Role of ABC transporters in pharmacoresistant epilepsy

The diploma thesis summarizes the recent knowledge on the possible role ABC transporters in mechanisms of refractory epilepsy. Approximately 30% of patients with epilepsy are refractory to treatment. An important characteristic of pharmacoresistant epilepsy (PE) is that most patients are at the same time resistant to several antiepileptics (AED) even though these drugs act by different mechanisms. It was suggested that one of the main cause of pharmacoresistance is the decreased drug uptake into the brain mediated by multidrug transporters in the blood-brain barier (BBB). The ABC transporters studied in relation to PE are the following: P-glycoprotein (Pgp, coded by MDR1 gene), some members of the multidrug resistance protein (MRP) family and the breast cancer resistance protein (BCRP). The over-expression of the ABC transporters was demonstrated in capillary endothelial cells of BBB, and astrocytes and neurons of epileptogenic brain tissue. Therefore modulation of ABC efflux transporters of the BBB could be a novel strategy to enhance the penetration of AED into the brain. Coadministration of inhibitors with less toxicity, higher potency and specifity to Pgp together with AED could thus reverse PE and achieve seizure control in epilepsy. Employee Education in the Process of Adaptation

5 ABSTRACT The presented thesis, which is complemented with a case study, deals with education of employees in the process of adaptation. Attention is paid to the theoretical anchoring of this issue by specialised literature. On the theoretical level, the thesis outlines adaptation and adaptation process, employee training as well as education of employees who are in the adaptation process. This theme is deliberately shown in connections and actual context through introduction of the system of education of employees in the adaptation process as it is set in a selected company. It discusses the way of identification of educational needs, and the planning, implementation as well as subsequent evaluation of the education. The aim of the thesis is to provide a comprehensive view of education of employees in the process of adaptation, and to find out within empirical research how the system of education of employees in adaptation process is set in a specific selected company. Logistics and Production Management in a Selected Company

The goal of the Master´s thesis is to analyse operations of the company dealing with the production of plastic and wooden windows and to focus on production organization and distribution and to suggest improvements measures. The Master´s thesis concentrates on production analysis and distribution of company WindowStar s.r.o which is one of the daughter companies of Austrian holding D.E.I.N. Haus. Company WindowStar s.r.o. resides in Planá nad Lužnicí and deals exclusively with the production of plastic and wooden windows. There are frequent changes of trends at purchasing windows, competition in this branch is great and therefore the company has necessarily to increase its competitiveness and financial stability. Analysing the production and distribution there were found out some places possible to be improved. Afterwards there were suggested particular measures leading to savings. I recommend the company to change buyer of plastic waste and non-congruous plastic products, further, to make a trim-to-size plan for cutting aluminium both for plastic and wooden windows. Finally, I suggest changing the supplier of glazing slats necessary for production of wooden windows and giving more effort to negotiations and cooperation with suppliers.
Information and Material Flows in a Selected Building Company

The aim of the dissertation is to analyse material and information flows in the specific conditions of the custom-made production in the building industry, possibly to suggest sectional measures. The dissertation is focused on analysis of material and information flows in the Austrian company ELK a.s. residing in Planá nad Lužnicí. The company deals with the custom-made production of low-energy prefabricated houses. The production of prefabricated houses is becoming more and more popular in the sphere of the building industry, therefore it is necessary to improve material and information flows. Analysis described in this dissertation was carried out on the basis of particular order realized by this company. There were found out several problematic spots and the company was recommended to eliminate them. On the basis of the carried out analysis it is recommended to create a complex awareness system which should involve not only changes in the company, elaboration of forms for checking which would be possible right on the building site, but also to introduce the system on the level of the web interface which would improve cooperation with subcontractors. The suggested changes will help the company to improve the quality of services and to reduce the extra costs which occurred in the process of solving the problematic spots.
