Additionally, it allows readers to navigate through the abundance of published documents and standards and provides an overview of wireless communication methods. As this topic is extensive, this publication is intended to be a practical tool for everyday use or a reference point for finding more detailed information in the specific area of interest.
Frost weathering of sandstones: mechanisms and laboratory experiments

Frost weathering of sandstones: mechanisms and laboratory experiments This thesis studies and compares different mechanisms of frost weathering, and further describes the characteristics of the rock material and the external environment, which affect this process. The effects of frost action were related to sandstones. The thesis consists of a literature review and practical experiments. The literature review indicates, that frost weathering of sandstones is due to their characteristics (higher porosity, grain structure) rather influenced by mechanisms based on water migration through the pore space of freezing rock. Beside the properties of the sandstone material, also the water content, the final temperature of freezing and the rate of the temperature fall are important. The experiments were performed on samples of Cretaceous sandstones (thick-bedded and argillaceous sandstones of Bohemian Paradise) and Carboniferous arkoses of Syřenov formation. Samples were subjected to repeated cycles of freezing at - 20 řC and thawing in distilled water. The weathering progress was monitored by measuring the weight loss and damage description. Design of the Southern Connection from the High-Speed Line to Svetla nad Sazavou Station

This master thesis contains the design of the alternative northern connection of high-speed track Prague – Brno – Breclav to station Svetla nad Sazavou, including optimization of the main track between kilometres 95.3 and 102.3. This connection will be used as the temporary end of the high-speed track. Design speeds of the connection are 160 kph and 200 kph, switches are in kilometres 95.9 and 98.5.

Identification and isolation of primordial gonocytes in sturgeon

Primordial gonocytes (PGCs) in some animals, including fish arise after fertilization in extragonadal region from maternally inherited germline cytoplasm, and migrate to the future gonads region during embryogenesis, where differentiate into gametes. PGCs formation and migration patterns have been studied in several species models, and it is known that these patterns differ from each other. Sturgeons belong to class ray-finned fishes(Actinoptergii), in which the sturgeon phylogenetic position is an out-group to teleost fishes, the sturgeon development pattern is more similar to amphibians than teleost fishes. In this study, we demonstrate an injection technique for sturgeon PGCs visualization by GFP nos1 3'UTR mRNA. We found that the Sterlet(A. ruthenus) PGCs are specified in the vegetative pole of the embryo. Subsequently, we reported the PGCs migration route. The arisen PGCs actively migrated on the yolky cell mass, yolky extension, and after that passively moved to gonadal ridge. This study provides evidence that the PGCsare specified by maternally inherited germplasm, located in the vegetative part of the embryo. Sturgeon PGCs specification was similar to that of anuras, but the migration pattern resembled that of teleost. Furthermore, we successfully isolated PGCs to next needed studies.
Location of Balbani cytoplasm in sturgeon eggs as primordial gonocytes precursor during fertilization

The oocytes of many organisms, including fish, contain germ plasm. Germ plasm is maternally inherited determinants of germ cells. New-found germ plasm has been also described in previtellogenic sturgeon oocytes as Balbani cytoplasm. It contains precursors of germinal granules so called nuage. Germinal granules have been detected as a part of primordial gonocytes (PGCs) and according to some studies it play the role in the formation PGCs. PGCs are embryonic cells from which the gametes are differentiated. These cells has potential to transmit genetic information to the next generation. PGCs or subsequently spermatogonia therefore have the potential to be of value for gene banking and cryopreservation, particularly via the production of donor gametes with germ?line chimeras. Nevertheless the first step for successfully transplantation is visualization of PGCs. It is usually done using injection of GFP (green fluorescent protein) mRNA conjugated with mRNA, such as vasa and nanos, to the area of egg with PGC precursor localization (nuage) early after fertilization. Electron microscopic observation at this study revealed three main principal layers (a topmost alveolar layer, a zona radiata externa and a downmost layer as an zona radiata interna), a layer of cortical granules in unfertilized eggs and a perivitelline space in fertilized eggs; then germ plasm, containing nuage, which was dispersed only in the vegetal pole of ovulated sturgeon eggs. In addition, we observed formation interspace between the zona radiata externa and zona radiata interna. Sturgeon eggs have too strong envelopes for injection by glass needle. The glass capillary needle penetration could be facilitated by separation of parts of the egg envelopes as the alveolar layer and the zona radiata externa from interspace, which created by separation of the zona radiata externa and zona radiate interna by tweezers.
