Possible perspective use of ghrelin mechanisms in the treatment of opioid addiction

This dissertation deals with the narrow issue of the new perspective research direction, which is research of the possibility of using ghrelin antagonism in the treatment of opioid addiction, which has been rarely studied yet. It summarizes the general principles of neurobiological mechanisms of drug addiction as well as treatment guidelines and procedures based on these findings. The definition of addiction as a disease together with the factors contributing to its formation and course, are presented in this dissertation. The definition includes individual repetitive phases (cycles) of the substance addiction, along with a description of the anticipated involvement of individual brain structures and basic neuromediators involved in these phases, including references to the most important literary findings on the role of ghrelin in the mechanisms of addiction, focused on mechanism of action of ghrelin in the opioid addiction. Göteborg University, Sweden; The Sahlgrenska Academy; Institute for Neuroscience and Physiology; Department of Pharmacology Nowadays, the number of patients with lower urinary tract symptoms increases. The goal of this project was to explore the effect of 4-DAMP (4-diphenylacetoxy-N- methylpiperidine) on the regulation of the overactive bladder syndrome (OAB). Furthermore, the effect of induced cystitis by cyclophosphamide on the urothelium changes, was observed. It was found that there are some unspecified mechanisms that can compensate for the present cystitis. But only poor effect on incontinence and different effects in different doses lead to conclusion that 4-DAMP is not the best for the treatment, but additional tests are needed.
Regulation of autonomic activation and inhibition in the urinary bladder

The regulation of the urinary bladder function involves interplay of many signaling systems. Any disturbance within these principles always result in bladder dysfunction, manifested by urge to void and in many cases leading to incontinence. The main objective of this thesis was to characterize interactions between muscarinic, purinergic and adrenergic receptor systems in in vitro study. The experiments were performed on rats (300-350g; Sprague-Dawley strain), that after killed with an overdose of pentobarbital, were removed a smooth muscle tissue sample (6x2 mm) and put through contraction studies in organ bath with the presence of muscarinic agonist (carbachol - reference concentration 10-5 M) and following agonists (ATP, 2-chloro-adenosine) and antagonists (pirenzepine, methoctramine, 4-DAMP, 8-sulfophenyltheophylline) in different, ANS active, concentrations. The volume of the substances, added to the bath, was always 100 μl. The results revealed a high potential of antimuscarinic 4-DAMP on carbachol evoked contractions inhibition. This effect was 2000x higher compared with methoctramine and 600x higher than pirenzepine. The Bachelor thesis studies the process of providing financial services, mainly mortgage loans, via Internet. The introduction (first part) includes a detailed description of the topic. In the main body (second part), I am analyzing the process of providing financial services via Internet. In conclusion (the third part), I use my analysis to study a new approach and a possible solution, whose feasibility I will try to prove with my new project.
