Mitochondrial permeability transition pore and its role in cardioprotection

The mitochondrial permeability transition pore (MPTP) is a non-specific voltage dependent channel which is located in the inner mitochondrial membrane. High calcium concentration and oxidative stress are main inducers of MPTP opening in a tissue which is affected by ischaemia and subsequent reperfusion. Morbidity and mortality of pacients who suffer from acute myocardial infaction or cardiochirugical operation, depends on the size of ischemic- reperfusion injury (IRI). The methods of IRI attenuation are based on the inhibition of the MPTP through farmacological intervention or ischemic conditioning. This thesis summarizes the current knowledge about the MPTP structure, regulation and its role in cardioprotection. The role of the adrenergic system in the process of cold acclimation

The ability to keep constant body temperature is necessary for mammals. There are effective strategies developed during the evolution - shivering and nonshivering thermogenesis, which start during cold exposure. This thesis is focused on the nonshivering thermogenesis which occurs in brown adipose tissue and is controlled by norepinephrine released from the sympathetic nerves if the cold stress persists for a long time. The principle of the heat production is based on function of the uncoupling protein 1 which disrupts the proton gradient in mitochondria thereby releasing heat the waste product. Similar mechanism of energy dissipation is used not only during cold exposure but also after digestion of high fat diet, which may provide clues for potential therapeutic treatment of obesity and associated metabolic diseases. This study summarizes current knowledge about the role of adrenergic signalling in the process of cold acclimation. The role of incretin hormones in the development of type 2 diabetes mellitus

Incretins are gut hormones secreted by cells located in the small intestine in response to ingestion of nutrients. The aim of this thesis is to describe their effect on the control of glucose homeostasis. Incretin receptors are widely distributed in multiple tissues and organs suggesting their complex effects including the regulation of glucose homeostasis and food intake by various mechanisms including both peripheral and central effects. Owing to their ability to regulate insulin secretion and glucose metabolism incretins have a potential in the treatment of type 2 diabetes mellitus. Incretin-based therapy strategies are discussed and compared in this thesis. Analogues of glucagon-like peptide-1, one of the incretin hormones, have the ability to lower body weight and therefore are considered as a possible obesity treatment both in patients with and without diabetes. The role of incretins in pathophysiology of obesity and studies carried out in order to evaluate its potential in the treatment of this disease are depicted. Analysis of internal communication in a specific organization

The theme of the Bachelor's thesis is the analysis of the system of internal communication in a specific organization. The first part, I define the concept of information, every detail of the theoretical concepts related to the topic of communication, the system of internal communication channels of communication. In the second part I will prepare an analysis of the internal communication in a transnational organization Takko Fashion clothing, s. r. o., I will survey its information flows and resources and I will create the schematic diagram of the Organization internal communications network. In the form of questionnaires, find out what effect they have on staff. In conclusion, summarise the results and propose a solution to eliminate the barriers to corporate communications. Role of mitochonodrial genome in myocardial ischemia-reperfusion injury of spontaneously hypertensive rats (SHR) adapted to hypoxia.

Diplomová práce Abstract - Iveta Brabcová Abstract Ischemia-reperfusion heart injury is one of the most significant diseases affecting mankind and therefore current research pays more attention to its prevention and knowledge of the possible mechanisms which protect the heart. Adaptation to hypoxia has been known for several decades as a cardioprotective intervention but the main issues of protective mechanisms which are induced by the adaptation are still not completely understood. An important role of mitochondria as the main producers of energy and reactive oxygen species which can play a signalizing role in these mechanisms is confirmed in many studies. For this reason a special conplastic strain SHR/OlaIpcv-mtBN/Crl was created. This strain carries the nuclear genome of spontaneously hypertensive rat (SHR) and the mitochondrial genome of normotensive, highly resistant strain Brown Norway (BN). The aim of this study was to compare the expression of selected gene transcripts in the area of energy metabolism, of genes which are related to mitochondrial biogenesis and signaling and antioxidant systems. Comparing the expression was analyzed between strains and after chronic hypoxia adaptation, which cause cardioprotective phenotype in both of these strains.
