Utilization of stem cells

The aim of the thesis is to summarize the basic information about the stem cells and outline their possible use in medicine. The beginning of this thesis is focused on basic description of the stem cells, their properties, classification and method of acquisition. In next part of the thesis we are focused on typology of each of these cells and cell dividing. Our focus is at utilization of stem cells in current medicine that is pivotal theme in mine thesis, use of mesenchymal cells, their transplantation and use in the treatment of arthrosis and in plastic surgery. The thesis is further focused on the use of hematopoietic, embryonic and stem cells from the umbilical cord blood - here are described diseases which can be treated with allogeneic umbilical cord blood, and also for their use in the treatment of diseases that can be treatable by cell therapy but which are still being researched such as diabetes mellitus and Parkinson's disease or the use of stem cells in preterm infants.

Bachelor thesis Morphological characteristics of alterations in lungs in a mouse model of atherosclerosis Author: Martina Vyskočilová Study course: Medical laboratory technician Charles University Faculty of Pharmacy in Hradec Králové Background: Atherosclerosis is a multifactorial disease caused by deposition of blood lipids into the intima layer of blood vessels. Due to the fact that process of atherogenesis is associated with immune reactions, disease is considered as an inflammatory process. Atherosclerotic plaques rarely affect pulmonary arteries, where this condition is accompanied by an increase of pulmonary pressure, which is very likely the trigger of disease. Aim of this study is to find out whether there are morphological changes in the lungs of ApoE/LDLr-/- mouse model of atherosclerosis, characteristic for pulmonary atherosclerosis, compared to lungs of healthy mice. Methods: Lungs of ApoE/LDLr-/- and healthy C57BL/6J control mice have been fixed in 4% neutral formaldehyde, embedded in paraffin and sliced into 4 μm thin sections. One of the sections has been stained with hematoxylin-eosin, the other one with green trichrome. Method: C57BL/6J female mice were used for the experiment. 6 mice were fed with the high fat diet (experimental group), which contains 1,25 % of cholesterol and 40 % of fat, for the period of 3 months. Six mice (representing control group) were fed with the chow diet the whole time. Immunohistochemical analysis was performed by means of avidin-biotin complex method. Results: The expression of eNOS was visible on the vascular endothelium but there were no differences between the control group and the experimental group. Similarly, ICAM-1 expression was detected only in endothelium in aorta but no differences in the expression between control and experimental group were visible. Endothelial dysfunction represents the first step in the development of atherosclerosis and it is characterized by disruption of endothelial homeostasis. Statins are drugs of choice in the treatment of atherosclerosis and they can decrease LDL cholesterol levels and positively affect levels of HDL cholesterol. Statins, however have a wide range of other effects, which are referred to non-lipid or pleotropic effects. The most important non-lipid effects of statins are lipid- independent modulation of endothelial function, antioxidant, anti-inflammatory, antithrombogenic and antiproliferative effects. This study aimed to evaluate the expression of specific markers of endothelium protection, inflammation and oxidative stress in cardiac wall of transgenic model of mice with high plasma levels of human soluble endoglin, which was fed a high fat diet. Methods: For this work, we used a female transgenic mouse model on a CBAxC57BL/6J background with high plasma levels of human soluble endoglin, fed a high fat (40%) and high cholesterol (1.25%) diet. As a control group, we used their female littermates showing undetectable levels of human soluble endoglin. Tyrosol has been formerly confirmed as a substance which shortens the lag phase of Candida albicans cell cycle and stimulates its conversion to the hyphal form, farnesol stimulates blastospores. This work covers the effect of tyrosol combined with two antimycotics, fungistatic fluconazole and fungicidal amphotericine B. Candida albicans strains used in the experiment were mostly isolated from samples taken from patients suffering from variously located Candida albicans infections and four laboratory standard strains were also used. The method used for the experiment was a modified broth dilution method using combinations of both tyrosol and antimycotics various concentrations, which were prepared by two-fold dilution. Using amphotericine B, 28 strains did not show any difference in MIC compared with the control sample, 6 strains showed mildly higher MIC and 3 strains shower mildly lower MIC. Diploma thesis Charles University in Prague, Faculty of Pharmacy in Hradec Králové Medical laboratory technician Background: The aim of this diploma thesis was to describe changes of expression eNOS and ICAM-1 in the wall of aorta of mice with low and high levels of soluble form of endoglin (Sol-Eng+). The attempt was to compare expression of both markers from the position of intensity and also from the position of area of staining. For this purpose we used immunohistochemical methods with detection in fluorescent microscope. Methods: For this study were used transgenic mice with high levels of human soluble endoglin (Sol-Eng+). Female mice were fed from their six months of age with high fat diet, which contained 1.25% of cholesterol and 40% of fat. The mice were fed with this diet for another three months. The same old female mice from the same brood with low level of soluble endoglin in plasma were used as a control group. The expression of eNOS and ICAM- 1 was defined on sections of aorta of the mice with fluorescent immunohistochemistry. PharmDr. Petr Nachtigal, Ph.D. Background: We studied the expression of P-selectin in brachiocephalic artery in the apoE−/− /LDLr−/− deficient mice, which were fed with the low carbohydrate, high protein diet. Methods: For this study we used the female apoE−/− /LDLr−/− deficient mice, which were divided into two groups, 6 mice in each group. Control mice were fed with AIN-93G diet (control group) and the second group was fed by low carbohydrate, high protein diet for 8 weeks. Immunohistochemical analysis was performed in tissue sections of brachiocephalic artery by avidin-biotin complex method with DAB detection. Results: Biochemical analysis revealed significantly increased levels of total cholesterol after feeding with low carbohydrate, high protein diet. Immunohistochemical analysis detected P- selectin expression only in endothelial cells, with more intensity in endothelial cells covering atherosclerotic plaques in both groups apoE−/− /LDLr−/− deficient mice. Author: Markéta Marešová Title of bachelor thesis: Coeliac dinase Bachelor thesis Charles Univerzity in Prague, Faculty of Pharmacy in Hradec Králové ABSTRACT: Background: The thesis deals with the issue of the celiac disease. In the theoretical part I dealt with the definition, pathogenesis and pathology of this disease. I also looked at different forms and possible complications of the celiac disease, epidemiology, diagnosis and therapy. In the practical part my goal was to evaluate the overall awareness of patients on the basis of questionnaire studies. Main findings: I also focused on the quality of their lives and the non-health associated complications which the celiac disease entails. Conclusions: The majority of patients are well-informed about their disease according to results of questionnaire studies.
