Determination of residues of the selected pesticide effective substances in wine using the GC/MS method

The grape is one of the oldest cultivated plants. It can be consumed as a fruit or it can be used as a raw material for making stews, juices or the cultivated drink – wine. Generations of wine-makers have been striving to improve both the cultivating conditions of vineyards and the quality of wine. Many harmful elements, such as herbivorous pests, diseases and storms are important factors that can have a negative impact on crop quality. To regulate the effects of these factors, an integrated protection system is used, incorporating agronomical measures, resistant varieties and a wide spectrum of pesticides. However, increasing the usage of pesticides brings certain dangers to both the production quality and the environment. This necessitates the monitoring of the negative effects, including the content of pesticide residues in the grapes. My thesis studies and experimentally assesses selected pesticide residues in certain wine samples.

The aim of the bachelor thesis is description of renting apartment, demarcation of basic concepts regarding this issue. Work has mainly specialized in the analysis and description of the rental housing and its accessibility for the population and the concept of social housing , applied in the Czech Republic. The aim of the work is an analysis of rental housing in the city Czech Cieszyn. In my work I have tried to summarize how you can increase the availability of housing and amenities. I tried to assess the situation on the housing market , especially rental housing and the concept of social housing in the Czech Republic. To analyze the state of housing fund in the Czech Cieszyn , I used the materials published by the municipality , housing cooperatives , churches and real estate companies . For an analysis of pricing conditions of the lease , I used data provided real estate brokers and data obtained from the survey.
Monitoring of vitamin E content in the samples of cereals

Thesis deals with the analysis of vitamin E in cereal samples, namely barley and wheat and malt produced from them. Selected samples were treated with low-temperature plasma, fungicide and biological treatment to elimination fungus and mycotoxins. The aim of the thesis was to find out the influence of the treatment, the combination of several types of treatment on vitamin E content in grain and malt. If the content of health-positive substances is not reduced and grain can be used for food, malt, or for feed purposes. The theoretical part focuses on the properties, occurrence, biochemistry and importance of vitamin E for living organisms. It also focuses on the properties of cereals that are a significant source of vitamin E. Malt production and sample treatment are also described. The experimental part describes the field experiment, individual types of treatment, extraction of vitamin E from the samples and determination of vitamin E content by high performance liquid chromatography with fluorescence detection. The results are compared with the current literature. It was found that the treatment did not have a significant negative effect on vitamin E content.

Enzyme content monitoring in wheat malt

The aim Master thesis was to evaluate the influence of malting technology on the content of hydrolytic enzymes -amylase and -amylase. Another aspect of this thesis was to select a wheat variety with the highest activity of enzymes -amylase and -amylase. The first part of the practical part of my work is shown measuring enzyme malt model varieties of wheat set (Triticum aestivum), where individual samples differ technological processing of malting and the second part is focused on the application of efficient malting conditions on other varieties from different locations in the Czech Republic in 2016 . All malt samples were analysed to -amylase and -amylase activity by assay Ceralpha method and Betamyl-3 method made by Megazyme company. After measuring the first part, the most effective malting condition was evaluated, such as a 7 day malting time, a 45% water content, a germination temperature of 18 ° C, and a drying temperature of 50 ° C. In the second part, the varieties are so malted and the highest content of enzyme -amylase was Rumor, it had averafe contanin -amylase 277 U/g, Bonanza – 255 U/g and Izzy –254 U/g. The biggest contain of enzymes -amylasehad varieties Matchball – 37,9 U/g , Fakir – 35,6 U/g, and Bernstein – 35,3 U/g.
Introducing of method of pyridoxine determination by liquid chromatography in food products and resources

This master‘s thesis deals with quantification of vitamin B6 in beverages, food supplements and raw materials in food industry. The literature retrieval part summarizes general information about vitamines, vitamine B6, nicotine acid and vitamine B1. Further on in this part there is described principle of high pressure liquid chromatography and quantification of individual vitamines. In the experimental there are listed used tools, apparatus and chemicals. There is described preparation of idividual samples of food supplements, energy drinks, multivitamine drinks, beers and brewer’s malts. This chapter also contains information about chromatographic separatory systems Shimadzu and SpectraSystem. The quantification was performed by high pressure liquid chromatography on a reverse phase with gradient elution and two ways of detection, fluorescent and spectrofotmetric. The result of experimental activities and vitamine content in analysed samples are presented in the next chapter. In the final contains summarization of results obtained in experimental part. The maser’s thesis was measured in the laboratory of Institute of Food Science and Biotechnology, Faculty of Chemistry, Brno University of Technology.
Determination of ergosterol content in brewing materials

Presented bachelor’s thesis is focused on determination of ergosterol content in barley and malt. In theoretical part the presence of ergosterol in barley and malt has been elaborated, the purposes of its determination, detailed overview of previously applied methods of extraction from samples of barley and malt as well as other biological materials and methods of quantification. Practical part is focused on optimizing of extraction and determination of ergosterol by high performance liquid chromatography with diode array detection (HPLC – DAD).
Monitoring of organic acid content in alcoholic drinks

The aim of the thesis is determination and optimization of organic acids in beers using the methods of capillary isotachophoresis (CITP) and high performance liquid chromatography with diode array detection (HPLC/UV). Beer is a complicated matrix therefore the samples need pretreatment using solid phase extraction (SPE). The diploma thesis discusses the optimization of the analytical methods (HPLC/UV and CITP) and optimization of SPE. In the theoretical part, history, characterization and technology of beer production are presented. Further, characteristics of organic acids and methods for organic acid determination are also discussed. The experimental part deals with the preparation of solutions, tested samples, calibration samples. Parameters and procedures of analytical methods (HPLC/UV and CITP) and pretreatment using the solid phase extraction are also described in the experimental part. All results are summarized and compared with the current literature in discussion and conclusion. Eleven beer samples from the retail stores were analyzed. In all samples, six organic acids (lactic, oxalic, succinic, acetic, citric, benzoic) were determined by CITP and five organic acids (lactic, oxalic, succinic, acetic, citric) by HPLC/UV were determined. Results of this study give an overview of the organic acid contents in beer samples.
Cave maps and their symbols in publications Speleo and Speleofórum

The aim of this bachelor's thesis was to tackle the problem of map symbols used in maps of caves, cave systems and similar naturally created landforms. Two publications dealing with this issue were analyzed, namely the anthology Speleo and Speleofórum. For the Speleo magazine, it was 74 issues published between 1990–2019 and for the Speleofórum magazine 23 issues published in the years 1989–2019. In total, more than 1,000 maps and their map symbols were recorded. I total it was 264 and 340 symbols for the Speleo anthology and the Speleoforum respectively. Subsequently, the symbols were sorted into categories and digitalized. Only a fraction of the maps included a legend, and therefore the meaning of used symbols had to be identified in the text the respective article. Maps with key have become more common in the last five years, thanks to more affordable digital technologies such as Therion, which automatically provides a unified legend.
Stability of selected mycotoxins in beer

Mycotoxins are secondary metabolites of moulds, which attack cereals, for example barley, from which mycotoxins then get to beer. This submitted work is focused on ochratoxin A, deoxynivalenol and zearalenone, which can occur in beer. The first part of this master’s thesis consists of literary research, which describes mycotoxins in general, points out their occurrence, prevention of their formation and delivers information about their physical and chemical properties and toxicity. Furthermore, the research contains basis of malt and beer technology, the occurrence of mycotoxins in beer and raw materials for its production. The research describes changes in concentration of mycotoxins across malt and beer production. The next part deals with possibilities of determination of mycotoxins in barley, malt and beer, compares individual methods of their determination and points out many difficulties of some analyses. The experimental part of this work pursues determination of ochratoxin A, deoxynivalenol and zearalenone in different types of beer with the help of UPLC-FLR, HPLC-MS and ELISA. Instrumental techniques are validated and gathered results are compared with the results in literature. The goal of this master’s thesis is to assess the stability of ochratoxin A and deoxynivalenol in beer over time. The gained results show that there are changes in the concentration of ochratoxin A over time, nevertheless those changes show no pattern. Overall, there was a decrease in concentration in 47 % of the samples and an increase in 28 % of them. In the rest of the samples the concentration did not change. The concentration of deoxynivalenol does not change over time. One of the other goals of this thesis is monitoring of selected mycotoxins in beer. The average concentration of ochratoxin A in the samples was 39 ng/l and deoxynivalenol 9,9 g/l. Zearalenone did not occur in any of the samples when determined by liquid chromatography. All results agree with literature. Next, the thesis compares different analytical methods for determination of ochratoxin A, deoxynivalenol and zearalenone. The screening method ELISA is compared to UPLC-FLR and HPLC-MS. The determination of ochratoxin A by ELISA has shown to be time consuming, nevertheless the results responded to instrumental technique. ELISA overestimated the results of determination of deoxynivalenol in beer by 363–697 % and with zearalenone there were found false positive results.
