Identification of the binding sites on transient receptor potential cation channel TRPC6 for Calmodulin and S100A1
Bílý, Jan ; Teisinger, Jan (advisor) ; Krůšek, Jan (referee) ; Pavlíček, Jiří (referee)
Identification of the binding sites on transient receptor potential cation channel TRPC6 for Calmodulin and S100A1 The TRP (transient receptor potential) group of ion channels represents a large subset of membrane receptors. A part of this supergroup are canonical TRPC channels with a sequence homology analogical to TRP receptor first discovered at fruit fly (Drosophila melanogaster). These membrane channels are involved in a variety of physiological functions in different cell types and tissues. TRPC6 is a non-selective cation channel that modulates the calcium level in eukaryotic cells (including sensory receptor cells) in response to external signals. TRPC6 channel contains binding domain CIBR (Calmodulin inositol binding region), which is also able to adapt to calcium binding protein S100A1. Characterisation of the integrative binding site for calmodulin (CaM) and S100A1 at the C-tail of TRPC6 is presented in this work. Using site-directed mutagenesis, soluble protein fragments TRPC6 CT (801-787) were prepared with intentional changes in amino acid sequence. Several positively charged amino acid residues (Arg852, Lys856, Lys859, Arg860 and Arg864) were determined by measurement of fluorescence anisotropy influence and their participation in the calcium-dependent binding of CaM and/or S100A1 to...

The mirror comparison of Czech and German statistics of mutual trade
Heczková, Markéta ; Taušer, Josef (advisor) ; Bílý, Jan (referee)
The final thesis is aimed at mirror comparison of mutual trade between the Czech Republic and Germany. On the basis of the comparison of Czech and German methodologies of foreign trade and consequential analysis of their mirror comparison focused especially on effect of the Czech Republic's accession to the EU (access effect) were individual abnormalities affecting mirror comparison identified and described. There were several solutions put forward so that already detected abnormalities could be effectively dissolved, moreover their figure in mirror comparison could be eliminated and data acquired from the comparison would reach as high level of credibility as possible. The Single flow and Qualified single flow systems are described and discussed in relation to mirror comparison because they are possible and already existing plans for simplification of Intrastat.
Coaching and its contribution to corporate management in the Czech Republic
Bílý, Jan ; Campbell, Jan (advisor) ; Valášek, Petr (referee)
This bachelor thesis focuses on approach of Czech corporate management to coaching. The coaching and mentoring of people has become a trending topic lately. Coaching allows us not only to discover a potential within ourselves but also to release it fully. On the basis of qualitative research, the thesis analyses the benefits of coaching for the coached person, from the point of view of the coach, the ordering party and of the coached person him/herself using the self-assessing scales. According to the results, the most important asset for the coached person is higher autonomy and responsibility for one's work. From the manager's point of view, coaching helps promoting better communication, openness and also a possibility to praise and acknowledge employees and subordinates in a better way. The final part of the thesis evaluates the fulfillment of the laid down objectives.
The development of foreign trade of the Czech Republic after World War 2
Heczková, Markéta ; Štěrbová, Ludmila (advisor) ; Bílý, Jan (referee)
This work analyses foreign trade of Czechoslovakia between years 1945 and 1992, respectively 1. 1. 1993. This period is terminated by the end of World War 2 and the establishment of independent state -- the Czech Republic. The work is concerned with foreign trade commodity, as well as territorial structure, also with the terms of trade and their action at state economy. Last but not least are studied changes of direction in foreign trade of Czechoslovakia after the breakdown of socialism. The aim of the dissertation is then evaluate trade with Germany during this period.
