classic way, using cloud computing. In this case the forecasts were to materialize, that cloud computing is based on the cheaper end customer. The structure of work is the definition of the concept and its distribution for better understanding. Then I planted the notion of cloud computing in its historical context, I can answer two questions, namely whether it is a buzzword, and why cloud computing flourishes today. Then I conducted a market analysis, cloud computing, when I tried to search and select products that I use for finding work in the case study, comparing two solutions.
The Psychological and Spiritual Support in the Police of the Czech Republic

The thesis deals with the description of the psychological and spiritual support for the members of the police force. Its basis is the definition of the police force as the service to the public while the policeman is perceived as both the representative of the state force and the human being. In the part dealing with the professional ethics one points out to the usefulness of the ethical codex for a policeman as a possible inspiration for one's responsibility in the role of a policeman.Next, negative phenomena influencing a policeman are described, such as stress, a traumatic event,crisis,and a burn-out syndrom. The thesis considers the needs of the man linked to the needs of a policeman proper. Based on the information collected it then deals with the psychological and spiritual support. The psychological support in the police force has got a certain framework defined in the form of a crisis intervention, a psychological advisory service and a posttraumatic intervention care. A spiritual support has no specific concept defined. A priest has his role in posttraumatic intervention teams only. Based on the research carried out and on the literature available the possibility is outlined for the provision of not only psychological but also for the spiritual support.
