Synthesis of unsymmetrical derivatives of azaphthalocyanines IV. Zuzana Dvořáková Department of Pharmaceutical Chemistry and Drug Control, Faculty of Pharmacy in Hradec Kralove, Charles University in Prague, Heyrovskeho 1203, Hradec Kralove, 50005, Czech Republic Phthalocyanine (Pc) and their azaanalogues (AzaPC, which have some carbons in their macrocyclic system replaced by nitrogens) belong to group of photosensitizers, which are used for photodynamic therapy in treatment of various cancer diseases. The effect is given by ability to generate reactive oxygen species (e.g. singlet oxygen) after light absorption. The aim of the work is to synthesize AzaPc with optimal properties and with a high efficiency in singlet oxygen production. New unsymmetrical AzaPc with different numbers of pyridyl on periphery were synthesized with zinc as central metal. Different ZnAzaPc were prepared using a statistical condensation starting from two precursors (5,6-bis(tert-butylsulfanyl)- pyrazine-2,3-dicarbonitrile (A) and 5,6-di(pyridin-2-yl)pyrazine-2,3-dicarbonitrile (B)). From the mixture of six posibble AzaPc, four of them (AAAB, ABAB, AABB, ABBB) were isolated by column chromatography. Social network discourse - Facebook, the virtual community

Having focused on Facebook, the virtual community, the aim of this thesis was to study the social network discourse. Due to very complicated structure of the social network my study dealt with the discourse on its contextual level, which enabled us to interconnect particular attributes of this difficult system and gradually revealed their mutual influence. The contextual line of appropriate chapters then joined the discoursive factors of the social network, starting with their makro level continuing to their very specific manifestation - the communication acts. Within the conclusions of this thesis it is to finally analyze in which form the concrete discoursive perspectives have projected into the more exact demonstration of the social network discourse.
Designing of the curriculum for vocational education and specific part of the school leaving examination at the secondary school providing four-year course of upper secondary vocational education

This thesis describes designing of the curriculum for vocational education and of the branch specific part of the school leaving examination. I have set two main targets: 1. To design the Framework Educational Programme (RVP) for the branches with the graduation from the "Ecology and Environment". 2. To observe the approach to the school leaving examination at "střední odborné školy (SOŠ)" (secondary school providing four-year course of upper secondary vocational education) that are focused on the natural science. Both of these goals were accomplished. Ad 1. As the coordinator for the ecological study branches at the National Institute of Technical and Vocational Education in Prague (NUOV), I coordinated and participated in the designing of the branch specific part of the RVP for the new branch "Ecology and Environment". Four original branches were merged into this single one after the revision and change of the branch system. The new RVP for Vocational Education for the branches with the school leaving examination "16-01-M/ 01 Ecology and Environment" was submitted to the Ministry of Education and it was approved in the May, 2008. Accordingly, the schools have to create their own Schools Educational Programme (ŠVP) within two years and they have to apply it at the classes from 1 September, 2010.
The possibilities of assessment of cognitive functions in neurological patients. Adaptation and validation of the Neuropsychological Assessment Battery (NAB)

This dissertation focuses on available means of the assessment of cognitive functions in neurological patients in the Czech environment. The main goal was to verify the psychometric qualities and usability of the Czech version of the Neuropsychological Assessment Battery (NAB). NAB is a test battery that assesses attention, language, memory, and spatial and executive functions. The theoretical part of the dissertation introduces the basic cognitive functions and diagnostic tools for their assessment; the next chapter focuses on NAB and especially its psychometric characteristics. Third chapter describes specific diseases (epilepsy and psychogenic seizures), their manifestations and impact on cognition. Empirical part included participation in the adaptation of NAB to the Czech language. During the research, a group of healthy volunteers (n = 116) and patients from the Epilepsy Monitoring Unit (n = 60) were tested by the NAB Screening Module. To verify the convergent validity of NAB-SM we administered Repeatable Battery for the Assessment of Neuropsychological Status (including an Effort Index) to a part of the sample. Executive Dysufnctions in the Schizophrenia Spectrum Disorders

Schizophrenia is from a considerable part genetically conditioned. One of the heritable characteristics of this disorder is a cognitive deficit, which is to the some point even detected in healthly siblings of individuals with schizophrenia. One of the cognitive deficits, which according to recent research is a candidate for its consideration as a specific heritable characteristic as well, is also executive dysfunction. Executive dysfunction is correlated with changes of functional connectivity in the specific neural circuits. In this bachelor thesis, the functional changes are discussed and are associated with results of neuropsychological tests. Submitted research proposal aims to replicate the executive dysfunction (specifically in the domains of inhibitory control, working memory and cognitive flexibility) of schizophrenics and their first-degree relatives. In the matter of replication of this result, executive dysfunction and its neural correlation might be considered as a valid endophenotype, potentially useful in diagnostics and prevention of this serious disorder. This bachelor thesis aims to the psychological aspects of obesity with an emphasis to the role of depressive symptomatic. In the first chapter, there is brief information about depression and obesity as separate diagnoses. The second, the most comprehensive chapter, is concerned with a comorbidity of these diseases. It contains information about prevalence, development mechanisms and intervention options. The third chapter summarizes information about a role of psychology in bariatric surgery with an emphasis to depression. Selected Psychological Aspects of Healthy Lifestyle

The bachelor thesis is dedicated to psychosomatic aspects of mental disorders. It describes interdisciplinary knowledge about communication between brain, intestine and its microbiome. The theses summarizes the role of the Gut-Brain-Microbiome axis in the aspect of thinking and feeling. It describes what can happen when the microbiome is disturbed in relation to mood and health disorders. The theses also deals with the ways of improving a mental state by influencing the Gut-Brain- Microbiome axis. In particular, it focuses on the contribution of probiotic cultures and certain diets to depression and anxiety. DNA POLYMERASE STOP ASSAY FOR DETECTION OF G-QUADRUPLEXES

In this work we performed DNA polymerase stop assays while using template DNA derived from promoter regions of VEGF and c-Myc proto-oncogenes under variety of experimental conditions. Partial stopping of DNA synthesis along the template strand was observed in the presence of K+ ions due to formation of stable G-quadruplex structures. In contrast, Na+ ions alone were unable to stabilize G-quadruplexes to stop the reaction at their site. These data suggest that K+ and Na+ ions, which are both known to stabilize G-quadruplexes, do this in different manner or extent.
