Analysis of rRNA genes in variets Brassica napus

Brassica napus (AACC, 2n = 38) is an allotetraploid species derived from the parentel diploid species Brassica rapa (AA, 2n = 20) and Brassica oleracea (CC, 2n = 18). The aim of thesis was to carry out the genetic and epigenetic analysis of high-copy rRNA genes (or rDNA) in several varieties of hybrid species B. napus. The experiments involved determining the ratio of parental genes in hybrids, sequencing and methylation analysis of the promoter region of rDNA. Using Southern hybridization, it was revealed significant variability in the number of parental rDNA units between each variety. Data from sequence analysis were in good agreement with the results of Southern blot. Genetic recombination between parental rDNA units was revealed in one variety by DNA sequencing of promotor region. To study methylation, bisulfite sequencing was performed. It was found out that rDNA units of B. rapa origin have a higher value of methylation than units originated from B. oleracea.
Identification of probiotic lactobacili in dairy products

The genus Lactobacillus is part of intestinal microbiota which constitution plays important role in health and well-being of the host. Probiotic properties of lactobacili have been studied in lot of studies and clinical effects (immune enhancement, prevention of diarrhoeal disease etc.) of some probiotic species have been documented. Therefore bacteria of genus Lactobacillus are used in the production of fermented dairy products and probiotic preparations. PCR method based on amplification of 16S – 23S rRNA intergenic spacer regions was developed for identification of Lactobacillus species presented in dairy products. Whole DNA obtained by phenol extraction from dairy product Actimel Natur was used in this work as template for PCR. Purified DNA was amplificated by genus-specific PCR. Bacteria of genus Lactobacilllus were detected due to the detection of specific PCR product. Using species-specific PCR probiotic species Lactobacillus casei was detected in the Actimel Natur in quantity that approximately corresponded to declared amounts.

The theoretical part of the thesis explains the necessity of regulation the human reproduction since the population explosion launched by the agrarian revolution that was reduced by the number of illnesses and infections would lead to overpopulation. The contraceptine efforts having their roots in the ancient times led to the development of methods which vary as far as reliability is concerned . A midwife is in charge of providing high- quality health education to promote healthier family life, family planning and positive parenthood. Unfortunately, the Czech midwives have none of these authorities arising from the strategic documents. The aim of the empirical part is to find out to what extent the Czech women usány hormonal contraception are informed as well as their view on the role of a midwife in the sphere of family planning.
