Possibilities of removing odors on the sewer network
Šlechtová, Tereza ; Malaník,, Stanislav (referee) ; Hluštík, Petr (advisor)
The aim of this diploma thesis is to research the possibilities of an odor regulation in a sewerage system. The first part of the thesis is devoted to legislation in sewege odor problematics. The second part includes description of the cause of the odor and methodology related to a hydrogen sulfide elimination either from sewerage atmosphere or straight from waste water. The next part examines the results of a survey, which was given to operators of the sewerage system. The survey examined the cause of an odor, its control and possible solutions of an odor in a sewerage system. Practical part is devoted to a given geographical area, where these problems have occurred the past. In this area the measuring of hydrogen sulfide presence took place by adding chemicals into waste water. From the collected data an evaluation was made. This can serve as a possible recommendation to sewege operators.
Efficacy overflows
Šlechtová, Tereza ; Úterský, Michal (referee) ; Hluštík, Petr (advisor)
The aim of this bachelor´s thesis is to process the recherche of the overflow chambers The first part of this thesis includes the legislation of the overflow chambers. The following list provides a short description of the individual types of the overflow chambers and building materials which we use the most often. The thesis deals with technical proposal and hydraulic computation. The allovable pollution of diluted water is presented by dint of emission limits and pollution limits which are caused by the overflow chambers. In the practical part of this thesis are used valuable lessons learned from the theoretical parts. The aim of this part is recognition of the selected overflow chambers in the combined sewerage system in the city of Brno. The chambers were monitoring during measuring campaign in 2017. The overflow chambers were evaluated to their efficiency and construction conditions. In the latest bulit and current moved overflow chambers was calculated the ratio of dilution.
Study of peptide digestion kinetics by trypsin and chiral separations of biologically active compounds by HPLC
Šlechtová, Tereza ; Tesařová, Eva (advisor) ; Čabala, Radomír (referee) ; Mikšík, Ivan (referee)
This dissertation thesis composes of two parts; the first part focus on the characterization of trypsin, enzyme frequently used in proteomic research for the investigation and identification of protein sequences, and its peptide digestion kinetics. The second part is aimed to the enantioseparations of biologically active compounds. First part of this project focus on tryptic digestion of synthetic peptides and the development of HPLC method for the identification of synthetic peptides and their fragments. Using the in-solution digestion and HPLC method, relative kinetic constants were determined for problematic sequences. Amino acids responsible for the decrease in trypsin catalytic activity and their location towards the cleavage site were studied. Certain slight exopeptidase activity of trypsin was noted, especially at the end of peptide chain. Furthermore, three columns with immobilized trypsin used in HPLC were compared concerning their catalytic activity. The immobilization of enzymes on solid support is used to elevate the amount of enzyme present during digestion and to assure better repeatability and reproducibility of obtained results. Activity of a new trypsin column synthesized at the University of North Carolina at Chapel Hill was compared to two commercially available trypsin columns....
Determination of pesticides enantiomers in water
Šlechtová, Tereza ; Tesařová, Eva (advisor) ; Lokajová, Jana (referee)
This Barchelor's project deals with chiral herbicide mecoprop and its determination by high-performance liquid chromatography method with teicoplanin based chiral stationary phase. The teoretical part focus on basic facts conserning chirality, chiral pesticides and problems related to their usage and effects to the enviroment. It also involves the ways of their determination by the above mentioned method and gives several examples of previous separations. The experimental part is based on method optimalization and determination of changes in mecoprop enantiomeric ratio in various water samples during different storage conditions.
Comparison of methods for detection and determination of enantiomers of theanine in HPLC
Šlechtová, Tereza ; Tesařová, Eva (advisor) ; Bosáková, Zuzana (referee)
This thesis focused on optimization of separation methods for underivatized theanine enantiomers on two teicoplanin-based chiral stationary phases and separation methods for enantiomers derivatized by 9-fluorenylmethyloxycarbonyl chloride and dansylchloride. Using these optimized conditions the limits of detection and quantification were calculated. Under optimized conditions were also investigated the contents of theanine enantiomers in green tea and in selected dietary supplement containing L-theanine.
Demografické aspekty nezaměstnanosti v Pardubickém kraji v letech 2005-2014
Šlechtová, Tereza
This bachelor thesis is dedicated to unemployment of the Pardubice Region from the demographic factors point of view in period 2005-2014. The partial part of thesis is searching for causation of unemployment at the level of its districts. By using method of demographic analysis I solved mutual links and finding in the context of demographic relation of unemployment. On the basis of research problem I reached to a conclusion that Pardubice region is considerably different from the point of the unemployment, supply and structure of job vacancy and achieved degree of education. The biggest difference of unemployment we can find between districts Pardubice and Svitavy.
Vývoj demografických a socioekonomických ukazatelů nemocnosti obyvatelstva České republiky v letech 2007–2017
Šlechtová, Tereza
The thesis deals with morbidity in terms of socio-demographic and economic factors and mortality structure at the district level in the Czech Republic in 2007–2017. Using the composite indicator, cluster analysis and factor analysis, we investigated regional disparities of selected indicators. The results show that the area of morbidity and mortality of the population in the districts of the Czech Republic is regionally differentiated. Big impact plays the socio-economic situation (unemployment, the share of university graduates, housing payments), infant mortality or the old age dependency ratio. In general, the least favorable situation in the area of morbidity was in districts with negative socio-economic development.
