Detection of epithelial-mesenchymal transition markers (EMT) in cells in vitro
Špaček, Petr ; Szotáková, Barbora (advisor) ; Trejtnar, František (referee)
Charles University Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Petr Špaček Supervisor: Doc. Ing. Barbora Szotáková, Ph.D. Consultant: RNDr. Věra Králová, Ph.D. Title of diploma thesis: Detection of epithelial-mesenchymal transition (EMT) markers in cells in vitro Epithelial-mesenchymal transition (EMT) is a process during which motile mesenchymal-like cells develop from non-motile parent epithelial cells. Physiologically, EMT plays important roles during embryonic development and wound healing. Loss of control over this mechanism can lead to fibrosis and cancer progression. Motile mesenchymal-like cells can pass through the basal lamina, get into the blood vessels and spread to distant tissues. Transition is regulated by EMT biomarkers. The biomarkers comprise wide spectrum of proteins, including cell surface proteins (E-cadherin, N-cadherin), cytoskeletal proteins (vimentin), microRNA (miR 200) and transcription factors (Snail, Twist). In this study, expression of EMT biomarkers was evaluated using RT-PCR and Western blotting. The ability to migrate was assessed using real-time analysis with the x-CELLigence system. Two known triggers of EMT, the StemXVivo™ EMT Inducing Media Supplement (IS) and TGF-β, were compared in human oral cancer cell lines DOK and...

Frost susceptibility of improved soils to pavement subgrade
Lorenc, Daniel ; Špaček, Petr (referee) ; Stehlík, Dušan (advisor)
This diploma thesis deals with the issue of the effect of freezing on soils in subgrades of road structures. The theoretical part compares methods of determining the frost susceptibility of soils used in selected countries of the European Union and the Czech Republic. It also explains the principle of frost heave in subgrade layers. The aim of the practical part is to compare the direct and indirect method of determining frost susceptibility in the Czech Republic and the California bearing ration CBR or the immediate bearing ratio IBI.

The amount of damage determination in cases of defects and failures of flexible pavements road structures
Špaček, Petr ; Jůza, Petr (referee) ; Kledus, Robert (referee) ; Stehlík, Dušan (advisor)
This thesis is focused on possibilities of financial valuation of flexible pavements surface layers conditions, in connection with whole life cycle of the pavement and certain possibilities of their maintenance and repairs, in cases of defects and failures from forensic engineering point of view. There were carried out analysis of the topics connected with judging of flexible pavements surface layers defects and failures related to forensic investigation in the Czech Republic, as well as abroad. In this thesis there were created pricing models of the certain construction works which are related to repairs and maintenance of flexible pavements. Within the possibilities of the author, there were compared market prices of the particular construction works in selected regions with the prices according to generally used pricing systems in the Czech Republic. The goal of this thesis is to create unified methodology for amount of damage financial valuation in cases of defects and failures of flexible road pavements surface layers for forensic engineering practice in the Czech Republic.
Profiling of Non-Core Part of Security System
Špaček, Petr ; Michal, Bohumil (referee) ; Kašpárek, Tomáš (advisor)
The thesis is focused on identifying and eliminating performance problems in the security system "AVG Server Edition 8.5 for Linux/FreeBSD", which is used for filtering SMTP traffic. It includes explanation of the principle of SMTP proxy filter and of the interconnection between AVG and the Postfix mail system. It describes methodology of testing, sample designing, measurement and data evaluation. Several performance problems were found and steps to eliminate them were carried out. Suggested optimization was tested and found effective.
Project Management System
Špaček, Petr ; Jirák, Ota (referee) ; Ruttkay, Ladislav (advisor)
The work deals with project management.  The design software helps to manage and plan projects by using a simple graphic user interface. For implementation there are used technologies like Microsoft .NET, Microsoft SQL Server 2008.
SEO Analysis Tool
Špaček, Petr ; Očenášek, Pavel (referee) ; Ruttkay, Ladislav (advisor)
The thesis deals with SEO problems (Search Engine Optimization). The part of the thesis is creation of tool which analyzes web pages in multilanguage versions and results provides with appropriate educational effect for user. According the analyzing that can be also used in the regular interval it's possible to show optimization history.
